Developmental expression of perlecan during murine embryogenesis.

Perlecan is a modular heparan sulfate proteoglycan that is an intrinsic constituent of all basement membranes and extracellular matrices. Because of its strategic position and unique structure, perlecan has been implicated in modulating the activity of various growth factors required for normal development and tissue homeostasis. To gain insights into the potential function of perlecan in vivo, we examined the spatiotemporal distribution of its mRNA and protein core during murine embryogenesis. We utilized a new affinity-purified antibody that recognizes specifically the protein core of perlec

Rescue of the limb deformity in hammertoe mutant mice by retinoic acid-induced cell death.

Retinoids, used therapeutically primarily in the treatment of skin disorders, are potent teratogens. Several craniofacial, neural tube, and limb defects derive from a selective increase in cell death by retinoic acid in sites of spontaneous programmed cell death. Previously we showed that programmed cell death in the limb was apoptotic, and that the webbing of the foot of the Hammertoe mutant mouse correlates with diminished cell death in these regions of webbing. We therefore examined the effect of the induction of cell death by retinoic acid in normal and mutant limbs. Here we report that ex

Inactivation of two mouse alkaline phosphatase genes and establishment of a model of infantile hypophosphatasia.

We report the inactivation, via homologous recombination, of two of the three active mouse alkaline phosphatase genes, i.e., embryonic (EAP) and tissue nonspecific (TNAP). Whereas expression of the EAP isozyme was abolished in all tissues that express EAP developmentally (such as the preimplantation embryo, thymus, and testis), the EAP knock-out mice show no obvious phenotypic abnormalities. They reproduce normally and give birth to live offspring, indicating the nonessential role of EAP during embryonic development. Mice deficient in the TNAP gene mimic a severe form of hypophosphatasia. Thes

Collagenase-3 (MMP-13) is expressed by hypertrophic chondrocytes, periosteal cells, and osteoblasts during human fetal bone development.

Collagenase-3 (MMP-13) is a novel matrix metalloproteinase, the expression of which has so far only been documented in human breast carcinomas and osteoarthritic cartilage. In this study we have examined the expression of MMP-13 during human fetal development. Northern blot hybridizations revealed abundant expression of MMP-13 mRNAs in total RNA from fetal cartilage and calvaria at gestational age of 15 weeks. By in situ hybridization MMP-13 transcripts were detected in chondrocytes of hypertrophic cartilage in vertebrae of the spinal column and in the dorsal end of ribs undergoing ossificatio

Urokinase-type plasminogen activator regulates cranial neural crest cell migration in vitro.

Proper migration and differentiation of neural crest (NC) cells are required for normal development of craniofacial structures, heart and great vessels, sensory and autonomic nervous systems, and other organs with vertebrate embryos. Serine-protease inhibitors reduce NC cell migration in vitro, suggesting the extracellular proteases are important mediators of NC cell migration. While plasminogen activator activity levels are high in NC cells relative to other embryonic tissue, its ability to regulate NC cell migration has not been specifically tested in vivo or in vitro through its ability to

Mouse embryonic submandibular gland epithelium loses its tissue integrity during early branching morphogenesis.

During the development of the mouse submandibular gland, the epithelium undergoes not only shape changes to produce extensively branched lobules and stalk, but also changes in cell arrangement from a cell mass to a cavitated cell sheet. The present study examined the organization in the developing epithelium of intercellular adhesion systems and of actin-containing microfilaments. E-cadherin and beta-catenin, which are components of cell-to-cell adherens junctions in epithelial cells, were distributed along the cell periphery of almost the entire epithelium of the submandibular gland at all st

Tyrosinase is a new marker for cell populations in the mouse neural tube.

Tyrosinase, the key enzyme in melanin synthesis, is expressed in pigment cells derived from both neural crest and neuroectoderm. The present study was performed to detect tyrosinase promoter activity and tyrosinase gene expression during murine brain development. Mouse tyrosinase 5' region (6.1 Kb) was used to direct lacZ expression in transgenic mice. During embryogenesis, the transgene reproduced tyrosinase expression in pigment cells but was also observed in embryonic neuroectoderm and migrating neural crest cells. Both tyrosinase and lacZ were detected in cell populations often organized i

Fibulin-1 and fibulin-2 expression during organogenesis in the developing mouse embryo.

Fibulin-1 and fibulin-2 are extracellular matrix proteins with unique structural features. We used in situ hybridization and immunofluorescence staining to examine the expression of fibulin-1 and fibulin-2 during mouse embryogenesis. Both fibulins have previously been shown to be deposited at sites where polarized cells convert into mesenchyme, during early stages of development of endocardial cushion tissue and in neural crest cells. By Northern blots we confirm that expression of fibulin-2 is particularly high in the developing and newborn heart. We also show that fibulin-2 mRNA and protein

Chondrogenic potential of chick embryonic calvaria: II. Matrix calcium may repress cartilage differentiation.

Chick embryos cultured in the absence of their eggshell are rendered severely calcium-deficient, and develop a cartilage-like phenotype in the calvarium, a normally osteogenic tissue. In the preceding paper (Jacenko and Tuan [1995] Dev. Dyn. 202:13-26), experiments using organ cultured calvaria from day-12 normal and shell-less embryos showed that depletion of calcium alone may be responsible in promoting chondrogenic differentiation in calvaria. Here these findings were extended using an in vivo calvarial grafting technique, such that the extent of calvarial matrix calcification was a functio

Gene encoding a novel murine tissue inhibitor of metalloproteinases (TIMP), TIMP-3, is expressed in developing mouse epithelia, cartilage, and muscle, and is located on mouse chromosome 10.

Remodeling of the extracellular matrix (ECM) is an essential component of normal development and is also involved in the pathogenesis of arthritis and the spread of cancer. The matrix metalloproteinases and their natural inhibitors, the tissue inhibitors of metalloproteinases (TIMPs), play an important role in this context. We have isolated mouse cDNA clones encoding a novel member of the TIMP family, designated TIMP-3. We have assigned the Timp-3 locus to the [C1-D1] region of mouse chromosome 10 using both genetic and cytogenetic methods. The conceptual translation product of the Timp-3 cDNA

Growth dynamics of pancreatic islet cell populations during fetal and neonatal development of the rat.

Rats from 18 days fetal to 28 days neonatal ages were studied for the total population sizes and cell proliferation activities of insulin secreting B cells, glucagon secreting A cells, somatostatin secreting D cells, and pancreatic polypeptide secreting PP cells. Cell population sizes were assessed by morphometric quantitation of immunohistochemically stained cells by a linear scanning method and cell proliferation activities were estimated by [3H]-thymidine labeling indices of these cell populations. There was a continuous increase in population sizes for all 4 islet cell types, with the fast

Prevention of spinal neural tube defects in the curly tail mouse mutant by a specific effect of retinoic acid.

Curly tail mouse mutant embryos (ct/ct) develop spinal neural tube defects (NTD) in 54% of cases, comprising isolated tail flexion defects and spinal bifida with tail flexion defects. Both types of spinal NTD result from delayed closure of the posterior neuropore (PNP). Previous studies (Seller et al. [1979] Proc. R. Soc. Lond. Biol. 206:95-107; Seller and Perkins [1982] Prenat. Diagn. 2:297-300) described a paradoxical effect of retinoic acid (RA) on the phenotypic expression of the ct mutation: Treatment with low doses of RA on day 8 of gestation increased the incidence of total NTD, whereas

Fibrillin delineates the primary axis of the early avian embryo.

Chicken embryos were immunolabeled with antibodies to fibrillin, an extracellular matrix molecule implicated in the Marfan syndrome. The pattern of labeling, established using both whole-mounted and sectioned embryos, showed a striking localization at the primary axis of the early embryo, including Hensen's node. After stage 8, the labeling of Hensen's node diminished while other structures in the embryo exhibited increased immunolabeling. These structures include the region of the presumptive dorsal aorta, the ventral surface of the notochord, and the mesocardium. Injections of the antibody b

Changes in cellular dynamics during the development of the ciliary epithelium.

Eye growth and the formation of the ciliary folds during development depend on the maintenance of normal intraocular pressure. In this study we measured labeling index, packing density, height, and volume of the prospective ciliary epithelial cells during the early development of the eye in chicken embryos. Changes in these values were related to the formation of the ciliary folds and to experimental alterations of intraocular pressure. These variables changed independently in the outer, pigmented layer and the inner, non-pigmented epithelial layer of the ciliary epithelium. The labeling index

PRDM16 is necessary for sensory neuronal development and differentiation in the trigeminal ganglion.

BACKGROUND: Cranial neural crest cells (cNCC) generate craniofacial cartilage, bone, and peripheral neurons and glia, and birth defects arise when the cartilage/neuronal/glial progenitors fail to differentiate. PRDM16 is a transcriptional regulator containing both zinc-finger and SET domains, implicated in craniofacial development and orofacial clefting, but its role in cranial sensory ganglion formation has not been defined. RESULTS: Here, we demonstrate that prdm16 is required for trigeminal ganglion (TG) assembly and sensory neurogenesis from cranial neural crest lineages. In zebrafish, pr

Oogenesis and germinal bed morphology of the brown anole (A. sagrei).

BACKGROUND: The brown anole is a model species of the genus Anolis, a squamate (encompassing lizards and snakes) group widely studied in evolutionary, behavioral, and developmental biology. Full genome annotation, the establishment of gene editing techniques, and comprehensive description of reproductive tract morphology and embryogenesis in this species have laid the foundation for functional studies. However, analysis of brown anole oogenesis is still required and vital to optimize genome modification, mutant line establishment, and analyses of the evolution of reproductive developmental mec

The emerging role of ER-localized TMEM proteins in neurodevelopmental disorders.

Neurodevelopmental disorders (NDDs) comprise a spectrum of conditions that impair brain development and nervous system function, resulting in lifelong cognitive, behavioral, and motor disabilities. Globally, more than 3% of children are affected by NDDs. Although advances in next-generation sequencing technology have identified many disease-associated genes, the cellular and molecular mechanisms underlying NDD pathogenesis remain poorly understood. Recent genomic and clinical studies have highlighted the transmembrane (TMEM) family of proteins as emerging contributors to NDDs. TMEM proteins re

Orthodentine-osteodentine and osteocytic-anosteocytic transition inferred from Silurian stem chondrichthyan dermal jawbones with ever-white teeth overgrown by brown denticles.

BACKGROUND: The dentigerous jawbones of a 423-million-year-old stem chondrichthyan (ischnacanthid acanthodian) carry a characteristic combination of multi-row non-shedding white teeth with brownish overgrowing denticles. The color distinction makes it a good model for observing the interaction between two odontode lineages, but the nature of the opaque white material and its relationship to other tissues is unknown. RESULTS: Synchrotron microtomography demonstrates that the white tissue is a lightly mineralized orthodentine. By contrast, the orthodentine on the denticles is highly mineralized

Early ultraviolet exposure disrupts late dorsal fin skeletal development in zebrafish.

BACKGROUND: Most organisms on earth experience ultraviolet radiation. High levels of ultraviolet B radiation can cause DNA and protein damage. Zebrafish have become a popular model to investigate the impact of ultraviolet B radiation. Previous studies have identified that early ultraviolet B radiation damages the transient, larval fin fold by inducing apoptosis. However, until now it remained unknown how this early exposure affects late-forming skeletal structures. RESULTS: We determined that ultraviolet B radiation at early stages results in skeletal phenotypes appearing weeks later. Specifi

Seawater incubated embryos of a freshwater Aplocheilus lineatus (Aplocheilidae; Cyprinodontiformes) support the dispersal hypothesis of killifish intercontinental distribution.

BACKGROUND: Aplocheiloid killifishes (Cyprinodontiformes, Aplocheiloidei) inhabiting Africa, America, and South Asia are unique in that embryos of annual species are capable of entering a state of diapause and becoming extremely stress resistant. Distribution of aplocheiloids is a biogeographical enigma: how freshwater fish managed to cross long-distance oceanic barriers to become intercontinentally distributed? As a hypothesis, we proposed their embryos to be a dispersal stage. To test it, we compared embryonal development in seawater and freshwater incubated eggs of Aplocheilus lineatus from
