BACKGROUND: Xeroderma pigmentosum (XP) is an inherited photoaging syndrome caused by mutations in genes involved in the nucleotide excision repair (NER) pathway. XP patients exhibit hypersensitivity to ultraviolet (UV) radiation, leading to accelerated skin aging and requiring lifelong sun avoidance. In this study, we aimed to investigate UV-induced cellular senescence in XP melanocytes and explore potential senotherapeutic strategies.
METHODS: Melanocytes differentiated from iPSCs derived from an XP-A patient and a healthy individual, respectively, were used to induce senescence. Senescence …

T cell immunosenescence refers to the progressive functional decline of T lymphocytes with aging, characterized by the phenotypic markers, mitochondrial dysfunction, and the senescence-associated secretory phenotype (SASP), representing a pivotal aspect of overall immune aging. This review systematically elucidates the critical role of T cell immunosenescence in the pathogenesis of common inflammatory skin diseases, including psoriasis, atopic dermatitis, rosacea, and seborrheic dermatitis. Senescent T cells drive the production of a disease-specific SASP via internally dysregulated signaling

Osteoarthritis (OA) is a common and debilitating joint disorder, with its pathogenesis significantly influenced by factors such as aging and obesity. A critical aspect of OA development is the senescence of chondrocytes, which is characterized by irreversible cell cycle arrest and the secretion of pro-inflammatory molecules, collectively known as the senescence-associated secretory phenotype (SASP). Senescent chondrocytes compromise the maintenance of the extracellular matrix (ECM) and accelerate cartilage degradation, thereby exacerbating the progression of OA. Contributing factors to chondro

The skin is the largest organ in the human body and is made up of various cells and structures. Over time, the skin will age, which is not only influenced by internal factors, but also by external environmental factors, especially ultraviolet radiation. Aging causes immune system weakening in the elderly, which makes them more susceptible to dermatosis, such as type 2 inflammatory mediated pruritus. The immune response in this condition is marked by senescent cells consistently releasing low amounts of pro-inflammatory cytokines through a senescence-associated secretory phenotype (SASP). This

Bacterial genotoxins damage host cells by targeting their chromosomal DNA. In the present study, we demonstrate that a genotoxin of Salmonella Typhi, typhoid toxin, triggers the senescence-associated secretory phenotype (SASP) by damaging mitochondrial DNA. The actions of typhoid toxin disrupt mitochondrial DNA integrity, leading to mitochondrial dysfunction and disturbance of redox homeostasis. Consequently, it facilitates the release of damaged mitochondrial DNA into the cytosol, activating type I interferon via the cGAS-STING pathway. We also reveal that the GCN2-mediated integrated stress

Pancreatic cancer is a highly aggressive tumor with a poor prognosis. The ubiquitin-proteasome system is crucial in maintaining protein homeostasis and regulating cellular senescence, thereby influencing tumorigenesis and progression; however, the specific mechanisms underlying this process remain unclear. Ubiquitin protein ligase E3A (UBE3A) is associated with various tumors; however, its potential role in pancreatic cancer warrants further investigation. The effects of UBE3A on cellular senescence were evaluated through in vitro and in vivo experiments, including SA-β-gal staining, EdU assay…

Cellular senescence and immune dysfunction intersect as central biological processes shaping disease progression and therapeutic responses across ageing-related pathologies. Rather than acting independently, senescent cells (SnCs) and immune cells engage in a dynamic and bidirectional crosstalk that influences immune surveillance, inflammatory signaling, and tissue remodeling. This review focuses on the molecular and cellular mechanisms through which SnCs modulate immune composition and function, emphasizing immune checkpoint ligands, stress-associated surface molecules, and senescence-associa…

Cellular senescence is a process of physiological growth arrest that can be induced by intrinsic or extrinsic stress signals. Some cancer therapies are associated with senescence of cancer cells with a typical cell cycle arrest. Doxorubicin (Dox) induces senescence by a p53-dependent pathway and telomere dysfunction of numerous cancers. However, cellular senescence induces suppression in proliferation activity, and these cells will remain metabolically active and play an important role in tumor relapse and development of drug resistance. In the current study, we investigated the apoptotic effe…

Intervertebral disc degeneration (IVDD) is a major cause and independent risk factor for low back pain. This study aims to explore the molecular mechanisms underlying IVDD and provide new ideas for targeted treatment. Cellular senescence is a key pathological event in IVDD, but its upstream regulatory network in nucleus pulposus cells (NPCs) remains unclear. This study found that MATN3 protein expression decreased significantly during IVDD, and its level was closely associated with NPC senescence. Functionality experiment data show that overexpression of MATN3 can effectively inhibits NPC sene…

Acute kidney injury (AKI), a life-threatening disorder marked by abrupt renal dysfunction, is increasingly recognized as a global healthcare challenge. It not only triggers immediate organ dysfunction but also heightens long-term risks of chronic kidney disease (CKD). The senescence of proximal tubular epithelial cells (PTECs) has a major impact on the occurrence and development of AKI. This review systematically analyzes existing evidence, which suggests that the senescence of PTECs may have a dual effect. Acute cellular senescence typically mitigates uncontrolled replication of damaged cells…

BACKGROUND: Cellular senescence is an irreversible cell cycle arrest that normal cells undergo in response to progressive shortening of telomeres, changes in telomeric structure, oncogene activation or oxidative stress and acts as an important tumour suppressor mechanism.
RESULTS: To identify the downstream effectors of the p53-p21 and p16-pRB tumour suppressor pathways crucial for mediating entry into senescence, we have carried out a loss-of-function RNA interference screen in conditionally immortalised human fibroblasts that can be induced to rapidly undergo senescence, whereas in primary …

SARS-Cov-2 infection, which has caused the COVID-19 global pandemic, triggers cellular senescence. In this study, we investigate the role of the SARS-COV-2 spike protein (S-protein) in regulating the senescence of RPE cells. The results showed that administration or overexpression of S-protein in ARPE-19 decreased cell proliferation with cell cycle arrest at the G1 phase. S-protein increased SA-β-Gal positive ARPE-19 cells with high expression of P53 and P21, senescence-associated inflammatory factors (e.g., IL-1β, IL-6, IL-8, ICAM, and VEGF), and ROS. Elimination of ROS by N-acetyl cysteine (…

Human Werner syndrome (WS) is an autosomal recessive progeria disease. A mouse model of WS manifests the disease through telomere dysfunction-induced aging phenotypes, which might result from cell cycle control and cellular senescence. Both p21Waf1/Cip1 (p21, encoded by the Cdkn1a gene) and p16Ink4a (p16, encoded by the Ink4a gene) are cell cycle inhibitors and are involved in regulating two key pathways of cellular senescence. To test the effect of p21 and p16 deficiencies in WS, we crossed WS mice (DKO) with p21 -/- or p16 -/- mice to construct triple knockout (p21-TKO or p16-TKO) mice. By s…

Cellular senescence is a stress response that cells can employ to resist cell death. Senescent cells rely on anti-apoptotic signaling for their survival, which can be targeted by senolytic agents, like the BCL-XL, BCL-2, BCL-W inhibitor ABT-263. However, the response to ABT-263 of senescent cancer cells ranges from highly sensitive to refractory. Using BH3 profiling, we identify here apoptotic blocks in cancer cells that are resistant to this senolytic treatment and discover a correlation between mitochondrial apoptotic priming and cellular sensitivity to ABT-263 in senescence. Intriguingly, A…

BACKGROUND: Fibroblast growth factor receptor 4 (FGFR4) is overexpressed in up to 50% of pancreatic ductal adenocarcinoma (PDAC) cases. BLU9931, a selective FGFR4 inhibitor, exerts cytotoxic effects on PDAC cells and induces cellular senescence. This study aimed to investigate the effects of BLU554, another selective FGFR4 inhibitor, whose efficacy and safety in hepatocellular carcinoma has been proven in clinical trials, on PDAC cell lines.
METHODS: BLU554 was applied to PDAC cell lines, and cytotoxicity in PK-1 and T3M-4 cells was assessed by counting viable cells. BLU554-induced senescence…

BACKGROUND: Radiation-induced skin injury (RISI) is a common complication of radiotherapy, affecting up to 95% of cancer patients. It manifests as acute erythema and ulceration or chronic fibrosis and telangiectasia, severely compromising patients' quality of life. The pathogenesis of RISI involves oxidative stress, inflammation, DNA damage, and cellular senescence. However, current treatments are largely supportive and fail to address underlying mechanisms. Berberine (BBR), a natural isoquinoline alkaloid, exhibits anti-inflammatory, antioxidant, and wound-healing properties, making it a prom…

Postmenopausal osteoporosis (PMOP) is a prevalent condition among postmenopausal women, closely linked to estrogen deficiency, aging, and oxidative stress. Cellular senescence, through mechanisms such as the senescence-associated secretory phenotype (SASP) and bone marrow stromal cells (BMSCs) differentiation imbalance, disrupts bone homeostasis in PMOP. Macrophages play a critical role in maintaining bone homeostasis. However, the extent of macrophage senescence in PMOP and the mechanisms by which it disrupts bone homeostasis have not yet been elucidated. Eldecalcitol (ED-71), a novel drug, h…

Osteoporosis is characterized by reduced bone mass and structural deterioration, leading to increased fracture risk, particularly in older adults. Parathyroid hormone (PTH) is a widely used anabolic therapy for osteoporosis; however, rapid bone loss after treatment discontinuation presents a significant clinical challenge. Cellular senescence has been implicated in age-related bone fragility. However, its role in PTH-induced bone remodeling and post-treatment bone loss remains unclear. This study aimed to investigate the effects of PTH administration frequency on bone microarchitecture and cel…

Ankylosing spondylitis (AS) is a chronic inflammatory disease possessing a morbid serum microenvironment with enhanced oxidative stress. Long-term exposure to an oxidative environment usually results in cellular senescence alone with cellular dysfunction. Mesenchymal stem cells (MSCs) are a kind of stem cell possessing strong capabilities for immunoregulation, and senescent MSCs may increase inflammation and participate in AS pathogenesis. The objective of this study was to explore whether and how the oxidative serum environment of AS induces MSC senescence. Here, we found that AS serum facili…

Inorganic arsenic (As) exposure is known to induce pulmonary fibrosis, yet the underlying mechanisms remain incompletely understood. This study investigated the crosstalk between sequestosome 1 (SQSTM1/p62) and nuclear factor erythroid 2-related factor 2 (Nrf2) in regulating cellular senescence and epithelial-mesenchymal transition (EMT) in As-exposed alveolar type II (AT2) cells. RNA sequencing analysis initially suggested that As exposure promotes EMT and senescence in MLE-12 cells. Subsequent experiments demonstrated that As exposure progressively decreased the epithelial marker E-cadherin …
