Dirofilaria immitis and Dirofilaria repens are mosquito-borne nematodes which infect primarily dogs as their main definitive hosts. They cause cardiopulmonary (D. immitis) or cutaneous (D. repens) dirofilariasis in canids and other carnivores and can accidentally be transmitted to humans where they can induce a variety of clinical outcomes depending on organ localization. Dirofilaria spp. infection in dogs was assessed using molecular methods (PCR and sequencing) to identify the different Dirofilaria species occurring in 200 dogs from Northern and Central Tunisia. The overall molecular prevale

OBJECTIVE: Paramyosin is a major protein produced by the metacestode cyst of Echinococcus granulosus, the causative agent of cystic hydatid disease. This protein has been shown to play an important role in modulating host immune responses. In this study, we attempted to characterize the noncoding sequence of the paramyosin gene.
METHODS: Genomic DNA was isolated from G1 Iranian hydatid cysts. A DNA fragment of 3200 bp in length was amplified from the paramyosin gene. The polymerase chain reaction (PCR) product was cloned to the pTZ57T vector and sequenced by M13 primers and then compared with

Economical animal breeding programs are important for achieving maximum gain, and any factors resulting in economical loss should be minimized or eliminated. An organism of concern is the liver fluke, Fasciola hepatica, which causes decreased yield and even death in sheep and dairy cattle. In an effort to eliminate or minimize the detrimental effect of this parasite in animals, it is important to understand the genetic diversity within the liver flukes and the relationship between this parasite and the host in the particular geographic area. The aim of this study was to explore genetic diversi

Paragonimosis is an important food-borne zoonosis especially in Asian countries. Among Paragonimus species, Paragonimus westermani followed by P. skrjabini complex are the major pathogens for human paragonimosis in Asia. In addition, P. heterotremus is an important pathogen in southern China and the Indochina Peninsula and is the only proven species to cause human paragonimosis in Vietnam. During a recent survey in Yenbai Province in northern Vietnam, we found small and large types of Paragonimus metacercariae often concurrently in mountainous crabs, Potamiscus tannanti. Adult worms from those

In 1975, an ancient corpse buried in 167 BC was found at Jiangling County, Hubei Province of China. The eggs of Clonorchis sinensis found in the gall bladder of the corpse were preserved well. In the present paper, we extracted the genomic DNA from the ancient eggs and modern eggs, respectively, and the internal transcribed spacer 1 and 2 (ITS1 and ITS2) at ribosomal RNA genes were studied. The results show that ITS2 sequences from the ancient sample were identical with those from modern samples, but in ITS1 differences in 15 nucleotide positions were found between the ancient and modern sampl

Esophagostomiasis in humans due to infection with Oesophagostomum bifurcum (nodular worm) is of major human health significance in northern Togo and Ghana, where Necator americanus (human hookworm) also exists at high prevalence. Yet, very little is known about the transmission patterns of O. bifurcum, which is in part due to the difficulties in diagnosis and in differentiating some life-cycle stages of O. bifurcum from N. americanus using morphological features. As a first step toward developing a molecular-diagnostic assay, it was evaluated whether ribosomal (r)DNA could provide genetic mark

Intraspecific genetic variation among 14 geographic isolates of Schistosoma mansoni was quantified using a molecular marker to examine individual genotypes. Genetic crosses demonstrated maternal inheritance of S. mansoni DNA element pSM750. This element revealed diagnostic banding profiles, which allowed accurate strain identification. Most strains had similarity indices greater than 0.75 indicating that within-strain variation in these laboratory parasite populations was low. However, individual parasites from the NMRI strain were quite diverse (S = 0.40). Genetic heterogeneity among strains

Mummichog (Fundulus heteroclitus) hosts a diverse array of parasites, including several species of Gyrodactylidae. We surveyed populations of mummichogs across three estuarine systems in South Carolina, USA, for gyrodactylids and studied their taxonomy, phylogenetics, and evolutionary history. Molecular analysis using concatenated sequences of 18S, ITS, and 5.8S rDNA fragments and morphology support the synonymization of Fundulotrema with Swingleus, as the former was shown paraphyletic. Five species of Gyrodactylidae were encountered. Gyrodactylus kritskyi n. sp. is described and four other sp

The trematode Maritrema gratiosum Nicoll, 1907 has a heteroxenous life cycle comprising a marine gastropod as its first intermediate host, a barnacle as its second intermediate host, and a bird as its definitive host. Globally, the species is widely distributed from subtropical waters of the South China Sea to arctic waters of the Barents Sea. In European waters (namely, western Europe and the Barents Sea), the complete life cycle of M. gratiosum is known. Elsewhere in its range, however, its life cycle remains incompletely understood. Parasitic nematodes of the family Onchocercidae (superfamily Filarioidea) have evolved alongside vertebrate hosts for millions of years. While morphological methods have played a central role in identifying species, many taxa remain cryptic and are challenging to differentiate, complicating diagnosis and the understanding of their epidemiology. Among the clinically relevant genera are Onchocerca and Dirofilaria, the latter being responsible for illnesses such as heartworm disease in canines and rare zoonotic infections in humans. Despite advances in our knowledge of the canine heartworm (Dirof
