Chemical Synthesis of Native ADP-Ribosylated Oligonucleotides Enables Analysis of DNA ADP-Ribosylation Hydrolase Specificity.

ADP-ribosylation (ADPr) is a modification by which an ADP-ribose moiety is conjugated to different molecules by ADP-ribosyltransferases (ARTs) to control various cellular processes in organisms from all kingdoms of life. While traditionally considered primarily as a post-translational modification of proteins, recent evidence has demonstrated that ADPr of nucleic acids (DNA and RNA) has emerged as a widespread type of ADPr. However, the precise biological roles and underlying mechanisms remain elusive, largely due to the lack of ADP-ribosylated nucleic acid molecular tools. Inspired by the nat

On the origin of the ionic strength control of the motility of kinesin-14.

Kinesin-14 motors are a special class of the kinesin family proteins that walk toward the minus end along the microtubule. It has the reverse directionality compared to the general conventional kinesins. The motility of kinesin-14 has been found to depend on the ionic strength. That is, when the ionic strength changes, the motility of the protein changes significantly. Understanding the origin of such effect is important both as a fundamental problem and as a part of the overall understanding of intracellular transportation. Here, we explore the molecular origin behind the lowering of motility

Deciphering the Specificity of Reversible DNA-Phosphate ADP-Ribosylation via the Precise Synthesis and Enzymatic Profiling of Nucleotide-Phospho-ADP-Ribosyl Probes.

ADP-ribosylation (ADPr), long recognized as a canonical protein post-translational modification, has recently expanded to include targeting nucleic acids, uncovering a diverse landscape of noncanonical biological functions. Emerging evidence suggests that ADPr at the 5'-phosphate terminus of DNA is implicated in the DNA damage response, yet understanding its precise molecular function has been hampered by the lack of structurally defined chemical probes. Here, we report the stereoselective synthesis of deoxynucleotide-phospho-ADPr (dN-P-ADPr) probes, representing native fragments of terminal D

Integrative network toxicology identifies SLC5A2 as a key mediator of tetrasodium pyrophosphate-induced renal fibrosis.

Tetrasodium pyrophosphate (TSPP) is a widely used phosphate-based food additive that may contribute to exogenous inorganic phosphate exposure, however, its potential role in renal fibrosis remains poorly understood. This study investigated the molecular targets and mechanisms underlying TSPP-induced renal fibrosis using integrative network toxicology combined with experimental validation. Network toxicology identified 39 overlapping targets, which were mainly enriched in renal tubular ion transport, ion homeostasis, and mineral metabolism. Target prioritization and transcriptomic analysis furt

Cytoprotection as an Innovative Therapeutic Strategy to Cardiogenic Shock: Exploring the Potential of Cytidine-5-Diphosphocholine to Mitigate Target Organ Damage.

BACKGROUND: Preservation of organ function and viability is a crucial factor for survival in cardiogenic shock (CS) patients. There is not information enough on cytoprotective substances that may delay organs damage in CS. We hypothesize that cytidine-5-diphosphocholine (CDP-choline) can act as a cytoprotective pharmacological measure that diminishes the target organ damage. So, we aimed to perform a review of works carried out in our institution to evaluate the effect of therapeutic cytoprotection of the CDP-choline. SUMMARY: CDP-choline is an intermediate metabolite in the synthesis of phos

Resurrecting ancestral antibiotics: unveiling the origins of modern lipid II targeting glycopeptides.

Antibiotics are central to modern medicine, and yet they are mainly the products of intra and inter-kingdom evolutionary warfare. To understand how nature evolves antibiotics around a common mechanism of action, we investigated the origins of an extremely valuable class of compounds, lipid II targeting glycopeptide antibiotics (GPAs, exemplified by teicoplanin and vancomycin), which are used as last resort for the treatment of antibiotic resistant bacterial infections. Using a molecule-centred approach and computational techniques, we first predicted the nonribosomal peptide synthetase assembl

UDP-glucuronosyltransferase is involved in susceptibility of Chironomus kiiensis Tokunaga, 1936 (Diptera: Chironomidae) to insecticides.

UDP-glucuronosyltransferases (UGTs) could transform various exogenous and endogenous compounds, which help detoxification of pesticides in insects. To investigate the role of UGTs in the detoxification metabolism of insecticides in Chironomus kiiensis, CkUGT302M1, CkUGT302N1, CkUGT308N1 and CkUGT36J1 genes were identified with 1449-1599 bp encoding 482-532 amino acids. Four UGT genes shared 40.86∼53.36% identity with other homologous insect species, and expressed in all developmental stages, notably in the larval and adult stages. Expression of CkUGTs was higher in the gastric caecum, midgut a

Ablation of Pyrophosphate Regulators Promotes Periodontal Regeneration.

Biomineralization is regulated by inorganic pyrophosphate (PPi), a potent physiological inhibitor of hydroxyapatite crystal growth. Progressive ankylosis protein (ANK) and ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1) act to increase local extracellular levels of PPi, inhibiting mineralization. The periodontal complex includes 2 mineralized tissues, cementum and alveolar bone (AB), both essential for tooth attachment. Previous studies demonstrated that loss of function of ANK or ENPP1 (reducing PPi) resulted in increased cementum formation, suggesting PPi metabolism may be a targe

Citicoline and Retinal Ganglion Cells: Effects on Morphology and Function.

BACKGROUND: Retinal ganglion cells (RGCs) are the nervous retinal elements which connect the visual receptors to the brain forming the nervous visual system. Functional and/or morphological involvement of RGCs occurs in several ocular and neurological disorders and therefore these cells are targeted in neuroprotective strategies. Cytidine 5-diphosphocholine or Citicoline is an endogenous compound that acts in the biosynthesis of phospholipids of cell membranes and increases neurotransmitters' levels in the Central Nervous System. Experimental studies suggested the neuromodulator effect and the

RNA-dependent RNA polymerases from flaviviruses and Picornaviridae.

Flaviviruses and picornaviruses are positive-strand RNA viruses that encode the RNA-dependent RNA polymerase (RdRp) required for replicating the viral genome in infected cells. Because of their specific and essential role in the virus life cycle, RdRps are prime targets for antiviral drugs. Recent structural data have shed light on the different strategies used by RdRps from flaviviruses and Picornaviridae to initiate RNA polymerization. New details about the catalytic mechanism, the role of metal ions, how these RdRps interact with other nonstructural (NS) viral and host-cell proteins as well

Evaluation of phosphate fertilizers for the stabilization of cadmium in highly contaminated soils.

The efficiency of three phosphate fertilizers including triple superphosphate (TSP), diammonium phosphate (DAP), and phosphate rock (PR) as stabilizing agents of cadmium-contaminated soils has been assessed in this study. Two types of assessment criteria, (a) the reduction of leachable cadmium concentration; and (b) the changes in Cd association with specific operational soil fraction based on the sequential extraction data, are used in the evaluation of stabilization performance of each fertilizer. Results of the study showed that after the 60-day stabilization, the leachable concentrations o

Noncooperative stabilization effect of phalloidin on ADP.BeFx- and ADP.AlF4-actin filaments.

Actin plays important roles in eukaryotic cell motility. During actin polymerization, the actin-bound ATP is hydrolyzed to ADP and P i. We carried out differential scanning calorimetry experiments to characterize the cooperativity of the stabilizing effect of phalloidin on actin filaments in their ADP.P i state. The ADP.P i state was mimicked by using ADP.BeF x or ADP.AlF 4. The results showed that the binding of the nucleotide analogues or phalloidin stabilized the actin filaments to a similar extent when added separately. Phalloidin binding to ADP.BeF x- or ADP.AlF 4-actin filaments further

Ligand-specific glucocorticoid receptor activation in human platelets.

Few studies have addressed the effects of classical anti-inflammatory glucocorticoids on platelet function. Here, we report for the first time that human platelets contain the glucocorticoid receptor (GR) as identified by a combination of biochemical and functional techniques. Ligand-binding studies revealed the presence of a high- and low-affinity binding site for [3H]-dexamethasone in platelets. The 2 GR ligands prednisolone and dexamethasone competed for [3H]-dexamethasone binding, as did the mineralocorticoid aldosterone. However, while prednisolone (1-10 microM) reduced adenosine diphosph
