Examination of social network members' influence on daily drinking: A pilot study.

OBJECTIVE: Research on young adult alcohol use often overlooks the influence of specific social network members on daily alcohol use. This pilot study combined egocentric social network methods with a daily diary design to examine how network members influence drinking at the day level. METHODS: Participants (N = 21) identified six social network members they frequently drank with and saw in person and then reported on these individuals in a 21-day study. Daily reports captured self-reported alcohol use and social network members presence and alcohol use from the previous day. RESULTS: Acros…

Functional remodeling of the parasubthalamic nucleus drives alcohol drinking escalation in dependence.

The neurocircuitry changes mediating the development and maintenance of an alcohol use disorder are complex and dynamic. The parasubthalamic nucleus (PSTN), a small nucleus of the posterior lateral hypothalamus best known for suppressing appetite, is interconnected with brain regions disrupted in addiction; yet its potential role in the regulation of alcohol consumption had never been examined. Here we show that the PSTN exerts potent control over alcohol drinking in mice. Remarkably, the influence of endogenous PSTN activity on voluntary alcohol consumption switches from inhibitory to stimula…

"We're all hammered in Chile": Drinking norms and tactics from adolescents' perspectives.

BACKGROUND: Understanding adolescents' perspectives on how alcohol policies and the normalization of alcohol consumption affect their behavi … cents' early experiences with alcohol in the home, while these attitudes-together with parents' own drinking behaviors-further reinforce the normalization of alcohol use. CONCLUSIONS: Prevention strategies for adolescents should address the normalization of alcohol use in Chile and its embedding in social contexts, such as families. The enforcement of alc …

Alcohol-Induced Blackouts and Subsequent Drinking Trajectories: Evidence From Three Longitudinal Samples.

BACKGROUND: Alcohol-induced blackouts are widely recognized as markers of acute intoxication and elevated risk for alcohol-related harm. Blackout experiences may also represent psychologically salient events capable of influencing subsequent drinking behavior. The present study examined whether recent blackout predicts changes in alcohol use, alcohol-related consequences, and blackouts over time. METHOD: Data were drawn from three independent longitudinal samples of college student drinkers (Ns = 568, 484, and 200). Generalized linear mixed models with negative binomial distributions were use…

Hippocampus-Ventral Striatum Connectivity Is Associated With Binge Drinking Frequency in Adults.

Contextual information, such as spatial locations and social groups, are known to propagate reward-seeking behavior including binge drinking. Animal models suggest that context-evoked drug and alcohol seeking emerges from hippocampal projections to the ventral striatum (i.e., nucleus accumbens). While binge drinking has been associated with adverse structure and function of both the ventral striatum and hippocampus, the anatomical relevance of this circuit in humans has largely been inferred from volumetric studies investigating these regions separately. Using diffusion MRI tractography in adu…

Associations among alcohol sensitivity, acquired tolerance, and drinking outcomes in late adolescence.

Alcohol tolerance is a core diagnostic feature of alcohol use disorder, yet little research has investigated its role in risky drinking during late adolescence. Low sensitivity to alcohol sedation is also linked to alcohol use disorder risk, but few studies have examined how acquired tolerance and alcohol sensitivity concurrently relate to heavy drinking. This study examined the concurrent and prospective associations of acquired tolerance and alcohol sensitivity with heavy drinking among late adolescent heavy drinkers (N = 308, ages 18-21, 52.6% female). At baseline, participants completed se…

Organ-Specific Proteomic Aging Clocks Reveal Complexities Underlying the Healthy Drinking Paradox.

The "Работает drinking" paradox refers to situations where alcohol consumption is misleadingly associated with better health outcomes, a pattern frequently observed in observational studies that are prone to a range of confounding factors. Some recent studies developing organ-specific proteomic aging clocks report that alcohol consumption is linked to slower aging in select organs, even as it accelerates aging in others, thus echoing this paradox, with substantial discrepancies across such studies. In this Perspective, I summarize findings to date, discuss potential reasons for these contradict…

Fluoride Levels in Austrian Drinking Water Are Insufficient for Effective Caries Prevention.

Background/Objectives: Fluorides play a well-established role in preventing dental caries, primarily by enhancing enamel resistance and inhibiting demineralization. Drinking water is among the most important sources of systemic fluoride intake. In 1993 and 2007, national analyses of Austrian drinking water revealed fluoride levels below 0.5 mg/L in almost all regions, which is insufficient for effective caries prevention. The present study aimed to re-examine the fluoride concentration in Austrian drinking water. Methods: Drinking water was collected in a total of 1985 Austrian municipalities.…

Sewage intrusion into drinking water distribution systems: implications for water resource management.

Sewage infiltration of drinking water distribution systems (DWDS) is a major challenge for global water resource management, exacerbated by outdated infrastructure and rising water pressure. According to studies, even small sewage intrusion can result in major microbiological changes, such as appreciable increases in bacterial biomass, bacterial diversity, and dissolved organic matter (DOM) composition. These changes intensify the risk of waterborne infections and complicate treatment procedures required for ensuring safe drinking water. The present work is a review/state of science that synth…

Differential impact of recent heavy drinking on first and recurrent acute pancreatitis.

BACKGROUND: While alcohol is known to sensitize the pancreas to acute injury, the role of short-term episodic drinking in regular drinkers is unknown. METHODS: We conducted a case-crossover study to (1) determine the hazardous period of drinking prior to a first episode of acute pancreatitis (FAP) or recurrent acute pancreatitis (RAP) and (2) evaluate the dose-response association between short-term drinking and FAP/RAP. Patients hospitalized for FAP/RAP with an AUDIT-C score of ≥3 were enrolled. Recent and lifetime drinking history were collected through interviews. Drinking prior to the ind…

Sensory Preference of Drinking Water Influenced by Subthreshold-Level Mineral Salt Mixtures.

This study investigated the impact of subthreshold-level mixtures of KCl and MgSO4 on the sensory preference of drinking water and identified key sensory drivers. Absolute thresholds were first determined, followed by sensory evaluation of individual and blended salt solutions using a 9-point hedonic scale and CATA test. Design of Experiments (DOE) approach with quadratic polynomial regression optimized the binary mixtures. Results showed samples near the absolute thresholds (0.95 mg/L KCl; 0.65 mg/L MgSO4) were most preferred. Beyond these thresholds, mineral salt concentration negatively cor…

Exploring associations between drinking contexts and alcohol consumption: An analysis of photographs.

Drinking contexts are theorized to represent a core factor driving hazardous consumption and ultimately susceptibility to alcohol use disorder. Yet capturing and characterizing contextual influences on alcohol consumption has posed a significant challenge. In this study, we employ objective ambulatory assessment methods to test a multiaxial framework for understanding contextual influences on drinking. Specifically, we propose a novel SPAIS framework, comprising five exogenous, objectively measurable domains of drinking context (schedule/time, physical, activity, interpersonal, and substance/d…

ADHD Symptomatology is Associated with Alcohol Use and Consequences via Drinking Norms.

Background: Research suggests individuals with ADHD are at risk for alcohol use and related problems and are more susceptible to peer influence. Although previous research has focused on the relationship between alcohol use, related problems, and ADHD symptomatology, scant research has examined whether descriptive drinking norms (i.e., perception of others quantity of drinking) and level of alcohol use mediate this association. Objectives: Consequently, the current study tested a sequential mediation model to examine whether ADHD symptoms were associated with alcohol-related problems via drink…

Identifying drinking water safety hazards throughout an Arctic community's water supply system.

Arctic communities, ecosystems, and infrastructure are increasingly coming under scrutiny due to ongoing attempts at decolonization and reconciliation with Indigenous groups and the potential for geopolitical conflict in the region. There is also increased attention to the region from various industries because of its economic potential, especially as climate change allows for increased access for development. This increase in attention has highlighted the lack of understanding about drinking water sources and infrastructure in the Arctic. In this study we conducted extensive water quality mon…

Effects of binge-like ethanol drinking on nest building behavior in mice.

Nest building is a natural behavior that can readily be analyzed in mice in the home cage environment. Nest building is involved in thermoregulation, positive motivational states, and motor function, and has been proposed as an index for ethanol withdrawal severity in mice. However, nest building outcomes after voluntary ethanol consumption have not been examined. Here, we tested male and female C57BL/6J mice on a 4-day drinking in the dark (DID) paradigm of binge-like drinking with either ethanol or a water control and analyzed nest scores at two time points (48 hours and 7 days) after the la…

Sex differences in nucleus accumbens circuitry engaged with binge-like ethanol drinking.

RATIONALE: Women tend to progress from initial alcohol use to Alcohol Use Disorder (AUD) more quickly than men, highlighting the need to study sex differences in models of early-stage alcohol use. In humans and rodents, the nucleus accumbens (NAc) regulates alcohol binge drinking, a risk factor for developing AUD. However, it is unknown whether similar brain regions and NAc inputs are engaged in males and females during binge-like drinking. METHODS: We labeled NAc inputs with a viral retrograde tracer (GFP) and quantified whole-brain c-Fos to determine the regions and NAc inputs differentiall…

10-Butyl Ether Minocycline (BEM) Reduced Ethanol Consumption in Binge Drinking Mice.

Alcohol use disorder (AUD) is a complex brain disease with high morbidity, mortality, and limited treatment options, causing significant soc … es. We hypothesized that BEM would reduce high ethanol consumption in common murine models of binge drinking without affecting ethanol elimination. The Drinking-In-the-Dark (DID) paradigm was used to assess BEM's efficacy in reducing binge-like ethanol consumption. BEM dose-responsively reduced ethanol consumption in both female and male mice.
