Acetazolamide alleviates motion sickness by inhibiting inner ear carbonic anhydrase 2 and reducing endolymph volume.

Motion sickness is common in aerospace, aviation and maritime operations, and travel by vehicles or ships. Existing preventive and therapeutic drugs for motion sickness induce central nervous system (CNS)-related side effects; therefore, there is an urgent need to find new anti-motion sickness targets and to develop novel drugs with reduced adverse effects. In this study, we found that rotational stimulation significantly upregulated carbonic anhydrase 2 (CA2) expression in the inner ears of guinea pigs and mice. Pretreatment with acetazolamide (AZ), an inhibitor of carbonic anhydrase, effecti

Transcriptomic profiling of inner ear hair cell spontaneous regeneration reveals the key role of dlx5a.

Sensory hair cells (HCs) are susceptible to damage, typically causing permanent hearing and balance deficits in mammals due to limited regeneration. Understanding spontaneous regeneration in non-mammalian models offers key insights for restoring sensory function. Here, we establish a zebrafish semicircular canal crista HC injury model and characterize subsequent regeneration and functional recovery processes. Lineage tracing demonstrates that regenerated crista HCs derive from preexisting and newly formed supporting cells (SCs) that emerged after crista HC injury via both mitotic and non-mitot

Rapid Postmortem Temporal Bone Extraction and Dissection of Human Inner Ear Organs From Organ Donors.

We developed a postmortem protocol for temporal bone extraction and dissection in organ donors, enabling atraumatic bilateral temporal bone retrieval within 1-2 h of ischemia, followed by stepwise microsurgical dissection of neurosensory organs. Combining rapid extraction, abbreviated decalcification, and atraumatic microsurgical dissection, this protocol establishes a scientifically versatile framework for human inner ear tissue acquisition.

Inner Ear Anatomy of Caenagnathidae (Theropoda: Oviraptorosauria) Emphasizes Mosaic Evolution of the Avian Neurosensory System.

The endocranial anatomy of oviraptorosaurian theropods, and their inferred sensory adaptations, has played a key role in understanding the origin of bird-like senses among avian ancestors. However, within oviraptorosaurs, sampling has thus far been limited, and a major gap exists in our understanding of the endocranial anatomy of an enigmatic subclade, Caenagnathidae. Here, we describe the inner ear morphology of three caenagnathid oviraptorosaurs, spanning a wide range of stratigraphy and body size. Our data show that the incipiently bird-like inner ears of caenagnathids were relatively simil

Exploring AAV-Mediated Gene Therapy for Inner Ear Diseases: from Preclinical Success to Clinical Potential.

Hearing loss imposes a significant global health burden and often results from genetic factors and various external influences, such as noise exposure and the use of ototoxic drugs. Recent advancements in adeno-associated virus (AAV)-mediated gene therapy offer promising new avenues for the treatment of inner ear diseases. Clinical trials of AAV-mediated gene therapies show impressive preliminary results, although further research is needed to evaluate the safety and long-term effects of these therapies. Preclinical AAV-mediated gene therapy is notable for its high transduction efficiency and

A diagnostic pitfall: Advanced labyrinthitis ossificans simulating congenital inner ear malformation on CT and MRI.

Labyrinthitis ossificans (LO) is a rare pathological ossification of the membranous labyrinth, typically arising after inflammatory or infectious insults to the inner ear. We present a case of a 65-year-old male with type 2 diabetes presenting with progressive unilateral sensorineural hearing loss and a history of recurrent childhood ear infections treated with repeated lavages. Imaging revealed advanced ossification of the right cochlea, vestibule, and semicircular canals, despite no history of meningitis or trauma. Magnetic resonance imaging (MRI) revealed near-complete loss of the normal fl

Prevalence and Intriguing Clinical Profiles of Autoimmune Inner Ear Diseases in Sudden Sensorineural Hearing Loss.

INTRODUCTION: Sudden sensorineural hearing loss (SSNHL) is idiopathic in 70-90% of cases with 10-30% linked to vascular injuries, viral infections, or autoimmune inner ear disease (AIED). AIED contributes to less than 1% of all hearing losses, categorized into primary, affecting only the inner ear, and secondary, associated with systemic autoimmune diseases (SAID). This study examines the prevalence and features of AIED in SSNHL in a tertiary referral center. MATERIALS AND METHODS: We conducted a prospective study from November 2021 to December 2022 including SSNHL patients who exhibited symp

Ultrasound-induced microbubble cavitation via a transcanal or transcranial approach facilitates inner ear drug delivery.

BACKGROUND: Ionizing radiation induces a variety of genetic damages through the formation free radicals such as reactive oxygen species (ROS). Appropriate antioxidant intervention may inhibit or reduce free radical toxicity and thus offer protection against radiation. Rutin (RUT) and quercetin (QRT) are flavonoids known to be potent dietary antioxidants. METHODS: The present study tested the antigenotoxic effect of RUT and QRT in vivo against radiation- induced chromosomal damage. Swiss albino mice were administered orally with RUT and QRT (10 and 20 mg/kg b.wt.) once daily for five consecuti

Noise-induced hearing loss in zebrafish: investigating structural and functional inner ear damage and recovery.

Exposure to continuous moderate noise levels is known to impair the auditory system leading to Noise-Induced Hearing Loss (NIHL) in animals including humans. The mechanism underlying noise-dependent auditory Temporary Threshold Shifts (TTS) is not fully understood. In fact, only limited information is available on vertebrates such as fishes, which share homologous inner ear structures to mammals and have the ability to regenerate hair cells. The zebrafish Danio rerio is a well-established model in hearing research providing an unmatched opportunity to investigate the molecular and physiologica

Rescue of Hearing by Gene Delivery to Inner-Ear Hair Cells Using Exosome-Associated AAV.

Adeno-associated virus (AAV) is a safe and effective vector for gene therapy for retinal disorders. Gene therapy for hearing disorders is not as advanced, in part because gene delivery to sensory hair cells of the inner ear is inefficient. Although AAV transduces the inner hair cells of the mouse cochlea, outer hair cells remain refractory to transduction. Here, we demonstrate that a vector, exosome-associated AAV (exo-AAV), is a potent carrier of transgenes to all inner ear hair cells. Exo-AAV1-GFP is more efficient than conventional AAV1-GFP, both in mouse cochlear explants in vitro and with

Vasopressin induces endolymphatic hydrops in mouse inner ear, as evaluated with repeated 9.4 T MRI.

From histopathological specimens, endolymphatic hydrops has been demonstrated in association with inner ear disorders. Recent studies have observed findings suggestive of hydrops using MRI in humans. Previous studies suggest that vasopressin may play a critical role in endolymph homeostasis and may be involved in the development of Ménière's disease. In this study we evaluate the effect of vasopressin administration in vivo in longitudinal studies using two mouse strains. High resolution MRI at 9.4 T in combination with intraperitoneally delivered Gadolinium contrast, was performed before and

Evidence for changes in beta- and gamma-actin proportions during inner ear hair cell life.

Cytoplasmic actin isoforms beta (β-) and gamma (γ-) perform crucial physiological roles in inner ear hair cells (HC). The stereocilium, which is structured by parallel actin filaments composed of both isoforms, is the responsive organelle to mechanical stimuli such as sound, gravity and head movements. Modifications in isoform proportions affect the function of the stereocilia as previously shown in genetic studies of mutant mice. Here, immunogold labeling TEM studies in mice showed that both β- and γ-actin isoforms colocalize throughout stereocilia actin filaments, adherens junctions and cuti

Experimental exploration of the soft tissue conduction pathway from skin stimulation site to inner ear.

OBJECTIVES: Auditory sensation can be elicited by air conduction (AC) and by bone conduction (BC). It is also possible to elicit such responses by applying the standard clinical bone vibrator to the skin over soft tissue sites on the head, neck, or thorax of humans and animals. This mode of auditory stimulation has been called soft tissue conduction (STC). This study was designed to investigate the pathway between soft tissue sites and the ear. METHODS: The air in the middle ear was replaced with saline solution in an animal with unique anatomy--the fat sand rat, in which about 70% of a thin-

Autoantibodies against inner ear proteins in patients with delayed endolymphatic hydrops and unilateral juvenile deafness.

CONCLUSIONS: Patients with the contralateral type of delayed endolymphatic hydrops (DEH) may undergo an autoimmune attack against the other inner ear. As patients with unilateral juvenile deafness show no progression, despite lengthy observation, the autoantibody against the 68-kDa protein may be unrelated to the pathogenesis of DEH. OBJECTIVE: The contralateral type of DEH is believed to have an autoimmune etiology, and sometimes develops from unilateral juvenile deafness. The purpose of this study was to determine whether autoantibodies are pathogenetically important in DEH. MATERIAL AND M
