Targeting dopamine D2 receptors to alter fear extinction in adolescent rats.

Anxiety disorders emerging in adolescence are especially challenging to treat, partly due to adolescents having poor extinction recall and high relapse rates following exposure therapy, relative to other ages. Despite its clear clinical significance, little work has examined treatments to enhance extinction in adolescence. Dopamine, a key neurotransmitter involved in learning and memory, plays a critical role in fear extinction in adults, but its role in extinction in adolescents has not been well-explored. In a series of experiments, we investigated the effects of sulpiride, a dopamine antago…

Piriform cortex gates learned fear through a brain-spleen neuroimmune axis.

Maladaptive learned fear responses to stress underlie several debilitating neuropsychiatric disorders. Here, we identify a brain-to-spleen neural pathway that mediates learned fear through coordinated neuroimmune interactions. Using a chronic acquired olfactory stress (CAOS) model, we demonstrate that sustained enhancement of the piriform cortex (Pir) excitability contributes to the transformation of stress-associated olfactory inputs into learned fear-avoidance behavior. Through comprehensive neural tracing approaches, we mapped a functional tetrasynaptic circuit (Pir→ventral hippocampus CA1 …

Assessing spider fear: Validity of a novel computerized behavioral avoidance test.

Spider phobia is one of the most common specific phobias and can be highly detrimental to one's quality of life. To evaluate the efficacy of treatments, reliable pre- and post-treatment assessments are needed. Behavioral avoidance tests (BATs), where one is asked to approach a spider, are commonly used as such measures. With the increasing popularity of computerized interventions, there is a demand for quick assessments for fear of spiders, that can be completed using computers or smartphones. Here, we tested the validity of a novel computerized BAT (cBAT) that we introduced in our previous wo…

PKCα-deficient mice show altered synaptic signaling and impaired fear extinction.

Protein kinase C alpha (PKCα), encoded by Prkca, has been implicated in neuronal signaling, plasticity, and memory. Human genetic studies id … ampus-dependent spatial memory, increased anxiety-like behavior, and a marked deficit in contextual fear extinction. These findings identify PKCα as part of a signaling program associated with synaptic function and support a role for PKCα-dependent processes in adaptive updating of learned fear.

Pre-Test Fear and Confidentiality in Premarital HIV Screening in Somalia.

Somalia has a low-level HIV epidemic, but low prevalence does not necessarily mean that HIV testing is socially or psychologically low risk. This commentary examines "pre-test fear", defined here as anticipatory concern before HIV testing about possible health, relational, social, or confidentiality consequences of being tested or receiving a reactive result. Somalia-specific evidence documents HIV-related stigma, non-consented disclosure, misinformation, and confidentiality concerns, but direct evidence on pre-test fear and the prevalence or regulation of premarital HIV testing remains limite…

Brain Mechanisms of Fear Reduction Underlying Habituation to Pain in Humans.

Habituation to painful stimuli reflects an endogenous pain alleviation mechanism, and reduced pain habituation has been demonstrated in many chronic pain conditions. In ethology, animals exhibit reduced fear responses while habituating to repeated threatening stimuli. It remains unclear whether pain habituation in humans involves a fear reduction mechanism. In an fMRI experiment, we investigated pain-related brain responses before and after the development of habituation to pain induced by repetitive painful stimulation in healthy adults. In another behavioral experiment, we examined emotional…

Teaching Autistic Adolescents to Identify Fear and Anger: A Preliminary Study.

Climate change poses significant threats to European agricultural production, with increasing frequency and severity of adverse weather events impacting crop yields. Agroforestry, the integration of woody elements into agricultural systems, is recognized as a vital agroecological approach for both climate change mitigation and adaptation, yet key mechanistic and long-term performance questions remain unresolved. This study utilized the mechanistic Hi-sAFe model to simulate 100 years (2001-2100) of silvoarable agroforestry performance at Wakelyns farm in southeastern England, focusing on winter

Endocannabinoids inhibit contextual fear memory generalization via hippocampal GABAergic synaptic transmission.

Memory generalization allows an organism to adapt to new conditions, but overgeneralization of fear or traumatic experiences can be detrimental to survival and contributes to the development of various mental disorders. However, the cellular and molecular mechanisms underlying fear memory generalization, especially in the hippocampus, remain largely unknown. In this study, utilizing a well-established mouse model of fear memory generalization, we investigated the role of endocannabinoids (eCBs)-mediated GABAergic synaptic inputs to hippocampal pyramidal neurons in regulating contextual fear me…

Developmental and molecular contributions to contextual fear memory emergence in mice.

Cognitive impairment is a common phenotype of neurodevelopmental disorders, but how these deficits arise remains elusive. Determining the onset of discrete cognitive capabilities facilitates studies in probing mechanisms underlying their emergence. The present study analyzed the emergence of contextual fear memory persistence (7-day memory retention) and remote memory (30-day memory retention). There was a rapid transition from postnatal day (P) 20 to P21, in which memory persistence emerged in C57Bl/6J male and female mice. Remote memory was present at P23, but expression was not robust compa…

Post-conditioning sleep deprivation facilitates delay and trace fear memory extinction.

Trace and delay auditory fear conditioning involve different memory association strategies based on working memory involvement; however, their differences in long-term processing through sleep and extinction training remain unclear. While females often exhibit more persistent fear, complicating psychiatric treatment, most studies have primarily focused on how sleep affects initial recall in male mice. We investigated the three-way interaction between tests (trace vs. delay), sleep states, and sex during initial recall, extinction, and post-extinction remote recall. A six-hour post-conditioning…

Cerebellar interneurons control fear memory consolidation via learning-induced HCN plasticity.

While synaptic plasticity is considered the basis of learning and memory, modifications of the intrinsic excitability of neurons can amplify the output of neuronal circuits and consequently change behavior. However, the mechanisms that underlie learning-induced changes in intrinsic excitability during memory formation are poorly understood. In the cerebellum, we find that silencing molecular layer interneurons completely abolishes fear memory, revealing their critical role in memory consolidation. The fear conditioning paradigm produces a lasting reduction in hyperpolarization-activated cyclic…

Changes in brain rhythms and connectivity tracking fear acquisition and reversal.

Fear conditioning is used to investigate the neural bases of threat and anxiety, and to understand their flexible modifications when the environment changes. This study aims to examine the temporal evolution of brain rhythms using electroencephalographic signals recorded in healthy volunteers during a protocol of Pavlovian fear conditioning and reversal. Power changes and Granger connectivity in theta, alpha, and gamma bands are investigated from neuroelectrical activity reconstructed on the cortex. Results show a significant increase in theta power in the left (contralateral to electrical sho…

The cerebellum and fear extinction: evidence from rodent and human studies.

The role of the cerebellum in emotional control has gained increasing interest, with studies showing it is involved in fear learning and memory in both humans and rodents. This review will focus on the contributions of the cerebellum to the extinction of learned fear responses. Extinction of fearful memories is critical for adaptive behaviour, and is clinically relevant to anxiety disorders such as post-traumatic stress disorder, in which deficits in extinction processes are thought to occur. We present evidence that supports cerebellar involvement in fear extinction, from rodent studies that …
