Vigabatrin-Induced Retinal Toxicity: Unraveling the Gamma-Aminobutyrate-Vitamin B6 Axis and Metabolic Crosstalk.

Vigabatrin (VGB) is an antiseizure medication used as an add-on therapy for refractory focal seizures. Despite its clinical efficacy, VGB use is limited by retinal toxicity that may lead to permanent visual loss. This narrative literature review aims to elucidate the emerging molecular and metabolic mechanisms underlying VGB-induced retinal toxicity, with particular emphasis on the interplay among gamma-aminobutyrate (GABA) accumulation, taurine depletion, ornithine metabolism dysregulation, and vitamin B6-dependent pathways to identify potential therapeutic targets for preventing visual impai

Regional-specific GABAergic imbalance in cisplatin-induced neuropathy and antiallodynic effects of intraplantar baclofen in rats.

Chemotherapy-induced peripheral neuropathy (CIPN) is a debilitating complication of cisplatin chemotherapy, often manifesting as mechanical allodynia, which frequently resists conventional treatments. This study explored the behavioral and molecular alterations induced by cisplatin, with a focus on regional dysfunction of the gamma-aminobutyric acid (GABA) system, and evaluated the therapeutic potential of intraplantar baclofen (a GABAB receptor agonist) in alleviating CIPN. Rats were administered cisplatin (2 mg/kg, i.p., once weekly for 4 weeks), and behavioral assessments revealed significa

Cistanche deserticola-derived exosome-like nanovesicles target the Microbiota-GABA signaling axis to ameliorate loperamide-induced constipation.

INTRODUCTION: Constipation is a prevalent gastrointestinal disorder with limited therapeutic options that often lead to laxative dependence. Gut microbiota and their metabolic regulation represent promising therapeutic targets. METHODS: In this study, we isolated and characterized exosome-like nanovesicles from the traditional Chinese herb Cistanche deserticola (CELNs) and evaluated their efficacy in a loperamide-induced mouse model of constipation. RESULTS: We found that CELNs administration significantly alleviated constipation phenotypes, as evidenced by increased fecal output, water cont

General, kappa, delta and mu opioid receptor antagonists mediate feeding elicited by the GABA-B agonist baclofen in the ventral tegmental area and nucleus accumbens shell in rats: reciprocal and regional interactions.

Food intake is significantly increased following administration of agonists of GABA and opioid receptors into the nucleus accumbens shell (NACs) and ventral tegmental area (VTA). GABA-A or GABA-B receptor antagonist pretreatment within the VTA or NACs differentially affects mu-opioid agonist-induced feeding elicited from the same site. Correspondingly, general or selective opioid receptor antagonist pretreatment within the VTA or NACs differentially affects GABA agonist-induced feeding elicited from the same site. Regional interactions have been evaluated in feeding studies by administering an

Synaptic activation of GABAA receptors induces neuronal uptake of Ca2+ in adult rat hippocampal slices.

Synaptically evoked transmembrane movements of Ca2+ in the adult CNS have almost exclusively been attributed to activation of glutamate receptor channels and the consequent triggering of voltage-gated calcium channels (VGCCs). Using microelectrodes for measuring free extracellular Ca2+ ([Ca2+]o) and extracellular space (ECS) volume, we show here for the first time that synaptic stimulation of gamma-aminobutyric acid-A (GABAA) receptors can result in a decrease in [Ca2+]o in adult rat hippocampal slices. High-frequency stimulation (100-200 Hz, 0.4-0.5 s) applied in stratum radiatum close (</=0.

Cognitive impairment and increased brain neurosteroids in adult rats perinatally exposed to low millimolar blood alcohol concentrations.

Epidemiological evidence suggests that adolescents and adults perinatally exposed to alcohol, even at low doses, show high prevalence of cognitive impairment and social behavior deficits, which may be in part related to alcohol-induced changes of the gamma-aminobutyric acid (GABA)ergic neurotransmission. The endogenous neurosteroid 3alpha-hydroxy,5alpha-pregnan-20-one (3alpha,5alpha-tetrahydroprogesterone/3alpha,5alpha-THP), a potent positive allosteric modulator of GABA(A) receptor function, is implicated in the physiological tuning of GABA-mediated fast inhibition and in various alcohol's ac

GABAergic modulation of a substance P-mediated reflex of slow time course in the isolated rat spinal cord.

The effects of gamma-aminobutyric acid (GABA) and other drugs which interact with GABA receptors were studied on a reflex of slow time course in the spinal cord preparation isolated from the neonatal rat. A single shock to a dorsal root (L3-L5) elicited a stereotyped series of reflexes, consisting of fast and slow components, recorded from the contralateral ventral root of the corresponding segment. The slow component, i.e. the contralateral slow ventral root potential (v.r.p.) had a time-to-peak of 2-5 s and lasted 20-30 s. Bath-application of GABA (5-20 microM) or muscimol (0.05-0.5 microM)

Neurochemical correlates of scene processing in the precuneus/posterior cingulate cortex: A multimodal fMRI and 1 H-MRS study.

Precuneus/posterior cingulate cortex (PCu/PCC) are key components of a midline network, activated during rest but also in tasks that involve construction of scene or situation models. Despite growing interest in PCu/PCC functional alterations in disease and disease risk, the underlying neurochemical modulators of PCu/PCC's task-evoked activity are largely unstudied. Here, a multimodal imaging approach was applied to investigate whether interindividual differences in PCu/PCC fMRI activity, elicited during perceptual discrimination of scene stimuli, were correlated with local brain metabolite le

Controlling Epileptic Activity by Modulating Putrescine Metabolism.

Glial mechanisms regulate neuronal excitability through multiple mechanisms, including the control of extracellular inhibitory signaling. One such mechanism is the Glu/GABA exchange process, in which glutamate uptake is coupled to GABA release, with GABA being synthesized from the polyamine putrescine, placing putrescine at a central position in metabolic pathways that may influence epileptiform activity. Here, we examined how pharmacological manipulation of key enzymes of putrescine metabolism affects seizure-like activity in the low-[Mg2+] in vitro model of frontotemporal epilepsy, complemen

Genomic and phenotypic analysis of probiotic properties of GABA-producing Enterococcus lactis A1 in Micropterus salmoides aquaculture.

Aquaculture contributes substantially to global food and nutrition security. Dysfunction of the fish intestinal mucosal barrier increases susceptibility to pathogens and affects aquaculture production. Indigenous probiotics, specifically those targeting intestinal health, are promising agents for disease control and growth promotion. Gamma-aminobutyric acid (GABA), derived from microorganisms, is a postbiotic with multiple beneficial functions. However, there is a lack of reports on GABA-producing probiotic strains in aquatic animals. Herein, we evaluated the probiotic properties of Enterococc

The Role of Ventral Tegmental Area Gamma-Aminobutyric Acid in Chronic Neuropathic Pain after Spinal Cord Injury in Rats.

Spinal cord injury (SCI) frequently results in chronic neuropathic pain (CNP). However, the understanding of brain neural circuits in CNP modulation is unclear. The present study examined the changes of ventral tegmental area (VTA) putative GABAergic and dopaminergic neuronal activity with CNP attenuation in rats. SCI was established by T10 clip compression injury (35 g, 1 min) in rats, and neuropathic pain behaviors, in vivo extracellular single-cell recording of putative VTA gamma-aminobutyric acid (GABA)/dopamine neurons, extracellular GABA level, glutamic acid decarboxylase (GAD), and vesi

Neurosteroids in hepatic encephalopathy: Novel insights and new therapeutic opportunities.

Hepatic encephalopathy (HE) is a serious neuropsychiatric disorder resulting from liver failure. Symptoms of HE include mild cognitive impairment, stupor and coma. Morphological changes to neuroglia (both astrocytes and microglia) occur in HE consisting of cytotoxic brain edema (astrocyte swelling) in acute liver failure and Alzheimer type-2 astrocytosis in cirrhosis. Visual-evoked responses in animals with liver failure and HE manifest striking similarities to those in animals treated with agonists of the GABA-A receptor complex. Neurosteroids are synthesized in brain following activation of

Effects of atorvastatin combined with trimetazidine on myocardial injury and inflammatory mediator in unstable angina patients during perioperative of percutaneous coronary intervention.

UNLABELLED: Two coordinated dynamic properties (adaptation and sensitization) are observed in retinal ganglion cells (RGCs) under the contrast stimulation. During sustained high-contrast period, adaptation decreases RGCs' responses while sensitization increases RGCs' responses. In mouse retina, adaptation and sensitization respectively show OFF- and ON-pathway-dominance. However, the mechanisms which drive the differentiation between adaptation and sensitization remain unclear. In the present study, multi-electrode recordings were conducted on isolated mouse retina under full-field contrast st

Overinhibition of corticostriatal activity following prenatal cocaine exposure.

OBJECTIVE: Prenatal cocaine exposure (PCE) can cause persistent neuropsychological and motor abnormalities in affected children, but the physiological consequences of PCE remain unclear. Conclusions drawn from clinical studies can sometimes be confounded by polysubstance abuse and nutritional deprivation. However, existing observations suggest that cocaine exposure in utero, as in adults, increases synaptic dopamine and promotes enduring dopamine-dependent plasticity at striatal synapses, altering behaviors and basal ganglia function. METHODS: We used a combination of behavioral measures, ele

Analgesia induced by 2- or 100-Hz electroacupuncture in the rat tail-flick test depends on the activation of different descending pain inhibitory mechanisms.

UNLABELLED: We evaluated the effectiveness of intrathecal antagonists of α1- (WB4101) and α2- (idazoxan) adrenoceptors and serotonergic (methysergide), opioid (naloxone), muscarinic (atropine), GABA(A) (bicuculline) and GABA(B) (phaclofen) receptors in blocking 2- or 100-Hz electroacupuncture (EA)-induced analgesia (EAIA) in the rat tail-flick test. EA was applied bilaterally to the Zusanli and Sanyinjiao acupoints in lightly anesthetized rats. EA increased tail-flick latency, where the effect of 2-Hz EA lasted longer than that produced by 100-Hz EA. The 2-Hz EAIA was inhibited by naloxone or

tDCS Modulates Visual Gamma Oscillations and Basal Alpha Activity in Occipital Cortices: Evidence from MEG.

Transcranial direct-current stimulation (tDCS) is now a widely used method for modulating the human brain, but the resulting physiological effects are not understood. Recent studies have combined magnetoencephalography (MEG) with simultaneous tDCS to evaluate online changes in occipital alpha and gamma oscillations, but no study to date has quantified the offline (i.e., after tDCS) alterations in these responses. Thirty-five healthy adults received active or sham anodal tDCS to the occipital cortices, and then completed a visual stimulation paradigm during MEG that is known to elicit robust ga

Connections between EM2- and SP-containing terminals and GABAergic neurons in the mouse spinal dorsal horn.

Endomorphin-2 (EM2) demonstrates a potent antinociceptive effect in pain modulation. To investigate the potential interactions of EM2- and substance P (SP)-containing primary afferents and γ-amino butyric acid (GABA)-containing interneurons in lamina II in nociceptive transmission, connections between EM2- and SP-containing terminals and GABAergic neurons in the spinal dorsal horn were studied. Double-immunofluorescent labeling showed that approximately 62.3 % of EM2-immunoreactive neurons exhibited SP-immunostaining, and 76.9 % of SP-immunoreactive neurons demonstrated EM2-immunoreactivities

Cocaine Dysregulates Dynorphin Modulation of Inhibitory Neurotransmission in the Ventral Pallidum in a Cell-Type-Specific Manner.

Cocaine-driven changes in the modulation of neurotransmission by neuromodulators are poorly understood. The ventral pallidum (VP) is a key structure in the reward system, in which GABA neurotransmission is regulated by opioid neuropeptides, including dynorphin. However, it is not known whether dynorphin acts differently on different cell types in the VP and whether its effects are altered by withdrawal from cocaine. Here, we trained wild-type, D1-Cre, A2A-Cre, or vGluT2-Cre:Ai9 male and female mice in a cocaine conditioned place preference protocol followed by 2 weeks of abstinence, and then r

The role of GABA, glutamate, and Glx levels in treatment of major depressive disorder: A systematic review and meta-analysis.

This systematic review and meta-analysis aims to explore a possible role of gamma-aminobutyric acid (GABA) and glutamate, as measured by Magnetic Resonance Spectroscopy (MRS), in the treatment outcomes of people with Major Depressive Disorder (MDD). Despite the prevalence of MDD and various treatment modalities, the neurobiological mechanisms of each remain poorly understood. We synthesised data from 41 longitudinal studies comprising 918 individuals with MDD, spanning four primary treatment modalities: selective serotonin reuptake inhibitors (SSRIs), ketamine, repetitive transcranial magnetic

Analysis of genes within the schizophrenia-linked 22q11.2 deletion identifies interaction of night owl/LZTR1 and NF1 in GABAergic sleep control.

The human 22q11.2 chromosomal deletion is one of the strongest identified genetic risk factors for schizophrenia. Although the deletion spans a number of known genes, the contribution of each of these to the 22q11.2 deletion syndrome (DS) is not known. To investigate the effect of individual genes within this interval on the pathophysiology associated with the deletion, we analyzed their role in sleep, a behavior affected in virtually all psychiatric disorders, including the 22q11.2 DS. We identified the gene LZTR1 (night owl, nowl) as a regulator of night-time sleep in Drosophila. In humans,
