Microsatellite-based assessment of genetic diversity and inbreeding in the Ukrainian water buffalo (Bubalus bubalis) population.

BACKGROUND AND AIM: The Ukrainian water buffalo (Bubalus bubalis) represents a small, geographically isolated population with limited genetic characterization. Such demographic constraints increase the risk of reduced genetic variability and inbreeding, potentially compromising long-term sustainability. This study aimed to evaluate the genetic diversity and allelic structure of the Ukrainian water buffalo population using standardized microsatellite markers recommended by the Food and Agriculture Organization-International Society for Animal Genetics (FAO-ISAG). MATERIALS AND METHODS: A total

Genetic Diversity and Population Structure of the Common Carp (Cyprinus carpio) in Albania Revealed by Microsatellite Markers.

Understanding genetic diversity and population structure is essential for the sustainable management of freshwater fish, particularly in systems influenced by long-term stocking. In this context, the genetic status of Albanian common carp (Cyprinus carpio) remains poorly understood. Here, 125 individuals from five populations representing natural lakes (Ohrid and Shkodra), semi-managed freshwater systems (Prespa and Belshi), and a hatchery stock (Tapiza) were genotyped at 12 polymorphic microsatellite loci. High genetic diversity was detected across all populations, with low genetic differenti

Genetic risk stratification of common diseases in breast cancer survivors: a population-based cohort study.

IMPORTANCE: Patients diagnosed with breast cancer (BCa) are at increased risk of multiple common diseases; however, the spectrum of these diseases and the contribution of inherited genetic susceptibility remain incompletely characterized. METHODS: We evaluated 15 common diseases and tested their associations with BCa exposure and disease-specific polygenic risk scores (PRS) in the UK Biobank (UKB; N = 254,736). Analyses were performed using cause-specific Cox proportional hazards models within a full-cohort framework, with time-updated BCa status, delayed entry at study recruitment, and age a

Genetic testing for hereditary breast and ovarian cancer in the Murcian population using a comprehensive NGS panel.

Traditionally, hereditary breast and ovarian cancer syndrome (HBOC) has been associated with germline pathogenic or likely pathogenic variants (PV/LPV) in BRCA1 and BRCA2. However, growing evidence indicates that this condition is genetically heterogeneous, and that PV/LPV in additional cancer predisposition genes also contribute significantly to disease susceptibility. In this study, 414 HBOC index cases (ICs) from the Region of Murcia, who fulfilled the 2019 Spanish Society of Medical Oncology (SEOM) criteria, were analyzed using next-generation sequencing (NGS). A 50-gene panel was applied,

Revealing genetic foundations underlying cardiometabolic traits: integrating twin and GWAS data in Asian populations.

BACKGROUND: Traditional twin studies have reported high heritability estimates for common cardiometabolic traits, yet discrepancies compared to genome-wide association studies (GWAS) derived estimates underscore the effects of "missing heritability", potentially attributed to genetic interactions and environmental influences. However, research exploring potential contributors to missing heritability in cardiometabolic phenotypes remains scarce and largely confined to European populations. METHODS: We analyzed twin data (n = 2548) from the Chinese National Twin Registry (CNTR) and GWAS summary

Phylogenetic and genetic population analyses of Amblyomma ovale (Parasitiformes: Ixodidae) from Brazil and Central America.

Amblyomma ovale is a widespread hard tick in Neotropics and it is an important vector of pathogens, particularly Rickettsia parkeri strain Atlantic rainforest, which causes a spotted fever illness in humans. Adult ticks primarily parasitize medium and large mammals, such as carnivores, while larvae and nymphs feed on small mammals and occasionally on birds. In Brazil, the species is found in all six major biomes (Amazon, Atlantic forest, Cerrado, Caatinga, Pantanal, Pampa), and is generally found in humid, forested areas. Despite its broad range, large-scale genetic studies have been lacking.

Biological Context-Informed and Population-Stratified Strategies Improve Genetic Diagnosis of CCDC22-Related Disorder.

BACKGROUND: The CCDC22 gene is a key regulator of endosomal trafficking and NF-κB signaling, and its dysfunction is implicated in a spectrum of X-linked neurodevelopmental disorders, including Ritscher-Schinzel syndrome 2, intellectual disability, and epilepsy. Despite the identification of CCDC22 variants having increased, a comprehensive biological characterization, including its spatiotemporal expression and functional molecular networks, remains to be systematically delineated. Furthermore, the rarity of CCDC22 variants created a significant challenge in distinguishing their pathogenicity.

Cascade screening and genetic variant pathogenicity assessment for familial hypercholesterolemia in a township‑based elderly population.

BACKGROUND: Familial hypercholesterolemia (FH) is a common, autosomal semi-dominant inherited disorder causing premature cardiovascular disease. However, it remains underdiagnosed, especially in community settings. Early diagnosis, particularly in children, is critical for timely intervention. Furthermore, the pathogenicity of genetic variants of uncertain significance poses a major challenge for clinical interpretation. METHODS: We conducted a systematic screening program in a town in Wujiang District, Suzhou, China. Eligible individuals (based on LDL-C ≥ 3.8 mmol/L, family history, or volun

Population genetic structure of zoonotic Toxoplasma gondii in China revealed using multilocus sequence typing.

BACKGROUND: The zoonotic pathogen Toxoplasma gondii exhibits a diverse global population structure, with a few dominant lineages primarily in the Northern Hemisphere. However, reliance on low-resolution, restriction-based genotyping methods has created a "resolution ceiling," potentially masking hidden genetic diversity and complex transmission dynamics. METHODS: In this study, we combined conventional PCR-restriction fragment length polymorphism (RFLP) screening with high-resolution Sanger sequencing targeting 16 genetic markers to unravel the fine-scale epidemiology of 96 T. gondii DNA samp

Genetic diversity and antifungal susceptibility among ecological niche populations of Aspergillus flavus in Yaoundé, Cameroon.

Aspergillus flavus is a ubiquitous fungus commonly found in a variety of ecological niches including soil, crops, the air, and humans. In humans, it is the second leading cause of invasive aspergillosis (IA) and is linked to other illnesses through contamination of agricultural products with aflatoxins. As such, A. flavus threatens food safety, economic wellbeing, and human health, particularly in less developed regions and countries such as Cameroon. To mitigate these effects, farmers and clinicians rely on triazoles to reduce aflatoxin contamination and treat IA. However, a consequence of in

Population genetic diversity of invasive Pomacea snails and surveillance of Angiostrongylus cantonensis in Shanghai, East China.

BACKGROUND: Golden apple snails (Gastropoda: Ampullariidae: Pomacea) were introduced into China in the 1980s for aquaculture and have since become widespread agricultural pests across East Asia. In addition to their invasive impact, they are a key intermediate host of the rat lungworm Angiostrongylus cantonensis (Secernentea: Angiostrongylidae) in China, the causative agent of eosinophilic meningitis in humans. METHODS: We conducted a malacological survey of 55 freshwater sites across Shanghai and neighboring East China provinces to assess Pomacea distribution, genetic diversity, and A. canto

Unraveling genetic diversity and population structure of pineapple germplasm using genome-wide SNP markers.

Pineapple (Ananas comosus (L.) Merr.) is one of the most important tropical fruits worldwide. It is primarily propagated clonally and exhibits high heterozygosity which can mask deleterious mutations, potentially leading to inbreeding depression and hindering breeding efforts. To address this, we conducted a comprehensive genome-wide analysis of 91 pineapple accessions and identified over 7.9 million high-quality SNPs. We utilized these SNPs to investigate the genetic structure and reproductive patterns of diverse cultivars and related varieties. Population structure analysis revealed a unique
