Comprehensive Bioactivity-Guided Isolation, DeepSAT, and LC-Q-TOF-MS/MS Analysis to Reveal Hypoglycemic Constituents of Momordica charantia L.

Activation of intestinal bitter taste receptors can stimulate the release of glucagon-like peptide-1 (GLP-1), thereby enhancing insulin secretion and exerting a hypoglycemic effect. In this study, a combination of bioactivity-guided screening and chemical analysis was applied to search for potential components from Momordica charantia L. that exert hypoglycemic effects via bitter taste receptors. Combined with molecular networking and DeepSAT analysis, the active fraction was found to be rich in cucurbitane-type triterpenoids, from which five pure compounds were isolated and identified. Potential Role of Epithelial Endoplasmic Reticulum Stress and Anterior Gradient Protein 2 Homologue in Crohn's Disease Fibrosis.

BACKGROUND AND AIMS: Intestinal fibrosis is a common complication of Crohn's disease [CD]. It is characterised by an accumulation of fibroblasts differentiating into myofibroblasts secreting excessive extracellular matrix. The potential role of the intestinal epithelium in this fibrotic process remains poorly defined. METHODS: We performed a pilot proteomic study comparing the proteome of surface epithelium, isolated by laser-capture microdissection, in normal and fibrotic zones of resected ileal CD strictures [13 zones collected in five patients]. Proteins of interests were validated by immu

Role of the incretin system in the remission of type 2 diabetes following bariatric surgery.

AIMS: It has been observed, as a collateral outcome of bariatric surgery, that morbidly obese patients with frank type 2 diabetes mellitus or impaired glucose tolerance undergone Roux-en-Y Gastric Bypass (RYGB) or bilio-pancreatic diversion (BPD) became and remained euglycemic since surgery. But, most interestingly, the conversion to euglycemia happened within few days from the operation, long before a significant weight loss could intervene. The purpose of this viewpoint is to try to elucidate the mechanisms involved in the resolution/remission of diabetes after bariatric surgery, in particul

Nocloprost, a unique prostaglandin E2 analog with local gastroprotective and ulcer-healing activity.

Nocloprost (9 beta-chloro-16,16-dimethyl prostaglandin E2 (PGE2)) was examined for gastroprotective and ulcer-healing activity and compared to 16,16-dimethyl PGE2 (dmPGE) in rats. Nocloprost given intragastrically (i.g.) at various doses (0.01-10 micrograms/kg) 30 min before 100% ethanol, acidified aspirin (ASA), acidified taurocholate, water immersion, or restraint stress dose dependently prevented the formation of gastric lesions, the ID50 values being 0.25, 0.58, 0.06 and 0.12 micrograms/kg, respectively. The gastroprotection provided by nocloprost given i.g. was somewhat enhanced by the pr

What's new in short bowel syndrome?

PURPOSE OF REVIEW: Short bowel syndrome (SBS) is a rare disease but with many complications due to intestinal failure, parenteral nutrition and underlying disease. A better prevention, comprehension and treatment could improve the outcome of these patients. RECENT FINDINGS: Recent studies have been published on acute intestinal failure, first cause of SBS, and gives us strategy to avoid extended intestinal resection and thus SBS. There has been progress in the comprehension of intestinal adaptation, characterized by improvements in intestinal absorption, changes on hormonal secretion, develop

In-depth study of gastrointestinal motility, intestinal barrier function and secretory function in an irritable bowel syndrome post-inflammatory rat model.

The presence of intestinal barrier dysfunction during 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced acute colitis in rats has been well described. However, it has not been studied extensively in the post-inflammatory model of irritable bowel syndrome (IBS) in pharmacological research, despite its frequent use. Therefore, we aimed to explore the intestinal barrier function in post-colitis rats in a comprehensive manner. To do so, colitis was induced in Sprague Dawley rats with a TNBS-enema, after which mucosal healing was followed via repeated colonoscopies. In the post-inflammatory phase,

Biochemical aspects of malabsorption in marasmus.

Sixty marasmic children and fifteen normal age-matched controls were investigated for the absorption of fats and proteins. Their duodenal juice samples were also analysed for bile salts and microflora. A significant amount of malabsorption, with respect to both the dietary ingredients, was observed in the majority of the marasmic children. The levels of conjugated bile acids in the duodenal juice samples of marasmic children were significantly lower as compared with those of normal controls, while the reverse was true for free bile acids and bacterial counts. Significant correlations were obse

A Guide to Human Zinc Absorption: General Overview and Recent Advances of In Vitro Intestinal Models.

Zinc absorption in the small intestine is one of the main mechanisms regulating the systemic homeostasis of this essential trace element. This review summarizes the key aspects of human zinc homeostasis and distribution. In particular, current knowledge on human intestinal zinc absorption and the influence of diet-derived factors on bioaccessibility and bioavailability as well as intrinsic luminal and basolateral factors with an impact on zinc uptake are discussed. Their investigation is increasingly performed using in vitro cellular intestinal models, which are continually being refined and k

Chronic ingestion of a potential food contaminant induces gastrointestinal inflammation in rats: role of nitric oxide and mast cells.

Chronic ingestion of xenobiotics could be pathogenic in the gastrointestinal tract. Recently, we showed that acute low administration of a food contaminant (diquat) induced intestinal secretion involving mast cells and nitric oxide. This work aimed to determine in rats: (1) the influence of a low level (0.1 mg/kg/day per os) chronic ingestion of diquat on gastrointestinal immune cells, and (2) the participation of nitric oxide synthases (NOS) in these effects. Diquat increased both gastric and jejunal myeloperoxidase activities, tissue histamine in vitro release after stimulation by 48/80, and

The role of neuroenteric hormones in intestinal infectious diseases.

It is now well established that communication among the enteric nerves, hormones, and neuropeptides plays a role in the pathogenesis of infectious gastrointestinal conditions. The results of several studies suggest that enteric nerves and hormones modulate important gastrointestinal functions such as intestinal motility and transport, intestinal permeability, fluid secretion, and inflammation in response to infectious agents. During the past year several gut-brain peptides, including substance P, neurotensin, and galanin, emerged as important mediators in the development and progress of intest

[Ileo-ileal intussusception over an islet of heterotopic gastric mucosa without Meckel's diverticulum].

Three laparotomies have been necessary to find the origin of intermittent episodes of abdominal pain for eleven years in a girl: appendectomy at the age of three, surgical exploration at nine, and emergency laparotomy at fourteen for acute intestinal obstruction: ileo-ileal, secondary iléo-colic intussusception, reduction and ileal resection discovering an 2 x 2 cm area of normal fundic gastric mucosa. Heterotopic gastric mucosa in the small bowel is a rare congenital anomaly (12 pediatric and 5 adult cases previously reported in the literature). These folds of gastric mucosa are located more

Hormonal control of gastrointestinal motility.

The motor responses of the gastrointestinal tract to a meal are tuned to provide the optimal conditions for digestion and absorption of food. Hormones released from the mucosa of the intestinal tract play an important role in regulating and integrating the various motor events which occur. Complex feedbacks, both nervous and humoral, control the release of intestinal hormones and also modify their actions. Preliminary studies suggest that abnormalities in the secretion or action of various gastrointestinal hormones may play an important role in the production of motor disorders of the gastroin

REGγ Mitigates Radiation-Induced Enteritis by Preserving Mucin Secretion and Sustaining Microbiome Homeostasis.

Radiation-induced enteritis, a significant concern in abdominal radiation therapy, is associated closely with gut microbiota dysbiosis. The mucus layer plays a pivotal role in preventing the translocation of commensal and pathogenic microbes. Although significant expression of REGγ in intestinal epithelial cells is well established, its role in modulating the mucus layer and gut microbiota remains unknown. The current study revealed notable changes in gut microorganisms and metabolites in irradiated mice lacking REGγ, as compared to wild-type mice. Concomitant with gut microbiota dysbiosis, RE

Effects of Ganoderic Acid A on Gastrointestinal Motility and Brain-Gut Peptide in Rats with Functional Dyspepsia.

Источник пометил публикацию как отозванную

OBJECTIVE: The therapeutic effect of drugs for functional dyspepsia (FD) is still limited. Ganoderic acid A (GAA) has anti-inflammatory and cellular protective activities. The aim of this study is to explore the therapeutic effect of GAA on FD. METHODS: The FD rat model was established via tail damping and forced exercise fatigue. The gastric emptying rate and intestinal propulsion rate of the rats in each group were then detected, and the pathological damage of gastric antrum and duodenum tissues was observed by hematoxylin-eosin (HE) staining. An enzyme-linked immunosorbent assay (ELISA) wa

Intestinal SGLT1 in metabolic health and disease.

The Na(+)-glucose cotransporter 1 (SGLT1/SLC5A1) is predominantly expressed in the small intestine. It transports glucose and galactose across the apical membrane in a process driven by a Na(+) gradient created by Na(+)-K(+)-ATPase. SGLT2 is the major form found in the kidney, and SGLT2-selective inhibitors are a new class of treatment for type 2 diabetes mellitus (T2DM). Recent data from patients treated with dual SGLT1/2 inhibitors or SGLT2-selective drugs such as canagliflozin (SGLT1 IC50 = 663 nM) warrant evaluation of SGLT1 inhibition for T2DM. SGLT1 activity is highly dynamic, with modul

Physiological and anti-inflammatory roles of dietary fiber and butyrate in intestinal functions.

BACKGROUND: We investigated the effects of pectin, a soluble dietary fiber, on the morphological parameters of the small intestine. In addition, we tested the effects of butyrate enemas on dextran sulfate sodium (DSS)-induced experimental colitis. METHODS: Male Wistar rats were fed an elemental diet containing 2.5% pectin for 14 days, and several parameters were then determined. DSS-induced colitis was evoked by the oral administration of water containing 3% DSS for 10 days. The butyrate enema (3 mL of 100 mmol/L butyrate per day) was begun 7 days before the DSS treatment. Interleukin (IL)-8

Increased survival of CFTR knockout mice with an oral osmotic laxative.

Mouse models of cystic fibrosis that are generated by targeted disruption (knockout) of the cystic fibrosis transmembrane conductance regulator gene, cftr(-/-), typically die shortly after weaning, from intestinal obstruction/rupture caused by an inability to secrete fluid into the bowel lumen. We investigated the use of a commercial osmotic laxative, Colyte, provided continuously in the drinking water, to increase the survival of cftr(-/-) mice. Genotype analysis of 623 offspring surviving at 10 days of age yielded 28.1% cftr(+/+), 59.6% cftr(+/-), and 12.4% cftr(-/-) mice (25% predicted), su
