Membrane blebs have important roles in cell migration, apoptosis, and intercellular communication through extracellular vesicles (EVs). While plasma membranes (PM) typically maintain phosphatidylserine (PS) on their cytoplasmic leaflet, most blebs have PS exposed on their outer leaflet, revealing that loss of steady-state lipid asymmetry often accompanies PM blebbing. How these changes in PM lipid organization regulate membrane properties and affect bleb formation remains unknown. We confirmed that lipid scrambling through the scramblase TMEM16F is essential for chemically induced membrane ble

A molecular dithiane-based approach to synthesis of novel photolabile phospholipids is developed. These lipids are used in formulations with egg-POPC and cholesterol to prepare light-sensitive liposomes. Irradiation of such liposomes in PBS buffer (medium pressure mercury lamp, Pyrex filter, lambda > 300 nm) significantly increases the bilayer permeability and accelerates the release of entrapped small organic molecules by an order of magnitude. A simple assay, based on (1)H or (19)F PFG NMR measurements of diffusion coefficients, is developed to monitor light-induced unloading of the probe mo

Severe hemolysis or myolysis occurring during pathological states, such as sickle cell disease, ischemia reperfusion, and malaria results in high levels of free heme, causing undesirable toxicity leading to organ, tissue, and cellular injury. Free heme catalyzes the oxidation, covalent cross-linking and aggregate formation of protein and its degradation to small peptides. It also catalyzes the formation of cytotoxic lipid peroxide via lipid peroxidation and damages DNA through oxidative stress. Heme being a lipophilic molecule intercalates in the membrane and impairs lipid bilayers and organel…

Determining the mechanism of action of antimicrobial peptides (AMPs) is critical if they are to be developed into the clinical setting. In recent years high resolution techniques such as atomic force microscopy (AFM) have increasingly been utilised to determine AMP mechanism of action on planar lipid bilayers and live bacteria. Here we present the biophysical characterisation of a prototypical AMP from the venom of the North African scorpion Scorpio maurus palmatus termed Smp24. Smp24 is an amphipathic helical peptide containing 24 residues with a charge of +3 and exhibits both antimicrobial a…

Lipid membranes play crucial roles in cellular functions and offer diverse engineering applications. Studying their properties is critical but computationally demanding through atomistic simulations. In this work, we develop coarse-grained (CG) models for phosphocholine lipids, aimed at balancing computational efficiency and predictive accuracy with chemical and temperature transferability. We introduce chargeless CG beads with 2:1 or 3:1 mapping and optimize the force fields through a combination of systematic and accelerated approaches that integrate particle swarm optimization algorithm wit

Oxidative stress generates oxidized phospholipids (OxPLs) that alter membrane structure and inflammatory lipid signaling, yet the underlying biophysical mechanisms remain poorly understood. Here, we examine how two structurally distinct truncated oxidized phosphatidylcholines (OxPCs), 1-palmitoyl-2-(5'-oxo-valeroyl)-sn-glycero-3-phosphocholine (POVPC) and 1-palmitoyl-2-glutaryl-sn-glycero-3-phosphocholine (PGPC), remodel membrane lateral organization and regulate secretory phospholipase A2 (sPLA2) activity. Nitrous oxide (N2O) is a widely used inhalational analgesic whose antinociceptive effects depend critically on the κ-opioid receptor (KOR) and its endogenous ligand dynorphin (Dyn). Although N2O is known to promote Dyn release, whether it can also modulate KOR structure and signaling more directly remains unclear. Here, using all-atom molecular dynamics simulations together with free-energy calculations, we identify a membrane-mediated mechanism by which N2O allosterically biases KOR toward its active state. N2O rapidly permeates POPC/cholesterol membranes and preferentially accumulates within

Laurdan (6-dodecanoyl-2-(dimethylamino)-naphthalene) is a solvatochromic fluorescent probe widely used for investigating membrane biophysical properties. Since its first application in phospholipid bilayers, laurdan has become a valuable tool in this field, owing to its sensitivity to the mobility and dynamics of surrounding lipid carbonyl groups and its ability to report on membrane phase behavior through the generalized polarization (GP) parameter. GP, a ratiometric empirical parameter, is derived from the ratio of emission intensities at approximately 440 and 490 nm, providing a measure of …

Antimicrobial peptides are vital to the innate immune system and serve as a safe alternative to traditional antibiotics for bacterial infections. However, their effectiveness is limited by the need for high concentrations to disrupt bacterial membranes and the challenge of protecting them from proteases that degrade them before reaching their target. Intending to improve the delivery of anoplin (ANP) to bacterial cells, we prepared ANP-coated gold nanoparticles (Au NPs) with a diameter of about 50 nm, and investigated their effects on the properties of model bacterial membranes using Raman and

Understanding the cell membrane penetration process of biomedical nanosystems and its dependence on nanomaterial properties and surface functionalization is crucial for the rational design of safe and efficient cellular internalization strategies. Computer simulations are powerful tools to evaluate the thermodynamic aspects of the process and to elucidate its underlying molecular mechanisms. In this work, the interaction between uncoated or polymer-coated graphene oxide (GO) dots and lipid bilayer models is investigated by coarse-grained (CG) molecular dynamics (MD) simulations. We first valid

NMR and molecular dynamics simulations revealed differences in the localization of the novel thiosemicarbazones: 2-benzoyl ((E)-N-(acridin-9-yl)-2-(phenyl(pyridin-2-yl)methylene)hydrazine-1-carbothioamide (AOBP) and 2-dipyridyl ((E)-N-(acridin-9-yl)-2-(di(pyridin-2-yl)methylene)hydrazine-1-carbothioamide (AODP) within the lipid membrane. It turned out that both thiosemicarbazones can penetrate inside the membrane, but AOBP is able to pass into the center of the hydrophobic region of the lipid bilayer, while AODP is distributed closer to the surface and freely leaves the membrane into the aqueo

Dynamic Nuclear Polarization (DNP) has emerged as a transformative technique for enhancing the sensitivity of Nuclear Magnetic Resonance (NMR) spectroscopy. In the quest to improve the polarization of 31P nuclei in liquid environments, previous efforts have yielded only limited success. This study introduces a novel approach utilizing the solid effect to effectively polarize pentavalent phosphorus within the phosphate groups of key biomolecules, including adenosine triphosphate, nucleic acid, and lipid bilayer in the fluid phase. We report a remarkable 31P DNP enhancement factor exceeding 20 f

Amyloid-β (Aβ1-42) peptides aggregated into plaques deposited in the brain are the main hallmark of Alzheimer's disease (AD), a social and economic burden worldwide. In this context, insoluble Aβ1-42 fibrils are the main components of plaques. The recent trials that used approved AD drugs show that they can remove the fibrils from AD patients' brains, but they did not halt the course of the disease. Mounting evidence envisages that the soluble Aβ1-42 oligomers' interactions with the neuronal membrane trigger higher cell death than Aβ1-42 fibril interactions. Developing a compound that can alle

BACKGROUND: The insufficient understanding of unintended biological impacts from nanomaterials (NMs) represents a serious impediment to their use for scientific, technological, and medical applications. While previous studies have focused on understanding nanotoxicity effects mostly resulting from cellular internalization, recent work indicates that NMs may interfere with transmembrane transport mechanisms, hence enabling contributions to nanotoxicity by affecting key biological activities dependent on transmembrane transport. In this line of inquiry, we investigated the effects of charged nan

The lateral mobility of proteins within cell membranes is usually thought to be dependent on their size and modulated by local heterogeneities of the membrane. Experiments using single-particle tracking on reconstituted membranes demonstrate that protein diffusion is significantly influenced by the interplay of membrane curvature, membrane tension, and protein shape. We find that the curvature-coupled voltage-gated potassium channel (KvAP) undergoes a significant increase in protein mobility under tension, whereas the mobility of the curvature-neutral water channel aquaporin 0 (AQP0) is insens
