Household-level risk factors for Aedes aegypti pupal density in Guayaquil, Ecuador.

BACKGROUND: Vector-borne diseases are a major cause of disease burden in Guayaquil, Ecuador, especially arboviruses spread by Aedes aegypti mosquitoes. Understanding which household characteristics and risk factors lead to higher Ae. aegypti densities and consequent disease risk can help inform and optimize vector control programs. METHODS: Cross-sectional entomological surveys were conducted in Guayaquil between 2013 and 2016, covering household demographics, municipal services, potential breeding containers, presence of Ae. aegypti larvae and pupae, and history of using mosquito control met…

Preparation of β-Myrcene-Chitosan Nanoparticles and Their Uptake and Toxicity in Aedes aegypti Larvae.

Mosquito control still relies heavily on synthetic molecules, which can lead to the selection of resistant populations and undesirable environmental problems. This study described the preparation of a nanoparticle of the plant-derived molecule, β-myrcene, with chitosan, and the assessment of its toxicity against larvae of the yellow fever mosquito, Aedes aegypti. By producing fluorescent chitosan nanoparticles, we were able to observe their distribution in the digestive tract of larvae of Ae. aegypti. Chitosan-based nanoparticles containing β-myrcene (238 mg/L) could kill 100% of the larvae te…

Mosquito-borne viral diseases and potential transmission blocking vaccine candidates.

Mosquito-borne viral diseases (MBVDs) have a complex biological cycle involving vectors and vertebrate hosts. These viruses are responsible for many deadly diseases worldwide. Although MBVDs threaten mostly developing countries, there is growing evidence indicating that they are also of concern in western countries where local transmission of arboviruses such as West Nile, Zika, Chikungunya and Dengue viruses have been recently reported. The rapid rise in human infections caused by these viruses is attributed to rapid climate change and travel facilities. Usually, the only way to control these

A Review: Aedes-Borne Arboviral Infections, Controls and Wolbachia-Based Strategies.

Arthropod-borne viruses (Arboviruses) continue to generate significant health and economic burdens for people living in endemic regions. Of these viruses, some of the most important (e.g., dengue, Zika, chikungunya, and yellow fever virus), are transmitted mainly by Aedes mosquitoes. Over the years, viral infection control has targeted vector population reduction and inhibition of arboviral replication and transmission. This control includes the vector control methods which are classified into chemical, environmental, and biological methods. Some of these control methods may be largely experim

Practices and insights from chikungunya prevention and Control in Guangdong, China, 2025.

Chikungunya, a mosquito-borne disease, has become a global public health concern. In 2025, an imported chikungunya case was detected in Foshan, Guangdong, followed by local spread. This study analyzed the spatiotemporal evolution of this outbreak using data from official channels. Measures like enhanced mosquito control, expanded nucleic acid testing, and activation of emergency response were implemented. As a result, the epidemic was effectively controlled, with no severe cases reported. This work provided valuable insights into chikungunya prevention and control strategies, highlighting the …

Potential for Co-Infection of a Mosquito-Specific Flavivirus, Nhumirim Virus, to Block West Nile Virus Transmission in Mosquitoes.

Nhumirim virus (NHUV) is an insect-specific virus that phylogenetically affiliates with dual-host mosquito-borne flaviviruses. Previous in vitro co-infection experiments demonstrated prior or concurrent infection of Aedes albopictus C6/36 mosquito cells with NHUV resulted in a 10,000-fold reduction in viral production of West Nile virus (WNV). This interference between WNV and NHUV was observed herein in an additional Ae. albopictus mosquito cell line, C7-10. A WNV 2K peptide (V9M) mutant capable of superinfection with a pre-established WNV infection demonstrated a comparable level of interfer

Key Factors Influencing the Incidence of West Nile Virus in Burleigh County, North Dakota.

The city of Bismarck, North Dakota has one of the highest numbers of West Nile Virus (WNV) cases per population in the U.S. Although the city conducts extensive mosquito surveillance, the mosquito abundance alone may not fully explain the occurrence of WNV. Here, we developed models to predict mosquito abundance and the number of WNV cases, independently, by statistically analyzing the most important climate and virus transmission factors. An analysis with the mosquito model indicated that the mosquito numbers increase during a warm and humid summer or after a severely cold winter. In addition

Mathematical model of Zika virus with vertical transmission.

Zika is a flavivirus transmitted to humans through either the bites of infected Aedes mosquitoes or sexual transmission. Zika has been linked to congenital anomalies such as microcephaly. In this paper, we analyze a new system of ordinary differential equations which incorporates human vertical transmission of Zika virus, the birth of babies with microcephaly and asymptomatically infected individuals. The Zika model is locally and globally asymptotically stable when the basic reproduction number is less than unity. Our model shows that asymptomatic individuals amplify the disease burden in the

Host-feeding patterns of mosquito species in Germany.

BACKGROUND: Mosquito-borne pathogens are of growing importance in many countries of Europe including Germany. At the same time, the transmission cycles of most mosquito-borne pathogens (e.g. viruses or filarial parasites) are not completely understood. There is especially a lack of knowledge about the vector capacity of the different mosquito species, which is strongly influenced by their host-feeding patterns. While this kind of information is important to identify the relevant vector species, e.g. to direct efficient control measures, studies about the host-feeding patterns of mosquito speci

Germline transformation of the West Nile virus and avian malaria vector Culex quinquefasciatus Say using the piggyBac transposon system.

Culex quinquefasciatus Say is a mosquito which acts as a vector for numerous diseases including West Nile virus, lymphatic filariasis and avian malaria, over a broad geographical range. As the effectiveness of insecticidal mosquito control methods declines, the need has grown to develop genetic control methods to curb the spread of disease. The piggyBac transposon system - the most widely used genetic transformation tool in insects, including mosquitoes - generates quasi-random insertions of donor DNA into the host genome. However, despite the broad reported species range of piggyBac, previous…

Colonization of Anopheles coustani, a neglected malaria vector in Madagascar.

Anopheles coustani has long been recognized as a secondary malaria vector in Africa. It has recently been involved in the transmission of both Plasmodium falciparum and P. vivax in Madagascar. As most secondary malaria vectors, An. coustani mainly bites outdoors, which renders the control of this mosquito species difficult using classical malaria control measures, such as the use of bed nets or indoor residual spraying of insecticides. For a better understanding of the biology and vector competence of a vector species, it is useful to rear the species in the laboratory. The absence of a colony

Linking mosquito infestation to resident socioeconomic status, knowledge, and source reduction practices in suburban Washington, DC.

Eliminating water-holding containers where mosquitoes oviposit and develop (source reduction) can help manage urban disease-vector mosquitoes. Source reduction requires residents to be knowledgeable of effective practices and motivated to implement them. We tested relationships between demographics, resident knowledge, attitudes, and practices (KAP), and mosquito infestation by administering larval mosquito surveys and KAP questionnaires in Washington, DC. Respondents who reported practicing source reduction had lower numbers of pupae-positive containers and Culex pipiens-positive containers,

A knockout screen of genes expressed specifically in Ae. aegypti pupae reveals a critical role for stretchin in mosquito flight.

Aedes aegypti is a critical vector for transmitting Zika, dengue, chikungunya, and yellow fever viruses to humans. Genetic strategies to limit mosquito survival based upon sex distortion or disruption of development may be valuable new tools to control Ae. aegypti populations. We identified six genes with expression limited to pupal development; osi8 and osi11 (Osiris protein family), CPRs and CPF (cuticle protein family), and stretchin (a muscle protein). Heritable CRISPR/Cas9-mediated gene knockout of these genes did not reveal any defects in pupal development. However, stretchin-null mutati

Chronology of sodium channel mutations associated with pyrethroid resistance in Aedes aegypti.

Aedes aegypti is the primary mosquito vector of dengue, yellow fever, Zika and chikungunya. Current strategies to control Ae. aegypti rely heavily on insecticide interventions. Pyrethroids are a major class of insecticides used for mosquito control because of their fast acting, highly insecticidal activities and low mammalian toxicity. However, Ae. aegypti populations around the world have begun to develop resistance to pyrethroids. So far, more than a dozen mutations in the sodium channel gene have been reported to be associated with pyrethroid resistance in Ae. aegypti. Co-occurrence of resi…

Comprehensive genomic analysis and functional characterization of a new entomopathogenic strain, Bacillus thuringiensis subsp. israelensis VCRC B646, antagonistic to mosquito vectors (Diptera: Culicidae), isolated from soil in Tamil Nadu, India.

BACKGROUND: Mosquito-borne diseases such as malaria, dengue and filariasis remain major public health threats, demanding effective and environmentally-safe control measures. Bacillus thuringiensis subsp. israelensis (Bti) is widely employed as a biopesticide owing to its Cry and Cyt δ-endotoxins. Continuous isolation and genomic characterization of potent indigenous strains are crucial for strengthening vector control strategies. This study aimed to assess the larvicidal efficacy, genomic features, plasmid architecture and environmental safety of a novel Indian isolate, Bti VCRC B646. RESULTS

The AaFoxA factor regulates female reproduction through chromatin remodeling in the mosquito vector Aedes aegypti.

Female mosquitoes are vectors of many devastating human diseases because they require blood feeding to initiate reproduction. Thus, elucidation of molecular mechanisms managing female mosquito reproduction is essential. Although the regulation of gene expression during the mosquito gonadotrophic cycle has been studied in detail, how this process is controlled at the chromatin level remains unclear. Chromatin must be accessible for transcription factors (TFs) governing gene expression. A specialized class of TFs, called pioneer factors (PFs), binds and remodels closed chromatin, permitting othe

Targeting Iron Homeostasis Curtails Mosquito Fertility and Plasmodium Transmission Potential.

Iron metabolism is essential for both mosquito reproductive fitness and Plasmodium development within the mosquito, yet this axis has remained insufficiently explored as a potential target for malaria control. Ferritin, the major iron-storage protein and a central regulator of iron homeostasis, has not been biochemically or functionally characterized in any hematophagous insect to date. Our computational and phylogenetic analyses reveal that mosquito ferritin represents a unique case, exhibiting a chimeric architecture that combines conserved iron-binding motifs with distinctive heme-coordinat
