D-ribose--an additive with caffeine.

Caffeine acts as a stimulant, in which approximately 90% of people in the United States consume daily. Besides its beneficial effects, many individuals have experienced unpleasant reactions following the consumption of caffeine: such as insomnia, an increase in heart rate, feelings of nervousness, headaches, occasional lightheadedness, a state of "jitters," and a potential "crash" state following its metabolism. Researchers have proposed mechanisms responsible for caffeine's interactions, which include its blocking capacity of adenosine receptors, its role with the pituitary gland, increasing

Pyruvate protects glucose-deprived Müller cells from nitric oxide-induced oxidative stress by radical scavenging.

The cellular defense of Müller cells against oxidative and nitrosative stress was examined after the addition of the nitric oxide donor papanonoate. Glucose concentrations of > or = 550 microM efficiently protected the Müller cells from cell death by maintaining high ATP and glutathione and allowing only a moderate increase of free radicals. Fluorescence-activated cell sorting (FACS) analysis showed that 22% of the cells underwent apoptosis whereas necrosis was strongly suppressed. Under glucose deprivation, the intracellular concentration of ATP declined to 15% after 1 h; glutathione dropped

Glucose-6-phosphate: a key compound in glycogenosis I and favism leading to hyper- or hypolipidaemia.

The glycogen storage disorders (GSD)-I, -III, -VI and -VIII are associated with hypertriglyceridaemia or mixed hyperlipidaemia which poses the question whether these patients have an increased risk for atherosclerosis. The atherogenicity of triglycerides has remained controversial, while increased plasma cholesterol levels are generally accepted as a significant risk factor for coronary heart disease. However, clinical data show that one has to differentiate between metabolic conditions where triglycerides are atherogenic and those which are not significantly related to early onset of atherosc

Sensitivity of central chemoreceptors controlling blood glucose and body temperature during glucose deprivation.

1. The rise in blood glucose and the fall in body temperature which follows the injection of a glucose analogue, 2-deoxy-D-glucose (2-DG) into the lateral cerebral ventricle (I.C.V) of unanaesthetized rats were studied and found to be dose-dependent. These 2-DG induced responses are elicited by the impairment of glucose metabolism within central "glucoreceptors'. 2. 2DG induced hyperglycaemia and hypothermia were completely prevented and even the converse effects occurred when fivefold equimolar amounts of D-fructose were simultaneously injected I.C.V.; fructose, at equimolar doses, did not mo

Comparative Metabolomics of Sesame Cultivars in Response to Seed Deterioration Stress Treated With Cis-Cinnamic Acid.

Seed deterioration is considered an abiotic stress that has significant destructive effects on seed germination and seedling establishment in oilseed crops. In this study, the responses to seed deterioration in two sesame cultivars were evaluated based on seed and seedling vigor traits, along with the alteration in the metabolic profile. The investigations were conducted using a factorial experiment, including stress (deterioration and non-deterioration of seed) and cis-cinnamic acid pretreatment (0, 30, and 50 μM). Results showed that CA improved germination (25.5% to 50.8%) and seedling vigo

Integrated multi-omics reveals lipid remodeling underlying population- and stage-associated aroma variation in chicken egg yolks.

Egg yolk aroma is a key determinant of consumer acceptance, yet the metabolic basis of aroma variation across chicken breeds and laying stages remains unclear. Here, integrated volatilomics, lipidomics, and metabolomics were employed to investigate egg yolk aroma variation in four genetically distinct chicken populations at the onset-of-lay and peak-laying stages. A total of 265 volatile compounds were tentatively annotated. Literature odor thresholds were available for 93 compounds, allowing their OAVs to be calculated; among these, aldehydes, alcohols, ketones, and terpenoids were the predom

PslG-Mediated Disruption of Pseudomonas aeruginosa Biofilms: A Multi-Omics Investigation of Matrix Glycan Release, Metabolic Reprogramming, and Structural Disintegration.

Pseudomonas aeruginosa biofilms are recalcitrant to conventional antibiotic therapy due to their extracellular polymeric substance (EPS) matrix, in which Psl polysaccharide serves as the primary structural scaffold. PslG glycoside hydrolase can degrade Psl and trigger biofilm dispersal, yet the dynamic glycan release profile and metabolic consequences remain poorly characterized. Mature P. aeruginosa PAO1 biofilms were treated with PslG (1-1000 nM) for 0-30 min. Biomass was quantified by crystal violet staining. Matrix glycan composition and untargeted metabolomics was performed using LC-MS/MS

Molecular mechanisms and targeted therapeutic strategies for glycolytic reprogramming in osteosarcoma.

Osteosarcoma, a highly invasive and metastatic malignant bone tumor, has a poor prognosis primarily because of early pulmonary metastasis and resistance to conventional treatments. The malignant progression of osteosarcoma cells significantly depends on glycometabolic reprogramming, a process characterized by enhanced aerobic glycolysis. This review begins by mapping the multi-dimensional networks driving sugar metabolism reprogramming, which revealed that functional abnormalities in core glycolytic enzymes, including hexokinase 2, pyruvate kinase M2, and lactate dehydrogenase A (LDHA), are dr

Perturbation of Metabolic Pathways Associated with Exposure of Human Renal Proximal Tubular Cells to Nephrotoxic Pollutants and Drugs.

Mammalian kidneys, although exposed to a diverse array of environmental pollutants and commonly used drugs that are dose-limited by nephrotoxicity, have a significant but limited capacity for repair and regeneration. Primary cultures of human proximal tubular (hPT) cells were used to investigate the cellular response to exposures to one of two environmental toxicants (mercuric chloride [HgCl2] or S-(1,2-dichlorovinyl)-L-cysteine [DCVC; key metabolite of trichloroethylene] or one of three nephrotoxic drugs (cisplatin [CDDP], polymyxin B [PmxB], tenofovir disoproxil fumarate [TDF]) at concentrat

Hydrophilic-hydrophobic imbalance is associated with alcohol-positive milk in Holstein cows: An untargeted metabolomics analysis.

Alcohol-positive milk (APM) is a prevalent milk quality defect causing economic losses in dairy production. Using a longitudinal case-control design, this study characterized the metabolomic profile of APM and clarified the core molecular signatures underlying its occurrence in Holstein cows. Twenty milk samples from Holstein cows (10 normal milk [NM] vs. 10 APM) were collected for case-control comparison, and 7 paired samples from spontaneously recovered APM cows (recovered alcohol-negative milk, RAPM) were obtained for longitudinal analysis. Untargeted metabolomic profiling was performed usi

TMEM132A is associated with metabolic reprogramming, macrophage-oriented immune remodeling, and breast cancer progression.

Breast cancer progression reflects malignant cell programs coupled with immune remodeling. Biomarkers that connect tumor intrinsic states with microenvironmental interactions remain needed. We evaluated TMEM132A as a candidate marker in breast cancer. Bulk cohorts from The Cancer Genome Atlas, Genotype Tissue Expression, and Gene Expression Omnibus were analyzed to quantify TMEM132A expression, diagnostic discrimination, and associations with clinicopathologic features and survival. Single cell RNA sequencing and spatial transcriptomics localized TMEM132A across cell populations and tissue reg

Integrated analysis of enzymes, mRNAs, and miRNAs provides insight into the regulatory potential of extracellular vesicles in recipient cell glucose metabolism.

INTRODUCTION: Extracellular vesicles (EVs) are increasingly recognized as active coordinators of metabolic processes rather than mere messengers. By carrying unique subsets of enzymes, metabolites, lipids, and nucleic acids, EVs can directly deliver functional metabolic machinery or dynamically alter intracellular metabolic fluxes in recipient cells. However, their role in regulating specific biochemical pathways remains largely unknown. METHODOLOGY: In the current in silico analysis, we explored the dominant metabolic role of EV cargo using publicly available multi-omics data. For this, the

Cereblon (CRBN) inhibits prostate cancer metastasis by negatively regulating 6-phosphogluconate dehydrogenase (6PGD).

Metastasis is the primary cause of mortality in advanced prostate cancer, and the emergence of resistance to androgen receptor (AR)-targeted therapies highlights the urgent need for alternative therapeutic strategies. Metabolic reprogramming has increasingly been recognized as a key driver of metastatic progression. In this study, we uncover a novel tumor-suppressive role for cereblon (CRBN), a substrate receptor of the CRL4CRBN E3 ubiquitin ligase complex, in modulating prostate cancer metastasis through regulation of 6-phosphogluconate dehydrogenase (6PGD), a critical enzyme in the oxidative

Carbonic anhydrase activity under low zinc in Phaeodactylum tricornutum largely depends on metal-free iota-carbonic anhydrase.

Carbonic anhydrases (CAs) typically possess a Zn2+ cofactor and are involved in a wide range of adaptive metabolisms involving pH and dissolved inorganic carbon homeostasis. In photosynthetic microorganisms, they are involved in CO2 concentrating mechanisms (CCM) that supply CO2 to ribulose-1,5-bisphosphate carboxylase-oxygenase. Here we show that the model diatom Phaeodactylum tricornutum possesses a CA from the newly identified iota class, P. tricornutum ι-carbonic anhydrase (Pt-ιCA), which exhibited catalytic activity without a metal cofactor, in contrast to most CAs. Metal-free Pt-ιCA acti

Metabolic pathways fueling human spermatozoa capacitation and hyperactivation: a roadmap for spermatozoa bioenergetics.

Human spermatozoa are highly specialized cells whose function depends on tightly regulated metabolic and signaling networks. Following spermatogenesis and epididymal maturation, ejaculated spermatozoa must undergo capacitation in the female reproductive tract, a complex process characterized by extensive biochemical remodeling and high energy demand that enables hyperactivated motility and fertilizing competence. Human spermatozoa bioenergetics remains a prominent theme in the field of Andrology, whose advances are challenged by inter-species differences and difficulties in extrapolating data
