Structural biology is undergoing a transformative era driven by advances in artificial intelligence (AI)-based protein structure prediction and cryo-electron microscopy. Predictive approaches have dramatically expanded structural coverage across proteomes and are increasingly integrated into experimental workflows. However, protein function frequently depends on dynamic molecular processes including ligand-dependent conformational remodeling, transient interactions, cooperative assembly, and transport-state transitions that remain difficult to define from static computational models alone. Photodynamic therapy is traditionally based on reactive-oxygen-species-mediated tumor cell killing, but its immunomodulatory capacity is not fully explored. Plant-derived exosome-like nanovesicles provide a natural photosensitizer platform with potential for dual tumoricidal and immune-modulating effects. This study investigated whether light activation of Hypericum perforatum-derived exosome nanovesicles (HPDENs) remodels their microRNA cargo to enhance antitumor activity and reprogram the tumor immune microenvironment. Photoactivation significantly up-regulated miR-172f-p3 in HPDENs, which d

Plant cyclic nucleotide gated channels (CNGCs) facilitate cytosolic Ca2+ influx as an early step in numerous signaling cascades. CNGC-mediated Ca2+ elevations are essential for plant immune defense and high temperature thermosensing. In the present study, we evaluated phenotypes of CNGC2, CNGC4, CNGC6, and CNGC12 null mutants in these two pathways. It is shown CNGC2, CNGC4, and CNGC6 physically interact in vivo, whereas CNGC12 does not. CNGC involvement in immune signaling was evaluated by monitoring mutant response to elicitor peptide Pep3. Pep3 response cascades involving CNGCs included mito

In plants, age-related resistance (ARR) refers to a gain of disease resistance during shoot or organ maturation. ARR associated with vegetative phase change, a transition from juvenile to adult stage, is a widespread agronomic trait affecting resistance against multiple pathogens. How innate immunity in a plant is differentially regulated during successive stages of shoot maturation is unclear. In this work, we found that Arabidopsis thaliana showed ARR against its bacterial pathogen Pseudomonas syringae pv. tomato DC3000 during vegetative phase change. The timing of the ARR activation was ass

Some herbal extracts contain relatively high amounts of lipopolysaccharide (LPS). Because orally administered LPS activates innate immunity without inducing inflammation, it plays a role as an active ingredient in herbal extracts. However, the LPS content in herbal extracts remains extensively unevaluated. This study aimed to create a database of LPS content in herbal extracts; therefore, the LPS content of 414 herbal extracts was measured and the macrophage activation potential was evaluated. The LPS content of these hot water extracts was determined using the kinetic-turbidimetric method. Th

The interaction between environmental stressors, such as cold exposure, and immune function significantly impacts human health. Research on effective therapeutic strategies to combat cold-induced immunosuppression is limited, despite its importance. In this study, we aim to investigate whether traditional herbal medicine can counteract cold-induced immunosuppression. We previously demonstrated that cold exposure elevated immunoglobulin G (IgG) levels in mice, similar to the effects of intravenous immunoglobulin (IVIg) treatments. This cold-induced rise in circulating IgG was mediated by the re

Salicylic acid (SA) is an important signaling molecule of the plant immune system. In Arabidopsis thaliana, SA biosynthesis is indirectly modulated by the closely related transcription factors TGACG-BINDING FACTOR 1 and 4 (TGA1 and TGA4, respectively). They activate expression of SYSTEMIC ACQUIRED RESISTANCE DEFICIENT1, the gene product of which regulates the key SA biosynthesis gene ISOCHORISMATE SYNTHASE 1. Since TGA1 interacts with the SA receptor NONEXPRESSOR OF PATHOGENESIS-RELATED GENES 1 (NPR1) in a redox-dependent manner and since the redox state of TGA1 is altered in SA-treated plants

Salicylic acid (SA), jasmonic acid (JA), and ethylene (ET) are each involved in the regulation of basal resistance against different pathogens. These three signals play important roles in induced resistance as well. SA is a key regulator of pathogen-induced systemic acquired resistance (SAR), whereas JA and ET are required for rhizobacteria-mediated induced systemic resistance (ISR). Both types of induced resistance are effective against a broad spectrum of pathogens. In this study, we compared the spectrum of effectiveness of SAR and ISR using an oomycete, a fungal, a bacterial, and a viral p

The authors present large numbers of data on influence of the various mercury compounds on the immune system of humans and laboratory animals. The effect of mercury on the immune system was studied mainly in small rodents intoxicated mostly with HgCl2 or methylmercury. Contrary to a number of these data, analogous observations in humans are rare. Stimulating activity of mercury vapours on the immunological system has been reported in workers at chloralkali plants. So far no data are available concerning influence of the mercury vapours on the resistance of humans to neoplasms and infections.

Nematodes are ubiquitous metazoans that occupy diverse ecological niches, exposing them to a wide range of viruses and positioning them as both viral hosts and vectors. Free-living nematodes, particularly Caenorhabditis elegans and Caenorhabditis briggsae, provide powerful genetic models for studying antiviral immune mechanisms. Natural infections with noda-like viruses have revealed powerful antiviral defense strategies in Caenorhabditis species, including RNA interference, the Intracellular Pathogen Response, Signal Transducers and Activators of Transcription (STAT)-like transcriptional regu

Multiple sclerosis (MS) is an autoimmune disease that remains in need of effective therapies. Plant-derived medicines have appealing properties for the treatment of autoimmune diseases. MYMD-1 is a synthetic plant alkaloid that has been shown to ameliorate the course of autoimmune thyroiditis. The goal of the present study was to determine whether MYMD-1 would produce similar beneficial effects in a mouse model of MS, experimental autoimmune encephalomyelitis (EAE) induced by immunization with myelin oligodendrocyte glycoprotein. MYMD-1 improved the course of EAE and suppressed activation of e

Medicinal and edible plant-derived exosome-like nanovesicles (MEPELNVs) have attracted considerable attention as naturally derived immunomodulatory nanoplatforms for inflammatory disease intervention, owing to their excellent biocompatibility, low immunogenicity, and abundant bioactive cargoes capable of regulating immune responses. While recent reviews have broadly surveyed plant-derived exosome-like nanovesicles (PELNVs) across diverse botanical sources or focused on general isolation technologies and drug delivery applications, a systematic synthesis that specifically centers on medicinal a

Natural sweet potato transgene Ib-rolB/C enhances abiotic and biotic stress tolerance through coordinated regulation of ROS signaling, antioxidant defense, immune activation, and secondary metabolism. Horizontal gene transfer from Agrobacterium has contributed to plant genome evolution, yet the physiological roles of naturally integrated T-DNA genes remain poorly understood. Cultivated sweet potato (Ipomoea batatas) contains two cellular T-DNA regions, including the IbT-DNA2 locus harboring the naturally acquired gene Ib-rolB/C. Here, we investigated the stress-responsive expression and functi

Therapeutics based on fusing a protein of interest to the IgG Fc domain have been enormously successful, though fewer studies have investigated the vaccine potential of IgG fusions. In this study, we systematically compared the key properties of seven different plant-made human IgG1 fusion vaccine candidates using Zika virus (ZIKV) envelope domain III (ZE3) as a model antigen. Complement protein C1q binding of the IgG fusions was enhanced by: 1) antigen fusion to the IgG N-terminus; 2) removal of the IgG light chain or Fab regions; 3) addition of hexamer-inducing mutations in the IgG Fc; 4) ad

Traditional medicines are an integral part of health care worldwide, even though their efficacy has not been scientifically proven. HIV-infected individuals may use them singularly or in combination with conventional medicines. Many in vitro studies have proven the anti-HIV, anti-Candida, and anti-herpes simplex virus potential of traditional plants and identified some of the mechanisms of action. Very few in vivo studies are available that involve a small number of participants and show controversial results. In addition, knowledge is limited of the role of traditional medicines in the enhanc

Plants encounter diverse pathogens and have evolved a 2-layered innate immune system to detect pathogen molecules and activate defense mechanisms that restrict infection. Most cloned plant Resistance (R) genes encode NLR immune receptors. NLR genes are often found in clusters of paralogs with sequence and copy number variation; whether these NLR clusters evolve in response to single or multiple pathogens has been unclear. We report here the isolation of a Phytophthora capsici resistance gene, Rpc2, along with a novel P. infestans resistance gene, Rpi-amr5, from 2 Solanum americanum accessions.

Systemic acquired resistance (SAR) is an induced immune mechanism in plants. Unlike vertebrate adaptive immunity, SAR is broad spectrum, with no specificity to the initial infection. An avirulent pathogen causing local programmed cell death can induce SAR through generation of mobile signals, accumulation of the defense hormone salicylic acid, and secretion of the antimicrobial PR (pathogenesis-related) proteins. Consequently, the rest of the plant is protected from secondary infection for a period of weeks to months. SAR can even be passed on to progeny through epigenetic regulation. The Arab
