A study was made of the dynamics of pathohistological changes occurring in the embryos of albino rats under conditions of a single action of teratogenic doses of daraprim of the 9th day of pregnancy. Decidual tissue proved to be the most sensitive to daraprim the first disturbances in this tissue were noted to soon as one hour after the injection. Cells of various embryonic tissues reacted to daraprim differently. Most pronounced cytotoxic effect of daraprim is seen on the embryonic mesoderm and on the extraembryonic ectoderm. It is seen in both inpatient and outpatient settings with variable presentations ranging from mild, reversible injury to advanced kidney disease. Two infants with prominent frontal bossing, hydrocephalus, microphthalmia, and small, malformed, low-set, undifferentiated ears were born to mothers who had taken isotretinoin in the first trimester of pregnancy. A Dandy-Walker malformation, microcephaly, hypertelorism, small ear canals, cleft palate, small mouth, and congenital heart disease were also observed. Isotretinoin is a potent teratogen in man; maternal ingestion early in pregnancy leads to a distinct clinical pattern of anomalies.

An infant with moderate growth retardation, convulsions, cranio-facial dysmorphia, absence deformity of limbs and vertebral segmentation defect is described. She was born to a young mother who had taken castor oil seed (Ricinus communis) orally as a contraceptive for eight weeks after conception. No other drugs were taken during pregnancy. The features of this case partially resemble Fetal Hydantoin Syndrome.
