Studies of the binding of chromium(III) complexes to phosphate groups of adenosine triphosphate.

The binding of a number of chromium complexes to ATP has been studied by 31P NMR spectroscopy. The results are consistent with the formation of outer-sphere complexes between ATP and the chromium species. The magnitude of the binding constant has been determined as approximately 500 mol-1 dm3 (pH = 7.00) for the interaction of trisethylenediamine chromium(III) with ATP. Under our experimental conditions no NMR signals are observed from complexes in which chromium(III) is bound directly to the phosphate groups of ATP. The relative stability of the inner sphere complexes of chromium(III) and glu

Correlation between levels of plasma adenosine triphosphate and stress to the fetus at delivery.

Adenosine triphosphate (ATP) and its metabolites are released from intracellular stores and levels of ATP in the plasma are affected by hypoxia and by exercise. We examined levels of plasma ATP in the fetus at the time of obstetrical delivery. The arterial (ACB), venous cord (VCB) and maternal venous blood (MVB) samples were collected immediately after clamping of the cord. ATP was determined by the bioluminescence method. The levels of plasma ATP were significantly higher in ACB than in VCB or in MVB. There were significant correlations between the levels of ATP in ACB and in VCB, between the…

Calcium transport and catabolism of adenosine triphosphate in the protozoan parasite Giardia lamblia.

1. Calcium uptake by washed trophozoites of Giardia lamblia was dependent on inorganic orthophosphate and stimulated by glucose. Uptake was both rapid and substantial: 224 +/- 73 nmoles Ca2+/mg protein/min. 2. Known inhibitors of Ca2+ uptake in mammalian cells also impeded Ca2+ influx into G. lamblia. 3. The inhibitor studies indicated that Ca2+ transport in G. lamblia was an active process. Energy for such a process could be provided by the action of ATPases. 4. Two types of ATPases were found in the parasite; one, a membrane-associated enzyme activated by Ca2+; the other, a soluble, cytosoli

The content of erythrocyte 2,3 diphosphoglycerate and adenosine-5'-triphosphate in myelomatosis and leukaemia.

Ery-DPG and thus the oxygen-releasing capacity of haemoglobin increases in meylomatosis and in chronic myelocytic and lymphocytic leukaemia at decreased concentrations of haemoglobin, to the same extent as in other types of anaemia. Consequently the need for blood transfusions is less than assumed in these disorders. Ery-ATP is considerably increased in chronic myelocytic leukaemia, the values correlating positively with the 'activity' of the disease. This finding can probably be used for diagnostic and prognostic purposes.

Requirement for adenosine triphosphate for stimulation in vitro of ox growth-hormone release.

Rotenone, carbonyl cyanide chloromethoxyphenylhydrazone and 2,4-dinitrophenol cause parallel falls in ox pituitary ATP content and growth-hormone release in the presence of 65mm-K(+). Carbonyl cyanide chloromethoxyphenylhydrazone (0.4mug/ml) prevented the rise in growth-hormone release, but not the rise in 3':5'-cyclic AMP content after addition of prostaglandin E(2) (1mug/ml). The rate of hormone release from unstimulated slices and from slices in the presence of Ba(2+) at 2.3 or 6.9mm was proportional to the ATP content of the slices. Carbonyl cyanide chloromethoxyphenylhydrazone at concentr

The effect of vitamin A and other dietary constituents on the activity of adenosine triphosphate sulphurylase.

1. ATP sulphurylase was assayed in various organs from vitamin A-deficient and pair-fed control rats at different stages of deficiency. Activity decreased slightly in the liver and markedly in the adrenal gland. Striking differences in liver activity were observed between pair-fed control and ad libitum-fed animals. This observation suggested that diet (apart from vitamin A) strongly influenced the activity of ATP sulphurylase. 2. Total starvation caused a severe decrease in activity in liver within 48hr. This was due to a lack of protein intake. 3. By feeding groups of vitamin A-deficient and

Protein-Independent Liquid-Liquid Phase Separation of Adenosine Triphosphate Under Crowded Conditions.

Adenosine triphosphate (ATP), a common constituent of protein-rich biomolecular condensates, itself undergoes liquid-liquid phase separation (LLPS) even in the absence of polypeptides. Using polyethylene glycol as macromolecular crowding agent, we observed robust ATP droplet formation at pH 2-11. Moderate NaCl concentrations and lower temperatures promote LLPS by lowering critical ATP and crowder concentrations. While very high salt reverses this trend, the droplets nevertheless survive in hypersaline environments. We argue that both ion-mediated bridging and anomalous underscreening likely co…

Adenosine triphosphate release inhibitors targeting pannexin1 improve recovery after spinal cord injury.

Traumatic spinal cord injury is characterized by immediate and irreversible tissue loss at the lesion site and secondary tissue damage. Secondary injuries should, in principle, be preventable, although no effective treatment options currently exist for patients with acute spinal cord injury. Traumatized tissues release excessive amounts of adenosine triphosphate and activate the P2X purinoceptor 7/pannexin1 complex, which is associated with secondary injury. We investigated the neuroprotective effects of the blue dye Brilliant Blue FCF, a selective inhibitor of P2X purinoceptor 7/pannexin1 tha…

Increased cerebrospinal fluid adenosine 5'-triphosphate in patients with amyotrophic lateral sclerosis.

BACKGROUND: Extracellular adenosine 5'-triphosphate (ATP) has been suggested to cause neuroinflammation and motor neuron degeneration by activating microglia and astrocytes in amyotrophic lateral sclerosis (ALS). Since we have developed a highly sensitive ATP assay system, we examined cerebrospinal fluid (CSF) ATP levels in patients with ALS whether it can be a useful biomarker in ALS. METHODS: Forty-eight CSF samples from 44 patients with ALS were assayed for ATP with a newly established, highly sensitive assay system using luciferase luminous reaction. CSF samples from patients with idiopat

Cholesterol metabolism, apolipoprotein E, adenosine triphosphate-binding cassette transporters, and Alzheimer's disease.

PURPOSE OF REVIEW: Recent evidence suggests that cholesterol metabolism participates in the pathogenesis of Alzheimer's disease. Apolipoprotein E is the main lipid carrier in the brain and the best-established risk factor for late-onset Alzheimer's disease. Intracellular cholesterol levels influence the generation of amyloid-beta peptides, the toxic species thought to be a primary cause of Alzheimer's disease. Finally, compounds that modulate cholesterol metabolism affect amyloid-beta generation. This review summarizes data linking apolipoprotein E and adenosine triphosphate-binding cassette t…

Intracellular mechanism of mitochondrial adenosine triphosphate-sensitive potassium channel activation with isoflurane.

UNLABELLED: The precise mechanism of isoflurane and mitochondrial adenosine triphosphate-sensitive potassium channel (mitoK(ATP)) interaction is still unclear, although the mitoK(ATP) is involved in isoflurane-induced preconditioning. We examined the role of various intracellular signaling systems in mitoK(ATP) activation with isoflurane. Mitochondrial flavoprotein fluorescence (MFF) was measured to quantify mitoK(ATP) activity in guinea pig cardiomyocytes. To confirm isoflurane-induced MFF, cells were exposed to Tyrode's solution containing either isoflurane (1.0 +/- 0.1 mM) or diazoxide and …

Role of adenosine 5'-triphosphate in vasospasm after subarachnoid hemorrhage: human investigations.

OBJECTIVE: Adenosine 5'-triphosphate (ATP) is a vasoactive compound found in high concentrations inside erythrocytes. This compound may contribute to vasospasm after subarachnoid hemorrhage (SAH). We assessed the hypothesis that ATP contributes to vasospasm in humans. METHODS: ATP and hemoglobin concentrations were measured in cerebrospinal fluid (CSF) from humans with SAH and in blood incubated in vitro. The vasoactivity of the human CSF samples and of fractionated (fractions with molecular weight greater than or less than 10 kDa) and unfractionated blood incubated in vitro was assessed by a

Usefulness of adenosine triphosphate-atropine stress echocardiography for detecting coronary artery stenosis.

There have been few studies on adenosine triphosphate (AT) stress echocardiography. The AT stress test may have fewer adverse effects than the adenosine stress test. The addition of atropine to AT echocardiography may enhance the sensitivity for detection of coronary artery disease (CAD). The purpose of this study was to determine the utility of AT-atropine echocardiography for detection of CAD. The group studied consisted of 112 patients with suspected CAD. Sixty-one patients did not have a history of prior myocardial infarction (group I) and 51 patients did (group II). AT was infused intrave…

Adenosine 5'-triphosphate modulates membrane potassium conductance in guinea-pig myenteric neurones.

1. Intracellular recordings were made from myenteric neurones isolated from the guinea-pig small intestine to study actions of adenosine 5'-triphosphate (ATP). ATP was applied by superfusion (10 nM-100 microM) or pressure ejection from ATP-containing glass pipettes. 2. Myenteric neurones have been classified into two groups: type I/S neurones and type II/AH neurones. ATP produced a membrane hyperpolarization in 80% of AH neurones and a membrane depolarization in 90% of S neurones in a dose-dependent manner. Adenosine caused responses similar to those induced by ATP in both AH and S neurones, b

Aberrant Adenosine Triphosphate Release and Impairment of P2Y2-Mediated Signaling in Sarcoglycanopathies.

Sarcoglycanopathies, limb-girdle muscular dystrophies (LGMD) caused by genetic loss-of-function of the membrane proteins sarcoglycans (SGs), are characterized by progressive degeneration of skeletal muscle. In these disorders, muscle necrosis is associated with immune-mediated damage, whose triggering and perpetuating molecular mechanisms are not fully elucidated yet. Extracellular adenosine triphosphate (eATP) seems to represent a crucial factor, with eATP activating purinergic receptors. Indeed, in vivo blockade of the eATP/P2X7 purinergic pathway ameliorated muscle disease progression. P2X7…

Decreased intravesical adenosine triphosphate in patients with refractory detrusor overactivity and bacteriuria.

PURPOSE: Although several studies have examined the relationship between adenosine triphosphate release from the urothelium and bladder sensations including painful filling and urgency, the association between bacteriuria and urothelial adenosine triphosphate release has not been well studied. We evaluated women with refractory detrusor overactivity who were experiencing an acute exacerbation of detrusor overactivity symptoms including frequency, urgency and nocturia (and/or urge incontinence). We measured changes in intravesical adenosine triphosphate levels in these women with and without ba…

An efficient extraction method for quantitation of adenosine triphosphate in mammalian tissues and cells.

Firefly bioluminescence is widely used in the measurement of adenosine 5'-triphosphate (ATP) levels in biological materials. For such assays in tissues and cells, ATP must be extracted away from protein in the initial step and extraction efficacy is the main determinant of the assay accuracy. Extraction reagents recommended in the commercially available ATP assay kits are chaotropic reagents, trichloroacetic acid (TCA), perchloric acid (PCA), and ethylene glycol (EG), which extract nucleotides through protein precipitation and/or nucleotidase inactivation. We found that these reagents are part

Lactate and adenosine triphosphate in the extender enhance the cryosurvival of rat epididymal sperm.

We evaluated the cryosurvival of rat epididymal sperm preserved in raffinose-modified Krebs-Ringer bicarbonate-egg yolk extender supplemented with various energy-yielding substrates (glucose, pyruvate, lactate, and ATP) and assessed the effect on sperm oxygen consumption. The incubation of sperm at 37 degrees C for 10 min in lactate-free extender decreased sperm motility and oxygen consumption before and after thawing compared with those of sperm in glucose- and pyruvate-free mediums. We then focused on the effect of supplementing the extender with lactate (0, 10.79, 21.58, 32.37, and 43.16 mM
