Ameloblastic fibrosarcoma in adolescence: A rare case of a malignant odontogenic tumor.

A female patient, 15 years old, presented to the primary health unit with a complaint of vestibular and lingual swelling in the mandibular region, with a duration of 5 months. Imaging analysis revealed a destructive lesion with poorly defined margins. After an incisional biopsy, the lesion showed expansive and rapid growth. Microscopic analysis revealed an odontogenic tumor characterized by a biphasic composition of epithelial and mesenchymal components. The epithelial component showed unequivocal ameloblastoma-like features, without evidence of malignancy. In contrast, the spindle cell stroma

Conservative treatment of ameloblastic fibroma a case report with review of literature.

INTRODUCTION AND IMPORTANCE: Ameloblastic fibroma (AF) is a benign mixed odontogenic tumor composed of epithelial and mesenchymal tissues. It accounts for approximately 1.5-4.5 % of all odontogenic tumors and is most commonly diagnosed during the first and second decades of life. Due to documented cases of malignant transformation, the optimal therapeutic approach for AF remains a subject of debate. CASE PRESENTATION: A 15-year-old female patient presented with pain, swelling, and purulent discharge in the right posterior mandible. Panoramic radiography revealed a unilocular radiolucent lesio

Ameloblastic Carcinoma in a Young Adult Male Patient - A Case Report.

Ameloblastic carcinoma is a rare malignant odontogenic tumor that is further classified into being primary or secondary arising from a preexisting benign ameloblastoma. It affects the mandible in two thirds of the patients. There is no standard treatment protocol for this lesion but radicalsurgical excision with or without radiotherapy is reported in the majority of cases. In this paper, we present a case of a 22 year old male diagnosed with Ameloblastic carcinoma of the mandible with a clinical course of typical aggressiveness and extensive destruction. Histopathological examination of the in

Ameloblastic Fibro-Odontoma of the Posterior Mandible: A Rare Pathological Entity.

Ameloblastic fibro-odontoma (AFO) is a rare, slow-growing neoplastic lesion classified as a benign, epithelial mixed odontogenic tumor with odontogenic mesenchyme. This tumor demonstrates the histological features characteristic of both ameloblastic fibromas and complex odontomas. The clinical manifestation of AFO is typically characterized by asymptomatic enlargement of the jawbones. Radiographically, it presents as a distinct radiolucent region, indicating the presence of radiopaque substances with varying degrees of irregularities in size and morphology. Standard therapeutic intervention in

Transformation of ameloblastoma to ameloblastic carcinoma in a 10-year-old child.

Ameloblastic carcinoma (AC) is a rare odontogenic malignant epithelial neoplasm of maxillofacial skeleton with a distinct predisposition of the mandible. It can occur in a wide range of age groups, with a sex predilection in males. It can arise either as a de novo lesion or from preexisting ameloblastoma. AC has a high propensity for local recurrence as well as distant metastasis (chiefly lungs), thus requiring an aggressive surgical approach and a strict surveillance. Owing to the rarity of publications describing AC, little is known about this entity in pediatric patients. We report a case o

Diagnosis, Treatment, and Total Rehabilitation of a Secondary Type Ameloblastic Carcinoma.

Ameloblastic carcinoma (AC) is an uncommon malignant odontogenic tumor, with about 200 cases published in the literature. Here, we report a case of AC occurring in the maxilla - at the same anatomical location of an ameloblastoma 8 years earlier. A 23-year-old woman with maxillary swelling of 6-month duration was referred to our service. Extraoral examination revealed a swelling over the right side of the face, lifting her lip and nose. Intraorally, a firm, reddish, nodular swelling involving the right maxilla was observed. Computed tomography showed a hypodense image invading the right maxill

Establishment and characterization of immortalized mouse ameloblast-like cell lines.

OBJECTIVES: Enamel organ epithelium (EOE) gives rise to the major epithelial-derived cell types of tooth including the ameloblasts. The formation of enamel, termed amelogenesis, occurs through the cytodifferentiation of ameloblasts, ultimately leading to apoptosis and necrosis of these cells with eruption. Therefore, studies regarding enamel matrix formation and bioengineering have been limited. In this study, we establish and characterize two mouse immortalized ameloblast-like cell lines using human papillomavirus 16 (HPV16) E6/E7 oncogenes for the first time. SETTING AND SAMPLE POPULATION: …

Ameloblastic carcinoma: report of an aggressive case with multiple bony metastases.

Ameloblastic carcinoma is a rare primary tumor of the maxillofacial skeleton with a distinct predilection for the mandible. These lesions may initially show histologic features of ameloblastoma that dedifferentiate over time. Other ameloblastic carcinomas initially present with morphologic features suggestive of ameloblastoma with areas of epithelial dedifferentiation. We herein report a rare case of aggressive ameloblastic carcinoma in a 22-year-old white man who developed widespread bony metastases and expired 4 years after initial diagnosis.

Overexpression of transforming growth factor-beta1 in teeth results in detachment of ameloblasts and enamel defects.

Transforming growth factor-beta1 (TGF-beta1) is a key regulator of many cellular processes, including cell adhesion, the immune response and synthesis of extracellular matrix proteins. In the present study, we report the characterization of enamel defects in a transgenic mouse model overexpressing TGF-beta1 in odontoblasts and ameloblasts, its expression being driven by the promoter sequences of the dentin sialophosphoprotein gene. As reported earlier, these mice develop distinct dentin defects similar to those seen in human dentin dysplasia and dentinogenesis imperfecta. A further detailed ex…

Specific binding sites for transferrin on ameloblasts of the enamel maturation zone in the rat incisor.

During enamel maturation in rodents, an iron-containing pigment is deposited into the surface layer of the enamel. Maturation zone ameloblasts presumably are responsible for this deposition. The presence of large amounts of ferritin in the cytoplasm of these cells suggests that they receive iron, presumably from circulating transferrin. An in vivo radioautographic binding assay using iodinated transferrin was used to determine if indeed maturation ameloblasts possess transferrin receptors at their cell surfaces. Experimental rats received systemic injections of labeled transferrin while contro…

A systematic review on the molecular pathways of Ameloblastic carcinoma when compared to Ameloblastoma.

BACKGROUND: Ameloblastic carcinoma (AC), malignancy originating from the odontogenic epithelium, shows histological overlap with ameloblastoma (AM). In order to unravel mechanisms driving AC, it is imperative to understand the molecular distinction between these two entities. This systematic review aims to highlight molecular and immunohistochemical markers involved in the pathogenesis of AC and to distinguish it from its histological mimic, AM. RESEARCH DESIGN AND METHODS: Literature search across three databases including PubMed, Web of Sciences, and Scopus was carried out from year 1999 to

A rare case of peripheral ameloblastic fibroma manifesting as a maxillary gingival mass.

BACKGROUND: Ameloblastic fibroma (AF) is a rare benign odontogenic tumor characterized by the coexistence of epithelial and mesenchymal components, typically arising intraosseously in the posterior mandible during the first two decades of life. Its peripheral variant, occurring extraosseously in gingival soft tissue without bone involvement, is exceedingly rare. Owing to its clinical resemblance to reactive gingival lesions, peripheral AF poses diagnostic challenges and is frequently misdiagnosed without histopathological confirmation. CASE PRESENTATION: We report a case of peripheral AF in a

Ameloblastic Carcinoma In Situ: Review of Literature and a Case Presentation in a Pediatric Patient.

Ameloblastic carcinomas are rare malignant lesions with 3 mandibular pediatric cases reported in the literature. We present a case of ameloblastic carcinoma in situ in a 15-year-old male with a right mandibular cystic lesion on computed tomography. The incisional biopsy revealed plexiform ameloblastoma. Due to the infiltrating and aggressive nature of the tumor, the patient underwent hemimandibulectomy and immediate reconstruction with a vascularized osteocutaneous fibula free flap. The final pathology was read as ameloblastic carcinoma in situ. Given the rarity of this disease in the pediatri

The secretion of amelogenins is associated with the induction of enamel and dentinoid in an ameloblastic fibro-odontoma.

Ameloblastic fibro-odontoma is the unique entity of epithelial-ectomesenchymal odontogenic tumors, which is characterized by enamel formation in addition to dentine. We examined immunohistochemically a case of this tumor in which enamel having prism structures was developed in the absence of odontoblast differentiation but was in contact with mesenchymal matrices. Histological examination showed diverse morphological features of epithelial tumor cells, e.g., cuboidal cells comprising tooth bud-like projections, ameloblast- and stellate reticulum-like cells, and residual cells in forms of exten

Calcifying odontogenic cyst with ameloblastic fibro-odontome: one lesion or two?

The calcifying odontogenic cyst is unusual in that it is frequently found in conjunction with the histologic features of a variety of other odontogenic conditions. Two cases are reported, one of which contained areas histologically similar to the ameloblastic fibro-odontome and also showed condensations of cells in the stroma beneath epithelial strands proliferating from the cyst lining. In both cases the "ghost cells" stained strongly for disulphide groups but only occasional areas were positive for sulphydryl groups. No amyloid or "amyloid-like" material was detected. As no part of the curre

Identification of the functional activity of the [A-4] amelogenin gene splice product in newborn mouse ameloblasts.

In the mouse tooth organ, shortly after birth, ameloblasts acquire their secretory phenotype, which is characterized by the prominent expression and subsequent secretion of two isoforms of amelogenin, M180 and M59 (LRAP, [A-4]). Amelogenin deposition into the ameloblast extracellular matrix promotes enamel biomineralization. A complex set of intercellular signaling events, reciprocal communications between the developing oral epithelium and its underlying dental mesenchyme, guide the expression of amelogenin mRNA, and limit it to a defined period of tooth development. In tooth germ organ cultu…

Ameloblastic Fibroma: A Case Report of a Rare Presentation Illustrating a Diagnostic Challenge.

This case report emphasizes the diagnostic role of cone-beam computed tomography (CBCT) in identifying a rare pediatric case of ameloblastic fibroma (AF), comprising approximately 2% of odontogenic tumors. The 10-year-old patient initially presented with painless left mandibular posterior gingival swelling, resembling radicular or dentigerous cysts in clinical and conventional radiographic assessments. However, CBCT revealed a distinct periapical radiolucency linked to deciduous molar roots and an unerupted premolar, exposing mandibular lingual plate thinning and buccal plate disruption. The l
