[The midgut bacterial flora in lab-reared Anopheles sinensis].

OBJECTIVE: To screen gram-negative bacteria in the midguts of lab-reared Anopheles sinensis at 3 development stages (larva, unfed adult and engorged adult). METHODS: Twenty engorged adult An. sinensis mosquitoes were dissected, and bacteria were isolated from their midguts by plate cultivation. The bacteria were identified by using 16S rDNA V3 sequence and dyed with Gram-reaction solutions. Using denaturing gradient gel electrophoresis (DGGE), 16S rDNA clone libraries from common bacteria in the midguts of lab-reared An. sinensis at 3 development stages (20 engorged adults, 10 larva and 50 un…

Reproduction-longevity trade-off in Anopheles gambiae (Diptera: Culicidae).

Reduced survival and future reproduction due to of current reproduction is a trade-off known as the cost of reproduction. Surprisingly, only a few studies have assessed the cost of reproduction in arthropod disease vectors, despite its effect on longevity, and thus on vectorial capacity. We evaluated the cost of reproduction on survival of Anopheles gambiae Giles by comparing mosquitoes that were denied exposure to the other sex, hereafter named virgins, and those that were allowed exposure to the other sex and mating, hereafter named mated. Merely 6 d of exposure to females with mating activi…

Description and bionomics of Anopheles (Cellia) ovengensis (Diptera: Culicidae), a new malaria vector species of the Anopheles nili group from south Cameroon.

Mosquito species of the Anopheles nili group (Diptera: Culicidae) transmit malaria to humans along rivers in Africa. To date, the An. nili group includes the species Anopheles nili s.s. and its pale-winged variant known as the "Congo form," Anopheles somalicus and Anopheles carnevalei. Larval and adult mosquito collections in the forest region of Campo, in southern Cameroon, uncovered an additional morphological variant provisionally called "Oveng form" that was subsequently found to be genetically distinct from the other members of the An. nili group. In this study, we provide further biologi…

Compound effector genes suppress malaria parasite infections in gene-drive population modification strains of the African malaria mosquitoes, Anopheles gambiae and Anopheles coluzzii.

Malaria remains a major global health burden and is caused by protozoan parasites in the genus Plasmodium. Parasites are transmitted to humans during blood feeding by anopheline mosquitoes, and members of the Anopheles gambiae species complex are important vectors in sub-Saharan Africa. Gene-drive technologies offer promising options for disease control by enabling the spread of genetic traits through mosquito populations that block parasite transmission. We report here the development and characterization of four population modification gene-drive strains in Anopheles gambiae s.s. and An. col…

Promising approach to reducing Malaria transmission by ivermectin: Sporontocidal effect against Plasmodium vivax in the South American vectors Anopheles aquasalis and Anopheles darlingi.

BACKGROUND: The mosquito resistance to the insecticides threatens malaria control efforts, potentially becoming a major public health issue. Alternative methods like ivermectin (IVM) administration to humans has been suggested as a possible vector control to reduce Plasmodium transmission. Anopheles aquasalis and Anopheles darlingi are competent vectors for Plasmodium vivax, and they have been responsible for various malaria outbreaks in the coast of Brazil and the Amazon Region of South America. METHODS: To determine the IVM susceptibility against P. vivax in An. aquasalis and An. darlingi, …

Linking Anopheles bionomics and human behaviour in the Lao PDR.

BACKGROUND: Laos aims to eliminate malaria by 2030, with residual transmission present in the south. Understanding how LLINs function in relation to vector and human behaviours, and linking this to epidemiological outcomes, is critical for improving vector control strategies. Integrating human behaviour observations (HBOs) with entomological and epidemiological data helps identify gaps in protection and inform national malaria programs. METHODS: Entomological surveillance of Anopheles mosquitoes was conducted alongside HBOs in both rainy (July-September 2022) and dry (February-April 2023) sea…

Molecular evolution of gustatory receptors in the Anopheles gambiae complex.

BACKGROUND: Mosquitoes in the Anopheles (An.) gambiae species complex are major vectors of Plasmodium falciparum malaria. One reason for this is the high anthropophily of the constituent species An. coluzzii, An. gambiae sensu stricto, and An. arabiensis. In contrast, their sister species An. quadriannulatus is highly zoophilic. Anopheles mosquitoes largely rely on chemical cues for host-seeking, which are primarily detected by four chemosensory gene families: olfactory receptors (Ors), ionotropic receptors (Irs), gustatory receptors (Grs), and odorant binding proteins (Obps). Genes from these…

Spatial modeling of the population dynamics of Anopheles mosquitoes in Madagascar.

BACKGROUND: Malaria, whose parasites are transmitted by Anopheles mosquitoes, remains a major public health burden in Madagascar despite the control measures led by the National Malaria Control Program. Understanding the population dynamics of Anopheles mosquitoes is therefore essential to optimize malaria surveillance and control. This study aimed to develop a model incorporating environmental, climatic and agricultural determinants of Anopheles abundance to predict their spatiotemporal distribution. METHODS: We developed a model of spatiotemporal dynamics for four Anopheles species, vectors…

Review of Anopheles Mosquito Species, Abundance, and Distribution in Ethiopia.

BACKGROUND: Malaria is a major mosquito-borne disease in Ethiopia, and it is one of the leading causes of morbidity and mortality. Plasmodium falciparum and P. vivax are the two malaria-causing parasitic species commonly known to cause human malaria in Ethiopia. To better manage and control vectors transmitting malaria parasites, the abundance, distribution, and updated annotated list of Anopheles species present in Ethiopia are very important. METHODS: In order to compile a list of the species recorded in Ethiopia, 33 original research articles were collected. This work gives an updated list…

Status of insecticide resistance in Anopheles gambiae (s.l.) of The Gambia.

BACKGROUND: Vector control activities, namely long-lasting insecticidal nets (LLIN) and indoor residual spraying (IRS), have contributed significantly to the decreasing malaria burden observed in The Gambia since 2008. Nevertheless, insecticide resistance may threaten such success; it is important to regularly assess the susceptibility of local malaria vectors to available insecticides. METHODS: In the transmission seasons of 2016 and 2017, Anopheles gambiae (s.l.) larvae were sampled in or around the nine vector surveillance sentinel sites of the Gambia National Malaria Control Programme (GN…

A transgenic tool to assess Anopheles mating competitiveness in the field.

BACKGROUND: Malaria parasites, transmitted by the bite of an anopheline mosquito, pose an immense public health burden on many tropical and subtropical regions. The most important malaria vectors in sub-Saharan Africa are mosquitoes of the Anopheles gambiae complex including An. gambiae (sensu stricto). Given the increasing rates of insecticide resistance in these mosquitoes, alternative control strategies based on the release of genetically modified males are being evaluated to stop transmission by these disease vectors. These strategies rely on the mating competitiveness of release males, ho…

An agent-based model of the population dynamics of Anopheles gambiae.

BACKGROUND: Agent-based models (ABMs) have been used to model the behaviour of individual mosquitoes and other aspects of malaria. In this paper, a conceptual entomological model of the population dynamics of Anopheles gambiae and the agent-based implementations derived from it are described. Hypothetical vector control interventions (HVCIs) are implemented to target specific activities in the mosquito life cycle, and their impacts are evaluated. METHODS: The core model is described in terms of the complete An. gambiae mosquito life cycle. Primary features include the development and mortalit…

The genome of Anopheles darlingi, the main neotropical malaria vector.

Anopheles darlingi is the principal neotropical malaria vector, responsible for more than a million cases of malaria per year on the American continent. Anopheles darlingi diverged from the African and Asian malaria vectors ∼100 million years ago (mya) and successfully adapted to the New World environment. Here we present an annotated reference A. darlingi genome, sequenced from a wild population of males and females collected in the Brazilian Amazon. A total of 10 481 predicted protein-coding genes were annotated, 72% of which have their closest counterpart in Anopheles gambiae and 21% have h…

A genetic module regulates the melanization response of Anopheles to Plasmodium.

Two modes of refractoriness to Plasmodium, ookinete lysis and melanization, are known in the malaria vector, Anopheles gambiae. Melanization, a potent insect immune response, is manifested in a genetically selected refractory strain and in susceptible mosquitoes that are depleted of specific C-type lectins (CTLs). Here we use a systematic in vivo RNA interference-mediated reverse genetic screen and other recent results to define a melanization-regulating genetic module or network. It encompasses at least 14 genes, including those that encode five Easter-like clip domain serine proteases and fo…

The polyubiquitin gene of the mosquito Anopheles gambiae: structure and expression.

The polyubiquitin gene from the mosquito Anopheles gambiae has been cloned and sequenced, and its structure is reported along with sequence analysis results. The gene consists of approximately seven tandem head-to-tail repeat units of the seventy-six amino acid-coding ubiquitin monomer. It is expressed constitutively in larvae, pupae and adults of An. gambiae, as well as in a cell line derived from this mosquito species. A probe made from a DNA fragment containing the coding region of the gene recognizes transcripts of approximately 3.6 kb and 4.4 kb in RNA isolated from all mosquito developme…

Anopheles gambiae as a host for geographic isolates of Plasmodium vivax.

The G-3 strain of Anopheles gambiae was compared with 2 other strains of An. gambiae and An. freeborni, An. stephensi, and An. dirus for susceptibility to infection with 7 different geographic strains of Plasmodium vivax. Ratios of infection varied, indicating that certain strains of P. vivax were more infectious to the G-3 strain of An. gambiae than to other anopheline species/strains. Based on the comparative number of oocysts per mosquito, the relationships between the 3 strains of An. gambiae were closer than between the G-3 strain of An. gambiae and the 3 other species of Anopheles. Anoph…
