The blood lymphocytes of 37 splenectomised patients were analysed by means of T and B lymphocyte surface markers. Sixteen patients had had a splenectomy for non-haematological and 21 for haematological reasons. The results show that 15 had normal numbers of T and B cells; decreased T cells were found in two patients, raised B cells in seven, raised T and B cells in eight, and raised T cells in five patients. Cutaneous anergy to a wide variety of antigens first prompted the suggestion that the underlying defect may be of importance in the aetiology or pathogenesis of the disorder. Counts of T and B lymphocytes were made in the peripheral blood from patients with active and inactive pulmonary tuberculosis and in healthy subjects. In patients with active pulmonary tuberculosis the numbers of T cells were significantly lower than in the controls. After treatment for two months, if the patient's condition improved the percentage of T cells returned to nearly normal levels. If the patient failed to improve, the numbers of T cells remained low. The mechanism of these phenomena is discussed.

Lymphocytes of peripheral blood of patients with systemic lupus erythematosus, scleroderma and dermatomyositis were investigated, and relatively, with regard to T- or B-cell characteristics. T-lymphocytes of most patients with SLE were decreased both absolutely. B-lymphocytes mostly showed only a relative increase. Successful immunosuppressive treatment usually accompanied by normalization of T-cell number and with decrease of B-cells. This differentiation of lymphocytes is an additional aid in the determination of activity of SLE and a useful index of immunosuppressive-cytostatic treatment.

Large granular lymphocytes (LGL) may exert regulatory influences on B cell immunoglobulin synthesis. We, therefore, investigated the influence of LGL from controls and B cell chronic lymphocytic leukemia patients (B-CLL) on control B cell proliferation to costimulation with the F(ab')2 fragment of goat antihuman mu and B cell growth factor (BCGF). Purified LGL (greater than 90% by morphology) from control and B-CLL peripheral blood were added in various concentrations to purified control B cells and incubated with anti-mu and BCGF for 3 days. [3H]-thymidine uptake of B cells was then measured.

The population of B lymphocytes was assessed in the blood of 16 patients with agammaglobulinemia using immunofluorescence and EAC1423 reactivity as B-cell markers. Lymphocytes were fractionated on gradients of bovine serum albumin which are capable of separating lymphocytes into B-cell-rich and T-cell-rich populations. There was complete absence of B lymphocytes in the majority of patients with X-linked agammaglobulinemia.
