[Value of basophil studies in the diagnosis of drug allergies].

Mastocytes and basophils are the two cell populations involved in the reactions seen during drug allergies. The testing of basophils is the only and simplest way of studying sensitization in the IgE system by a drug antigen, especially when the skin tests cannot be carried out. This study is carried out in two steps: straight after the drug-induced accident, where a basophil count will give an idea of its allergic origin; and some time after the accident, where the human basophil degranulation test (H.B.D.T.) helps to identify in over 50 p. 100 of cases the responsible drug. The basophil count…

Cutaneous basophil hypersensitivity in the guinea pig conjunctiva.

Cutaneous basophil hypersensitivity (CBH) is an immunologic reaction extensively studied in the guinea pig. We elicited this reaction in the conjunctiva by systemically sensitizing guinea pigs to ovalbumin in incomplete Freund's adjuvant. Skin tests showed basophils in the skin one week after sensitization when challenged with ovalbumin. However, the conjunctiva showed an eosinophilic reaction with few basophils. This is compatible with other extracutaneous manifestation of the CBH reaction.

Ocular findings in systemic cutaneous basophil hypersensitivity.

SCBH was induced by immunizing guinea pigs with a protein antigen and challenging 1 week later with a large intraperitoneal dose of the same antigen. Animals developed a delayed-onset, erythematous skin rash and dermal infiltration by basophils and eosinophils. The uveal tracts of these animals were infiltrated by eosinophils, as were several other internal organs. The eye is affected in SCBH, and as in other forms of ocular cell-mediated hypersensitivity, the eosinophil is a prominent cellular component of these reactions.

Staphylococcal Extracellular Complement-Binding Protein Induces IL-4 Production in Murine Basophils.

Staphylococcus aureus is responsible for a wide range of pyogenic infections, food poisoning, and allergic inflammation. S. aureus produces a very wide variety of proteinaceous exotoxins, which are classified by structure as β-barrel pore-forming toxin, oligonucleotide/oligosaccharide-binding (OB)-fold/β-grasp proteins, and triple-helix bundle-containing proteins. We previously reported that three staphylococcal exotoxins belonging to the first two groups activate basophils and mast cells, which are innate immune cells that play a central role in allergic inflammation. In this study, we focuse…

Pathobiology and Regulation of Eosinophils, Mast Cells, and Basophils in Allergic Asthma.

Eosinophilia is a hallmark of allergic disorders, including asthma, allergic rhinitis, and atopic dermatitis. The onset and maintenance of allergic inflammation in atopic adults involves the activation of selective hemopoietic processes and the migration of mature and immature eosinophils to allergic tissue, where these cells release mediators of inflammation that participate in the regulation of inflammation. Eosinophils function in close cooperation with basophils and mast cells in allergic tissue, where crosstalk between these central effector cells regulates the inflammatory process. This …

The Role of Basophils in Atopic Dermatitis, from Pathogenesis to Therapeutic Perspectives.

Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by intense pruritus. The principal pathological features include abnormalities in the structure and function of the epidermis, as well as skin inflammation marked by the overexpression of T helper 2 cell (Th2) cytokines. Throughout the progression of AD, various immune cells contribute to its pathogenesis. Basophils, the least abundant granulocytes in the human peripheral circulation, have historically been overlooked. However, the advent of novel research tools has facilitated a renewed focus on the role of basophils …

Circulating Basophils as a Prognostic Marker for Response to Bacillus Calmette-Guérin.

PURPOSE: To predict recurrence and progression in non-muscle-invasive bladder cancer (NMIBC) patients receiving bacillus Calmette-Guérin (BCG), we evaluated circulating basophils as a biomarker that could be detected from the complete blood count. PATIENTS AND METHODS: We use a pooled cohort of patients from the Centre Hospitalier Universitaire de Québec-Université Laval (2016-2020) and the Vancouver General Hospital (2010-2018) where a complete blood count was available before transurethral resection of bladder tumor (TURBT) of a high-grade NMIBC and subsequent BCG. Descriptive statistics de…

Higher Circulating Levels of Neutrophils and Basophils Are Linked to Myopic Retinopathy.

This retrospective study investigated circulating immune cell alteration in patients with myopic retinopathy. Blood test results and demographic and ocular information of 392 myopic patients and 129 emmetropia controls who attended Changsha Aier Eye Hospital from May 2017 to April 2022 were used in this study. Compared with emmetropia, the percentages of neutrophils and basophils and neutrophil/lymphocyte ratio were significantly higher in myopic patients, whereas the percentages of monocytes and lymphocytes and the counts of lymphocytes and eosinophils were significantly lower in myopic patie…

Studies of the role of basophils in aspirin-exacerbated respiratory disease pathogenesis.

BACKGROUND: Aspirin-exacerbated respiratory disease (AERD) is characterized by the triad of chronic rhinosinusitis with nasal polyps (CRSwNP), asthma, and intolerance to cyclooxygenase-1 enzyme inhibitors. The underlying mechanisms contributing to AERD pathogenesis are not fully understood, but AERD is characterized by an enhanced type 2 inflammatory phenotype. Basophils are potent type 2 effector cells, but their involvement in AERD pathophysiology remains unclear. OBJECTIVE: We sought to characterize the systemic and local basophil responses in patients with AERD compared with patients with…

Zinc-binding metallothioneins are key modulators of IL-4 production by basophils.

Zinc (Zn) is an essential nutrient, and Zn deficiency causes immunodeficiency and skin disorders. Basophils express FcɛRI on their surface and release multiple mediators after receptor cross-linking, including large amounts of IL-4. However, the mechanisms involved in the FcɛRI-mediated regulation of basophil IL-4 production are currently unclear. Here, we show that the Zn-binding metallothionein (MT) proteins are essential for the FcɛRI-induced basophil production of IL-4. Basophils from MT-I/II(-/-) mice produced significantly less FcɛRI-induced IL-4 than did wild-type basophils. The MTs wer…

Basophils help establish protective immunity induced by irradiated larval vaccination for filariasis.

Basophils are increasingly recognized as playing important roles in the immune response toward helminths. In this study, we evaluated the role of basophils in vaccine-mediated protection against filariae, tissue-invasive parasitic nematodes responsible for diseases such as elephantiasis and river blindness. Protective immunity and immunological responses were assessed in BALB/c mice vaccinated with irradiated L3 stage larvae and depleted of basophils with weekly injections of anti-CD200R3 antibody. Depletion of basophils after administration of the vaccination regimen but before challenge infe…

Basophils are essential initiators of a novel type of chronic allergic inflammation.

Basophils represent less than 1% of peripheral blood leukocytes and have often been considered as minor and possibly redundant circulating mast cells. The recent finding that basophils readily generate large quantities of T helper 2 (Th2) cytokines such as IL-4 provided new insights into the possible role of basophils in allergic disorders and immunity to pathogens. However, in-depth studies on basophils, particularly their functions in vivo, have been hampered by the lack of appropriate animal models, such as mutant animals deficient only in basophils. Here, we established a mAb that reacted …

Human basophils release interleukin-4 after stimulation with Schistosoma mansoni egg antigen.

The elevated interleukin (IL)-4 and IgE production in Schistosoma mansoni infection seems to be induced essentially by the egg stage of the parasite. The underlying mechanism, however, is not known. Since basophils from human peripheral blood can produce IL-4, we asked, whether soluble S. mansoni egg antigens (SEA) would trigger basophils to release IL-4. Basophils from healthy human donors (n = 32) without prior history of schistosomiasis were incubated with SEA in the presence of IL-3. In all donors, IL-4 was produced at different concentrations. The IL-4 production was dependent on the dose…

Histamine release from human basophils by the insect allergen Chi t I.

Hemoglobins of the Diptera species Chironomus thummi thummi (Chi t I) are potent inhalant allergens. Chi t I-specific histamine release was measured by a new radioimmunoassay in whole human blood taken from 20 sensitized patients, 11 exposed nonsensitized probands, and 11 nonexposed controls. The sensitized patients, who all had positive skin tests and radioallergosorbent test results with Chi t I, showed a significantly higher histamine release than the two other groups. However, within the patient group, the percentage of released histamine did not correlate with the intensity of the skin te

The pharmacologic control of mediator release from human basophils and mast cells.

Insight into the pathogenesis of human allergic and inflammatory disorders has been obtained through a combination of in vitro and in vivo studies. These investigations have demonstrated that human basophils and mast cells release mediators after nonimmunologic as well as immunologic activation in vitro and in vivo: nonimmunologic triggers include changes in osmolarity. Although these cells share many properties, including the presence of high-affinity receptors for IgE on their cell surface, the presence of histamine in granules, the ability to generate and release large quantities of leukotr…

5-hydroxyeicosatetraenoic acid and 12-hydroxyeicosapentaenoic acid regulate basophil and mast cell activation and basophil recruitment in chronic spontaneous urticaria.

BACKGROUND: The underlying pathomechanisms of wheal reaction enlargement in chronic spontaneous urticaria (CSU) remain largely unclear. We hypothesized that the activation of mast cells (MCs) and recruitment of basophils to locally inflamed skin would be enhanced by lipid mediators (LMs) following initial skin MC activation. This study aimed to identify the LMs responsible for enhancing the aggregation of IgE-mediated MC activation and basophil chemotaxis. METHODS: We enrolled 77 CSU patients and 36 non-atopic control (NC) subjects. Lipid profiling of plasma from these subjects was performed …

Anaphylactic degranulation of guinea pig basophilic leukocytes. II. Evidence for regranulation of mature basophils during recovery from degranulation in vitro.

Mature circulating granulocytes have been considered functionally end stage cells unable to reconstitute their specific cytoplasmic granules. We have reevaluated this assumption by studying guinea pig peripheral blood basophils maintained in vitro for periods up to 72 hours after anaphylactic degranulation. Guinea pig basophils degranulated in vitro by exposure to either specific antigen (sheep serum) or lectin (Concanavalin A) synthesized new cytoplasmic granules. This process was characterized by the appearance of abundant rough endoplasmic reticulum, activation of the Golgi zone, interioriz…
