Harnessing natural deep eutectic solvents Pickering emulsion droplets for biocatalytic production of diacylglycerols via glycerolysis: Elucidating the underlying mechanism of lipase thermostability.

Glycerolysis is a crucial approach for the preparation of diacylglycerols (DAG). The challenges such as enzyme stability and interfacial mass transfer have been insufficiently addressed. To validate the Pickering interfacial biocatalysis (PIB) for the desired target, a boosting system containing Pickering emulsion droplets based on natural deep eutectic solvents (NADES) was developed as a reaction medium for enhancing the catalysis efficiency of CALB-catalyzed (Candida antarctica lipase B, CALB) glycerolysis. The effects of types of oils and NADES on the properties of Pickering emulsions and c

A novel engineered malonyl-CoA synthetase from Astragalus mongholicus with improved catalytic performance.

Malonyl-CoA is a key precursor for the biosynthesis of numerous active natural products. Malonyl-CoA synthetases (MatBs) provide a direct route to malonyl-CoA from malonic acid and CoA. However, the limited availability of plant MatBs with high catalytic efficiency restricts their application in biocatalysis. In this study, we identified a novel malonyl-CoA synthetase AmMatB from Astragalus mongholicus. Biochemical assays confirmed that recombinant AmMatB directly catalyzes malonyl-CoA formation from malonic acid and CoA, while also accepting methylmalonate as a substrate. To enhance its utili

Thermoresponsive Hybrid Nanostructures for Indirect Regulation of Lipase Activity.

Stimuli-responsive regulation of enzyme activity is an important strategy to optimize biocatalytic performance across diverse operating environments. Herein, we report a thermoresponsive hybrid nanomaterial (HM) composed of Candida antarctica fraction B (CALB) immobilized on gold nanoparticles (AuNPs) and coated with poly(N-isopropylacrylamide) (PNIPAm). This system exhibits a reversible phase transition temperature (Tcp) at 33°C, enabling temperature-dependent modulation of the enzymatic activity. Our data show that, below Tcp, the extended conformation of PNIPAm acts as a protective layer, l

SAM-dependent methyltransferases: from chemoenzymatic natural product synthesis to industrial application.

Covering: up to 2026Over the past decades, research on S-adenosyl-L-methionine (SAM)-dependent methyltransferases (MTs) has led to their emergence as highly selective biocatalysts for late-stage methylation reactions, serving as new tools within the chemoenzymatic synthesis of complex natural products and chemical scaffolds. Studies focussing on the identification of novel MTs, their engineering and process optimisation, have not only expanded the accessible chemical space for biocatalytic methylations but have also significantly improved their applicability for reactions conducted at preparat

Enzymes as Immunotherapeutics.

Enzymes are attractive as immunotherapeutics because they can catalyze shifts in the local availability of immunostimulatory and immunosuppressive signals. Clinical success of enzyme immunotherapeutics frequently hinges upon achieving sustained biocatalysis over relevant time scales. The time scale and location of biocatalysis are often dictated by the location of the substrate. For example, therapeutic enzymes that convert substrates distributed systemically are typically designed to have a long half-life in circulation, whereas enzymes that convert substrates localized to a specific tissue o

Rigid-Flexible Layered Immobilization Enables Precise Confinement and Dynamic Activation of Small Enzymes.

Small enzymes (10-50 kDa) encounter challenges of unstable enzyme-carrier interactions and low activity retention during immobilization, with existing strategies lacking specificity. Unlike previous industrial enzyme-focused studies, a rigid-flexible layered immobilization strategy integrating networked metal-organic frameworks (MOFs) into flexible hydrogels is proposed. This innovative strategy stabilizes enzyme conformation, strengthens enzyme-matrix interactions, and prevents enzyme leakage via a dual-pore system. This synergistic system, composed of MOF micropores (50-150 nm, confinement)

Discovery of FeP/Carbon Dots Nanozymes for Enhanced Peroxidase-Like Catalytic and Antibacterial Activity.

Iron phosphide/carbon (FeP/C) serving as electrocatalysts exhibit excellent activity in oxygen reduction reaction (ORR) process. H2O2 catalyzed by peroxidase (POD) is similar to the formation of new electron transfer channels and the optimization of adsorption of oxygen-containing intermediates or desorption of products in ORR process. However, it is still a challenge to discover FeP/C with enhanced POD-like catalytic activity in the electrocatalytic database for biocatalysis. The discovery of FeP/carbon dots (FeP/CDs) nanozymes driven by electrocatalytic activity for enhanced POD-like ability

Designing artificial cofactors and their enzymes together.

When replacing expensive redox cofactors, making biomimetic cofactors with desirable thermostability and redox properties is only half of the equation; enzymes must also be able to use them. Ji and colleagues confront that trade-off by pairing the development of a thermostable carbocyclic cofactor with a dedicated regeneration enzyme for sustained biocatalysis.

Exploiting Archaeal/Thermostable Enzymes in Synthetic Chemistry: Back to the Future?

Billions of years of evolution have led to the selection of (hyper)thermophiles capable of flourishing at elevated temperatures. The corresponding native (hyper)thermophilic enzymes retain their tertiary and quaternary structures at near-boiling water temperatures and naturally retain catalytically competent conformational dynamics under these conditions. And yet, while hyper/thermophilic enzymes offer special opportunities in biocatalysis and in hybrid bio/chemocatalytic approaches to modern synthesis in both academia and industry, these enzymes remain underexplored in biocatalysis. Among the

Biocatalytic structural diversification of saponins in Zygophyllum decumbens: HR-LC-ESI-TOF-MS/MS profiling and bioactivity modulation.

BACKGROUND: Zygophyllum decumbens has been traditionally used in the treatment of gout, fungal infections, diabetes, and rheumatism. These therapeutic effects are primarily attributed to its triterpenoid saponins (TS). The present study explores the biocatalytic transformation of TS to examine structural modifications, identify rare saponins, and evaluate associated changes in biological activity. METHODS: Triterpenoid saponins were isolated from shade-dried aerial parts by cold maceration in aqueous alcohol, followed by fractionation and precipitation with acetone, yielding (TS). Three microb

[Research advances in efficient biocatalytic transformation mediated by metal-organic frameworks].

Due to its high selectivity, mild reaction conditions, and environmentally friendly characteristics, biocatalysis has shown important potential in the fields of fine chemical synthesis, bioenergy conversion, and environmental governance. However, biocatalysts, including enzymes and microorganisms, generally have problems such as insufficient stability, easy inactivation, difficult recycling, and poor reusability in practical applications, which seriously limit their industrialization process. Recently, metal-organic frameworks (MOFs) have gradually become ideal carriers for constructing effici

Enhancing Enzyme Activity Via Noncovalent Association with DNA-Functionalized Nanoparticles.

Enhancing enzyme performance without protein engineering remains a central challenge in biocatalysis. Here, we report the unusual discovery that nanoparticles densely functionalized with DNA, commonly known as spherical nucleic acids (SNAs), can bind enzymes without chemical conjugation, affinity tags, or engineered recognition motifs and substantially increase catalytic activity. Using cytochrome c as a model peroxidase, we show that noncovalent binding to SNAs increases the apparent turnover number by up to 15-fold. Mechanistic studies by varying DNA density, length, and nanoparticle core id

Repurposing haemoproteins for asymmetric metal-catalysed H atom transfer.

Transition metal-hydrides have been widely exploited in catalysis for the hydrofunctionalization of unsaturated moieties, including carbonyls, alkenes and alkynes1. To complement heterolytic metal-hydride bond cleavage, metal-hydride hydrogen atom transfer (MHAT) has recently gained attention, as a promising strategy for radical hydrofunctionalization of unactivated alkenes2, thus enabling late-stage diversification of complex molecules3,4. However, owing to the weak interactions between the prochiral organic radical and the enantiopure catalyst5, asymmetric MHAT6 remains challenging. Here we

AoiQ Catalyzes Geminal Dichlorination of 1,3-Diketone Natural Products.

Enzymes that can perform halogenation of aliphatic carbons are of significant interest to the synthetic and biocatalysis communities. Here we describe the characterization of AoiQ, a single-component flavin-dependent halogenase (FDH) that catalyzes gem-dichlorination of 1,3-diketone substrates in the biosynthesis of dichlorodiaporthin. AoiQ represents the first biochemically reconstituted FDH that can halogenate an enolizable sp3-hybridized carbon atom.

Endogenous crude Scutellaria baicalensis polysaccharide robustly enhances one-pot extraction and deglycosylation of baicalin.

With the extensive application of baicalein in the treatment of cardiovascular and cerebrovascular diseases, its clinical and market demand has gradually expanded. But the natural yield of baicalein is very low, and it is mainly prepared by the deglycosylation of baicalin. However, the insolubility of baicalin in water significantly limits the deglycosylation of it under biocatalysis. To make biocatalysis of baicalin more efficient and environmental, a strategy was designed to enhance its water solubility through the solubilization mechanism of endogenous biological macromolecules, and the eff

Construction of a highly efficient DNA nanotube sensor with peroxide-like activity.

The G-quadruplex/heme complexes are special DNA-based artificial metalloenzymes with peroxidase-like activity and are widely used in biosensing and biocatalysis. However, their peroxidase-like activity is not satisfactory. Due to the high programmability and good stability of DNA, DNA as a scaffold material is promising for enhancing the activity of artificial metalloenzymes. In this work, an effective DNA nanotube-based peroxidase was constructed using a self-assembly strategy. To improve the activity of G-quadruplex/heme complexes, a new method for the construction of G-quadruplex/heme compl

Repurposing Thiamine-Dependent Benzaldehyde Lyases as Visible-Light-Driven Radical Acyltransferases.

Thiamine diphosphate (ThDP)-dependent enzymes are among nature's most elegant biocatalysts for C-C bond formation and cleavage, typically operating through a two-electron umpolung mechanism. Although a small subset of ThDP enzymes, such as pyruvate: ferredoxin oxidoreductase (PFOR), has been shown to proceed via single-electron radical pathways, this knowledge had not yet been harnessed for asymmetric radical reactions. Inspired by thiamine biocatalysis, chemists have developed biomimetic N-heterocyclic carbene (NHC) complexes since the 1950s, and more recently, established radical NHC catalys

Asymmetric assembly of congested contiguous stereocenters via photobiocatalytic three-component radical coupling.

Despite rapid advances in new-to-nature biocatalysis, trimolecular enzymatic reactions that generate previously inaccessible molecular entities, particularly those bearing well-defined, congested stereochemical dyads, remain exceedingly rare. Here, we report a novel diastereo- and enantioselective photobiocatalytic three-component radical coupling unknown in both organic chemistry and enzymology, enabled by cooperative catalysis employing an evolved pyridoxal biocatalyst and a transition-metal photosensitizer.
