Influence of space flight on red blood cells.

Losses of red blood cell mass (RCM) averaging 10-15% have been observed consistently in astronauts after space flight; postflight recovery of RCM requires 4-6 wk. Although apparently not harmful to the health and effectiveness of crews during uncomplicated flights, decreased RCM could compromise health and performance in the event of illness, injury, or partial malfunction of the life support system. Whether the loss of RCM would worsen or stabilize in missions longer than 7 months is unknown. As a biological response, it is a significant, predictable reaction whose etiology, biological mechan

Interaction between intestinal and peripheral blood mononuclear cells.

Effective methods are available to isolated immunocytes from human intestinal mucosa. Isolated mononuclear cells from the mucosa of patients with Crohn's disease and normal controls were classified according to ultrastructure and cell-surface profile by the use of a panel of monoclonal antibodies. In both compartments of the mucosa, i.e., the lamina propria and the epithelial layer, no differences were found between intestinal tissue from control patients and unaffected mucosa of patients with Crohn's disease. Inflamed mucosa was characterized by an accumulation of surface-Ig+ cells and, to a

Electrophoretic analysis of red blood cells in polycythaemia.

In contrast with previously published observations, this study has found no increase in the net negative surface charge of the red blood cells (RBC) from patients with various forms of polycythaemia when compared with normal subjects. Two sub-populations of RBCs with different electrophoretic mobilities (EPM), termed fast and slow, were present in samples from both patients and normal individuals. Frequently, the slow cell population in RBCs from patients with polycythaemia, particularly the apparent polycythaemia sub-group, was of a lower EPM and contained more cells than the corresponding po…

Trophoblast cells in peripheral blood from pregnant women.

The presence of human trophoblast cells in maternal blood was investigated by the use of flow cytometry and a monoclonal antibody reacting against a specific antigen present on the surface of these cells. Three types of cells derived from the placenta could be detected in the peripheral blood obtained from women between 6 weeks' gestation and term. One group of cells were polynucleated, a second group were diploid, and a third consisted of anucleate cells derived from the syncytiotrophoblast. These cells should be suitable for prenatal diagnosis of chromosomal and biochemical abnormalities.

Blood chimeric twins: an example of blood cell chimerism.

A man and his twin sister were found to have two types of circulating red cells. The man had 89% A1 cells of his own and 11% A1B cells derived from his sister. The woman had 12% A1B cells of her own and 88% A1 cells derived from her brother. The two lines of red cells also had different Duffy and Kidd groups. The lymphocyte karyotype in the brother was 68% XY and 32% XX, and that in the sister was 60% XX and 40% XY. The possible mechanisms for producing different proportions of red cells and lymphocytes are discussed.

NK cells in peripheral blood carry trogocytosed tumor antigens from solid cancer cells.

The innate immune lymphocyte lineage natural killer (NK) cell infiltrates tumor environment where it can recognize and eliminate tumor cells. NK cell tumor infiltration is linked to patient prognosis. However, it is unknown if some of these antitumor NK cells leave the tumor environment. In blood-borne cancers, NK cells that have interacted with leukemic cells are recognized by the co-expression of two CD45 isoforms (CD45RARO cells) and/or the plasma membrane presence of tumor antigens (Ag), which NK cells acquire by trogocytosis. We evaluated solid tumor Ag uptake by trogocytosis on NK cells

The proliferative effects of asbestos-exposed peripheral blood mononuclear cells on mesothelial cells.

Malignant mesothelioma (MM) is thought to arise from the direct effect of asbestos on mesothelial cells. However, MM takes a long time to develop following exposure to asbestos, which suggests that the effects of asbestos are complex. The present study examined the effects of asbestos exposure on the cell growth of MeT-5A human mesothelial cells via cytokines produced by immune cells. Peripheral blood mononuclear cells (PBMCs) were stimulated with antibodies against cluster of differentiation (CD)3 and CD28 upon exposure to the asbestos chrysotile A (CA) or crocidolite (CR); the growth of MeT-

Antibodies in plasma and blood cells of horses.

BACKGROUND: Understanding the distribution of antibodies in the bloodstream is critical for advancing diagnostic and therapeutic approaches in veterinary medicine and comparative immunology. While most studies have focused on plasma antibodies, the presence and role of membrane-bound immunoglobulins remain underexplored, particularly in equine species such as the Jabe horse. AIM: This study aimed to analyze the amount of antibodies in the plasma and blood cells of Jabe horses for their potential future use. METHODS: Blood samples were collected from Jabe horses and separated into plasma and …

The lifelong transcriptomic landscape of human blood cells.

Single-cell technologies have transformed our view of normal and malignant hematopoiesis, yet a framework linking lifelong homeostatic hematopoiesis to hematological diseases in blood ecosystem remains incomplete. Here, we constructed a high-resolution landscape of the entire hematopoietic system by integrating transcriptomes of ∼1 million cells from 207 healthy samples spanning five developmental stages (fetal, neonatal, childhood, adult, elderly), and identified 96 blood cell clusters, including a VNN2-marked monocyte subset in the fetus and TSHZ2-marked T cell subsets exhibiting distinct ag

Adenosine receptors in blood cells: Biomarkers and beyond.

The identification of reliable, non-invasive biomarkers that reflect molecular alterations in deep and inaccessible tissues remains a major challenge in translational medicine. In this context, peripheral blood cells have emerged as valuable surrogates of tissue-level pathophysiology. Among molecular systems expressed in circulating cells, adenosine receptors (ARs) represent particularly attractive candidates due to their central role in regulating inflammation, immune responses, vascular function, and tissue homeostasis. This review focuses on the expression and functional modulation of AR su…

Case Series: Malignant Flower Cells in Peripheral Blood.

The presence of lymphoid cells with flower-like nuclei in peripheral blood is an uncommon morphologic finding, classically associated with Human T-lymphotropic virus type 1 (HTLV-1) infection related Adult T-cell Leukemia/Lymphoma (ATLL). However, similar cytologic features may be observed in other hematolymphoid malignancies. To describe cases of hematologic neoplasms presenting with flower cells in the peripheral blood and to highlight their broader diagnostic spectrum. We report a series of patients with distinct hematologic malignancies who demonstrated flower cells on peripheral blood sme

[When the blood cells suffer after a TAVR].

We present here a case of documented paraprosthetic valvular leak following TAVI treated medically initially. This led to a poorly tolerated hemolytic anemia. We were able to correct this paraprosthetic valvular leak by a postdilation of the TAVI valve with a good result and uncomplicated follow-up.

New putative phenol oxidase in ascidian blood cells.

The phenol oxidase system is ancient and ubiquitously distributed in all living organisms. In various groups it serves for the biosynthesis … ke haemocyanins, which are oxygen transporting plasma proteins, Tuphoxin is synthesised in ascidian blood cells and secreted in the extracellular matrix of the tunic-ascidian outer coverings. Single mature transcript coding for this phenol oxidase can give several protein products of different sizes. Thus limited proteolysis of the initial protein is suggeste …

GM-Free Generation of Blood-Derived Neuronal Cells.

Many human neurological disorders are caused by degeneration of neurons and glial cells in the brain. Due to limitations in pharmacological and other therapeutic strategies, there is currently no cure available for the injured or diseased brain. Cell replacement appears as a promising therapeutic strategy for neurodegenerative conditions. To this day, neural stem cells (NSCs) have been successfully generated from fetal tissues, human embryonic cells (ES) or induced pluripotent stem cells (iPSC). A process of dedifferentiation was initiated by activation of the novel human GPI-linked glycoprote

Vesiculation of healthy and defective red blood cells.

Vesiculation of mature red blood cells (RBCs) contributes to removal of defective patches of the erythrocyte membrane. In blood disorders, which are related to defects in proteins of the RBC membrane, vesiculation of the plasma membrane is intensified. Several hypotheses have been proposed to explain RBC vesiculation but the exact underlying mechanisms and what determines the sizes of the vesicles are still not completely understood. In this work, we apply a two-component coarse-grained molecular dynamics RBC membrane model to study how RBC vesiculation is controlled by the membrane spontaneou…

Human red blood cells alterations in primary aldosteronism.

CONTEXT: Aldosterone (Aldo) effects include NADPH oxidase activation involved in Aldo-related oxidative stress. Red blood cells (RBCs) are particularly sensitive to oxidative assault, and both the formation of high molecular weight aggregates (HMWAs) and the diamide-induced Tyr phosphorylation (Tyr-P) level of membrane band 3 can be used to monitor their redox status. OBJECTIVE: The Aldo-related alterations in erythrocytes were evaluated by comparing in vitro evidence. DESIGN: This was a multicenter comparative study. STUDY PARTICIPANTS: The study included 12 patients affected by primary al…
