Co-evolution and exploitation of host cell signaling pathways by bacterial pathogens.

Bacterial pathogens have evolved by combinations of gene acquisition, deletion, and modification, which increases their fitness. Additionally, bacteria are able to evolve in "quantum leaps" via the ability to promiscuously acquire new genes. Many bacterial pathogens - especially Gram-negative enteric pathogens - have evolved mechanisms by which to subvert signal transduction pathways of eukaryotic cells by expressing genes that mimic or regulate host protein factors involved in a variety of signaling cascades. This results in the ability to cause diseases ranging from tumor formation in plants

Nuclei number in human embryos co-cultured with human ampullary cells.

A study was undertaken to evaluate the effect on nuclei number in human embryos cultured in vitro with primary cell lines of human Fallopian tube epithelium. The development of 203 surplus human embryos, cultured in standard culture medium (Earle's balanced salt solution + 15% A5) with or without ampullary cells, was observed from day 2 to day 5.5 post-insemination. The expanded blastocysts in both culture conditions were analysed for nuclei numbers per blastocyst. Embryos transferred to co-culture at the 2-cell stage had an average of 120.7 nuclei per blastocyst, which was significantly highe

Cell composition and co-stored peptides in the endocrine pancreas of Rana arvalis.

The endocrine pancreas of Rana arvalis studied by immunochemistry consisted of islets and diffuse endocrine cells. The islets showed a mammalian-like arrangement with a central core of B cells and a peripheral mantle of A/PP cells. A few D and vasoactive intestinal polypeptide cells were also present. Consecutive sections or double-labeling studies allowed us to detect several regulatory peptides colocalized in the same endocrine cells. The so-called A/PP cells contained subpopulations of cells showing various types of immunoreactivity and varying degrees of immunolabeling. Generally, glucagon

Metabolic co-operation between embryonic and embryonal carcinoma cells of the mouse.

Mouse embryonal carcinoma (EC) cells form permeable junctions at their homotypic cell-to-cell contacts which permit intercellular exchange of metabolites (metabolic co-operation). Hooper & Slack (1977) showed how this exchange could be detected by autoradiography as the transfer of [3H]nucleotides between PC13 (a pluripotential EC line) and PC13TG8 (a variant of PC13 which is deficient in hypoxanthine guanine phosphoribosyltransferase). We now show that cells taken from several different tissues of early mouse embryos, that is, from the morula, the inner cell mass of the blastocyst, and the en

Multicompartment hydrogel microcapsules for creating spatially patterned cell co-cultures.

There is increasing clinical evidence that pancreatic dysfunction in diabetes needs to be viewed in the context of crosstalk with the liver as well as other organs. Our goal for this study was to develop a pancreas-liver co-culture system suited for mechanistic and therapy testing studies in the context of multi-organ cross talk. To achieve this goal, we developed a co-axial flow-focusing microfluidic device to fabricate multi-compartment hydrogel microcapsules. Each microcapsule contained two aqueous compartments or cores surrounded by poly(ethylene glycol) (PEG) hydrogel. Each microcapsule h

Tumor organoid-immune cell co-culture systems for precision oncology.

Three-dimensional tumor organoids, particularly patient-derived organoids (PDOs), recapitulate key morphological, genetic, and functional features of original tumors. Co-culture with immune cells enables studies of the tumor immune microenvironment (TIME) and holds promise for personalized immunotherapy. In this review, we critically evaluate established methodologies for tumor organoid-immune cell co-culture, including reductionist, holistic (tumor slice culture and air-liquid interface), and organoid-on-a-chip approaches. We provide quantitative benchmarking of success rates, immune cell per

Mechanistic dissection of GRHL2 and PR transcriptional co-regulation in breast cells.

Gene expression is controlled by complex transcriptional networks in which transcription factors and their cognate enhancer elements integrate developmental and environmental cues. The progesterone receptor (PR), a hormone-activated transcription factor, is essential for breast development and physiology, yet how it engages with the chromatin and lineage-specific cofactors remains unclear. Using an unbiased approach, we identify the epithelial transcription factor grainyhead-like 2 (GRHL2) as a key co-regulator of PR activity in hormone responsive breast cancer cells. We show that GRHL2 intera

A multidimensional platform to support intestinal epithelial and immune cell co-culture.

INTRODUCTION: Detailed research into intestinal immune exposure to luminal contents, including nutrients and microbial metabolites, could be well supported by accurate and reproducible models of the human intestinal milieu. Animal models provide insights but are not fully predictive of human biology, while human correlative studies are hampered by the complexity of feedback signals from established immune reactions. Therefore, characterization of human nutrient uptake and immediate effects on intestinal health will benefit from ex vivo models that provide controlled experimental conditions and

Brooklyn plots to identify co-expression dysregulation in single cell sequencing.

Altered open chromatin regions, impacting gene expression, is a feature of some human disorders. We discovered it is possible to detect global changes in genomically-related adjacent gene co-expression within single cell RNA sequencing (scRNA-seq) data. We built a software package to generate and test non-randomness using 'Brooklyn plots' to identify the percent of genes significantly co-expressed from the same chromosome in ∼10 MB intervals across the genome. These plots establish an expected low baseline of co-expression in scRNA-seq from most cell types, but, as seen in dilated cardiomyopat

Characterization of The Retinal Progenitor Cells Generated Using Co-Culture Systems.

OBJECTIVE: Degeneration of the photoreceptors due to retinal disorders can affect vision, and even lead to blindness. Recently therapeutic progress in retinal degeneration, using human embryonic stem cells (hESCs), has been facing technical challenges, demanding the development of simple and standardized protocols. In addition to the designing of the protocols, characterization of the obtained cells is highly required for confirming the reliability of the applied methods for future medical applications. Previously, we showed that human stem cells from apical papilla (SCAP) have stromal cell-de

In Vitro Generation of Brain Regulatory T Cells by Co-culturing With Astrocytes.

Regulatory T cells (Tregs) are normally born in the thymus and activated in secondary lymphoid tissues to suppress immune responses in the lymph node and at sites of inflammation. Tregs are also resident in various tissues or accumulate in damaged tissues, which are now called tissue Tregs, and contribute to homeostasis and tissue repair by interacting with non-immune cells. We have shown that Tregs accumulate in the brain during the chronic phase in a mouse cerebral infarction model, and these Tregs acquire the characteristic properties of brain Tregs and contribute to the recovery of neurolo

Adenovirus Co-Opts Neutrophilic Inflammation to Enhance Transduction of Epithelial Cells.

Human adenoviruses (HAdV) cause a variety of infections in human hosts, from self-limited upper respiratory tract infections in otherwise healthy people to fulminant pneumonia and death in immunocompromised patients. Many HAdV enter polarized epithelial cells by using the primary receptor, the Coxsackievirus and adenovirus receptor (CAR). Recently published data demonstrate that a potent neutrophil (PMN) chemoattractant, interleukin-8 (IL-8), stimulates airway epithelial cells to increase expression of the apical isoform of CAR (CAREx8), which results in increased epithelial HAdV type 5 (HAdV5

Co-signaling receptors regulate T-cell plasticity and immune tolerance.

BACKGROUND: Aphasia is a common neurocognitive disorder caused by impaired speech and language, with stroke being the most frequent cause. The neuroanatomical mechanism underlying this condition is not yet fully understood. CASE DESCRIPTION: This case describes a 74-year-old Caucasian woman admitted with a clinical picture of right total anterior circulation infarct (TACI) and aphasia, scoring 17 on the National Institutes of Health Stroke Scale. Neuroimaging showed a large cortico-subcortical frontotemporoparietal and insular infarct involving the basal ganglia of the right hemisphere and bi

Nocardia cyriacigeorgica and Aspergillus Co-infection in a Patient with Giant-cell Arteritis.

UNLABELLED: Immunosuppressed patients are at greater risk of unusual infections. The authors present the case of a woman with giant-cell arteritis, on oral steroids, who developed cavitating pneumonia due to co-infection with Aspergillus and Nocardia. Reports of such co-infection are rare in the literature. This case highlights the importance of considering rare pathogens in immunosuppressed patients who present with non-specific symptoms, as well as the impact of such pathogens on clinical management. Another important issue is the need for prophylaxis against Nocardia spp. in immunocompromis

The Contribution of Co-signaling Pathways to Anti-malarial T Cell Immunity.

Plasmodium spp., the causative agent of malaria, caused 212 million infections in 2016 with 445,000 deaths, mostly in children. Adults acquire enough immunity to prevent clinical symptoms but never develop sterile immunity. The only vaccine for malaria, RTS,S, shows promising protection of a limited duration against clinical malaria in infants but no significant protection against severe disease. There is now abundant evidence that T cell functions are inhibited during malaria, which may explain why vaccine are not efficacious. Studies have now clearly shown that T cell immunity against malari

Co-storage and secretion of growth hormone and secretoneurin in retinal ganglion cells.

It is well established that growth hormone (GH) and granins are co-stored and co-secreted from pituitary somatotrophs. In this work we demonstrate for the first time that GH- and secretoneurin (SN) immunoreactivity (the secretogranin II (SgII) fragment) are similarly present in retinal ganglion cells (RGCs), which is an extrapituitary site of GH expression, and in quail QNR/D cells, which provide an experimental RGC model. The expression of SgII and chromogranin A in the pituitary gland, neuroretina and QNR/D cells was confirmed by RT-PCR analysis. Western blotting also showed that the SN-immu

p53 Aggregates penetrate cells and induce the co-aggregation of intracellular p53.

Prion diseases are unique pathologies in which the infectious particles are prions, a protein aggregate. The prion protein has many particular features, such as spontaneous aggregation, conformation transmission to other native PrP proteins and transmission from an individual to another. Protein aggregation is now frequently associated to many human diseases, for example Alzheimer's disease, Parkinson's disease or type 2 diabetes. A few proteins associated to these conformational diseases are part of a new category of proteins, called prionoids: proteins that share some, but not all, of the ch

Co-localization of tryptase and cathepsin-G in mast cells in cutaneous mastocytosis.

Mastocytosis is a heterogeneous disease characterized by an abnormal growth and/or accumulation of clonal mast cells (MC) in one or more organs. The most frequent site of organ involvement is the skin. The aim of this study was to investigate the immunoreactivity to tryptase and to cathepsin-G of MC from human cutaneous mastocytosis and to compare their number in normal skin and cutaneous mastocytosis. Immunohistochemistry and dual immunofluorescence using anti-tryptase and anti-capthepsin-G antibodies was performed on biopsy specimens from 20 cases diagnosed as cutaneous mastocytosis. Tryptas

Metabolic stability of experimental chemotherapeutic agents in hepatocyte:tumor cell co-cultures.

A U.S. National Cancer Institute screening program for new anticancer drugs, based on the growth of primary human tumor cells in an in vitro soft agar colony formation assay, has resulted in the identification of a number of compounds that have cytotoxic activity against primary human tumor cells in vitro but are inactive in the conventional in vivo murine P388 leukemia animal model pre-screen. To investigate whether metabolic inactivation ov the compounds might be a factor in the lack of in vivo cytotoxicity we have co-cultured rat hepatocytes with A204 rhabdomyosarcoma and murine P388 leukem
