Intrinsic survival mechanisms for retinal ganglion cells.

Retinal ganglion cells (RGCs) undergo programmed cell death (apoptosis) after axonal injury. This cell death is mediated by several mechanis … d on the mechanisms of cell death after axonal injury, it may be that determining the regulation of cell survival mechanisms may lead to innovative methods for neuroprotection. The final common pathway of glaucomatous optic neuropathy is RGC death, probably via damage to their axons occurring at or near the lamina cribrosa. Axonal injury leads directly (1) or i …

Somatic cells compartmentalise their carbohydrate metabolism to sustain germ cell survival.

To ensure success in reproduction, organisms dedicate substantial resources to supporting the germline. In testes, somatic gonadal cells for … sing autonomous consumption of carbohydrates in somatic cells increases germ cell death. Thus, germ cell survival depends on functional metabolic compartmentalisation within gonadal somatic support cells.

Autophagy in PDGFRα+ mesenchymal cells is essential for intestinal stem cell survival.

Autophagy defects are a risk factor for inflammatory bowel diseases (IBDs) through unknown mechanisms. Whole-body conditional deletion of autophagy-related gene (Atg) Atg7 in adult mice (Atg7Δ/Δ) causes tissue damage and death within 3 mo due to neurodegeneration without substantial effect on intestine. In contrast, we report here that whole-body conditional deletion of other essential Atg genes Atg5 or Fip200/Atg17 in adult mice (Atg5Δ/Δ or Fip200Δ/Δ) caused death within 5 d due to rapid autophagy inhibition, elimination of ileum stem cells, and loss of barrier function. Atg5Δ/Δ mice lost PDG

miR-181c regulates MCL1 and cell survival in GATA2 deficient cells.

GATA2 is a transcription factor critical for hematopoiesis. Germline mutations in GATA binding protein 2 (GATA2) led to haploinsufficiency, severe cytopenias of multiple cell lineages, susceptibility to infections and strong propensity to develop myelodysplastic syndrome, and acute myeloid leukemia. Mechanisms of progressive cytopenias remain unclear. MicroRNA (miRNA) represents a unique mechanism of post-transcriptional gene regulation. In this study, miRNA profiles were evaluated and eight miRNAs were found to be differentially expressed (≥2-fold, P ≤ 0.05) in patient-derived cell lines (N =

Effect of cell-bound proteins on the in vivo survival of circulating blood cells.

Normal circulating red blood cells (RBCs) and platelets have been shown to have small amounts of IgG on their membranes. The cell-bound IgG may be cytophilic (IgG nonspecifically adsorbed from the plasma) and/or IgG autoantibody. It has been suggested that most of the RBC-bound IgG is on older RBCs and is an autoantibody directed against senescent cell antigen (SCA). The accumulation of this RBC-bound IgG leads to Fc-dependent removal of senescent RBCs by macrophages in the reticuloendothelial system. RBCs also have complement components on their membrane; it is not clear how this accumulates

Survival by Design: Cell Fate Dynamics in Classical Hodgkin Lymphoma.

Classical Hodgkin lymphoma (cHL) is singular because rare Hodgkin and Reed-Sternberg (HRS) cells survive within an abundant inflammatory tumor microenvironment (TME) - despite originating from germinal center (GC) B cells that would normally be negatively selected, they persist through coordinated disruption of lineage fidelity, immune recognition, and cell-death execution. The present review integrates mechanistic, translational, and therapeutic evidence on apoptosis resistance, survival signaling, microenvironmental support, immune evasion, Epstein-Barr virus-associated rescue mechanisms, an

LAMTOR2 Reprograms Energy Metabolism to Sustain Hepatocellular Carcinoma Cell Survival.

Hepatocellular carcinoma (HCC) is characterized by profound metabolic reprogramming to support rapid proliferation and survival under nutrient stress. However, the central regulatory hubs that coordinate these diverse metabolic programs remain poorly understood. Here, we identify LAMTOR2 as a critical coordinator of energy metabolism and survival in HCC cells. LAMTOR2 is markedly upregulated in HCC tissues, and high LAMTOR2 expression correlates with poor survival. LAMTOR2 depletion inhibits HCC cell proliferation and induces significant cell death, effects that are substantially rescued by LA

Tumor virus-induced lineage survival circuit drives Merkel cell carcinogenesis.

Background: Artificial intelligence (AI) integrated circuits (IC) have memory devices as the key component. Due to more complex algorithms and architectures required by neuroscience and other medical applications, various memory structures have been widely proposed and investigated by involving nanomaterials, such as memristors. Methods: Due to reliability issues of mass production, the dominant memory devices in many computers are still dynamic random access memory (DRAM). A DRAM has one transistor and one capacitor, and so it contains two devices and requires a more compact design to replace

Epigenetic modifications in retinal ganglion cell survival and axonal regeneration.

The optic nerve, a component of the central nervous system (CNS), comprises axons from retinal ganglion cells (RGCs) that exhibit limited regenerative capacity following injury. Recent advances have substantially deepened our understanding of the epigenetic mechanisms underlying RGC survival and axonal regeneration, encompassing DNA methylation, histone modifications, noncoding RNAs (ncRNAs), RNA methylation, and their complex interplay. Here, we review emerging research paradigms that underscore the potential of epigenetic modulation in RGC survival promotion and axonal regeneration. We furth

Hydrogen sulfide: A whiff of trouble for cancer cell survival.

Hydrogen sulfide (H2S) can regulate biological processes by post-translational persulfidation of proteins at select cysteine residues. In this issue of Molecular Cell, Zheng et al.1 identify the enzyme SAHH as an H2S substrate, which upon persulfidation disrupts homocysteine metabolism and sensitizes lung cancer cells to ferroptosis.

Survival of total hip arthroplasty (THA) in sickle cell disease.

BACKGROUND: The objective of this study was to evaluate the outcome of THA done in patients with sickle cell disease at a single center, and compare with rest of the world before we could recommend routine THA in SCD patients. METHODS: In 92 patients, 117 THAs were performed in sickle cell patients. Seventy-six (76) patients had homozygous hemoglobin (Hgb SS) disease, whereas sixteen had sickle cell trait (Hgb AS). Patients with Grade III Ficat and above avascular necrosis of the head of femur (ANFH) with cementless porous-coated proximal femur fixation were included in this analysis. Harris

Cell-Autonomous Processes That Impair Xenograft Survival into the Cerebellum.

In immunocompetent animals, numerous factors including the immune system of the host regulate the survival of neuro-glial precursors transplanted into the cerebellum. We transplanted human neuro-glial precursors derived in vitro from partial differentiation of IPS cells into the developing cerebellum of mice and rats before maturation of the host immune system. These approaches should facilitate the development of immune-tolerance for the transplanted cells. However, we found that human cells survived the engraftment and integrated into the host cerebellum and brain stem up to about 1 month po

Appendiceal goblet cell carcinomas have poor survival despite completion surgery.

PURPOSE: Appendiceal goblet cell carcinomas (aGCCs) are rare but aggressive tumours associated with significant mortality. We retrospectively reviewed the outcomes of aGCC patients treated at our tertiary referral centre. METHODS: We analysed aGCC patients, diagnosed between 1990-2016, assessing the impact of completion surgery and tumour factors on survival. Survival was assessed using Kaplan-Meier analysis. RESULTS: We identified 41 patients (23 F, 18 M); median age 61 (range 27-79) years. Mean tumour size was 10.5 (range 0.5-50) mm; most tumours were located in the appendiceal tip (n = 18

Genetic modifiers of long-term survival in sickle cell anemia.

BACKGROUND: Sickle cell anemia (SCA) is a clinically heterogeneous, monogenic disorder. Medical care has less-than-optimal impact on clinical outcomes in SCA in Africa due to several factors, including patient accessibility, poor access to resources, and non-availability of specific effective interventions for SCA. METHODS: Against this background, we investigated 192 African participants who underwent whole exome sequencing. Participants included 105 SCA patients spanning variable clinical expression: a "long survivor" group (age over 40 years), a "stroke" group (at least one episode of over

Assessing fractional hair cell survival in archival human temporal bones.

OBJECTIVES/HYPOTHESIS: Histopathological analysis of hair cell survival in human temporal bone sections has historically been binarized such that each hair cell row is rated as either present or absent, thereby greatly underestimating the amount of hair cell loss. Here, we describe and validate a technique to reliably assess fractional hair cell survival in archival sections stained with hematoxylin and eosin (H&E) using high-resolution light microscopy and optical sectioning. STUDY DESIGN: Technique validation. METHODS: Hair cell counts in archival temporal bone slide sets were performed by…
