Ceramides increase mitochondrial permeabilization to trigger mtDNA-dependent inflammation in astrocytes during brain ischemia.

The brain is rich in lipids, and disorders or abnormalities in lipid metabolism can induce neurotoxicity. Ceramides are the central intermediates of sphingolipid metabolism. This study was designed to investigate the potential lipotoxicity of ceramides in brain ischemia. First, a pseudo-targeted lipidomics analysis of plasma samples from stroke patients found significantly elevated levels of long-chain ceramides. A similar observation was made in mice subjected to permanent middle cerebral artery occlusion (pMCAO) surgery. In cultured cells, it was found that the altered ceramides were mainly …

Alterations in urinary ceramides, sphingoid bases, and their phosphates among patients with kidney disease.

BACKGROUND: To avoid an invasive renal biopsy, noninvasive laboratory testing for the differential diagnosis of kidney diseases is a desirable goal. As sphingolipids are demonstrated to be involved in the pathogenesis of various kidney diseases, we investigated the possible usefulness of the simultaneous measurement of urinary sphingolipids for differentiating kidney diseases. MATERIALS AND METHODS: Residual urine specimens were collected from patients who had been clinically diagnosed with chronic glomerulonephritis (CGN), diabetic mellitus (DM), systemic lupus erythematosus (SLE), and arter

Inhibiting tau-induced elevated nSMase2 activity and ceramides is therapeutic in murine Alzheimer's disease.

BACKGROUND: Cognitive decline in Alzheimer's disease (AD) is associated with prion-like tau propagation between neurons along synaptically connected networks, in part via extracellular vesicles (EV). EV biogenesis is triggered by ceramide enrichment at the plasma membrane from neutral sphingomyelinase2(nSMase2)-mediated cleavage of sphingomyelin. We report, for the first time, that tau expression triggers an elevation in brain ceramides and nSMase2 activity. METHODS: To determine the therapeutic benefit of inhibiting this elevation, we evaluated the efficacy of PDDC, the first potent, selecti…

Ceramide Metabolic Pathways and Their Regulation in Adolescent Acne Vulgaris.

Ceramides constitute approximately 50% of the lipid mass in the stratum corneum and are essential for epidermal permeability barrier function. In acne vulgaris, barrier disruption is accompanied by altered ceramide composition, yet the molecular mechanisms that connect acne-associated signals to specific changes in ceramide metabolism remain incompletely understood. This review integrates evidence from lipidomic profiling, enzymatic pathway characterization, and inflammatory signaling studies to examine how ceramide biosynthesis and turnover are modulated in acne-prone skin. Particular attenti…

Ceramide: A critical lipid mediator in the progression of diabetic nephropathy.

Diabetic nephropathy (DN) is one of the most severe microvascular complications of diabetes mellitus (DM). The complex pathogenesis is driven by the synergistic effects of glycotoxicity and lipotoxicity. In recent years, ceramides, especially C16:0 ceramides as the core hub of sphingolipid metabolism, have been shown to participate in the development of DN. This systematic review examines the mechanisms of ceramides metabolic dysregulation in DN, elucidating the molecular triggers responsible for selective accumulation of C16:0 ceramides under hyperglycemia and insulin-resistant conditions. Cutaneous ceramide synthesis is dysregulated in pediatric eosinophilic esophagitis.

BACKGROUND: Eosinophilic esophagitis (EoE), like atopic dermatitis (AD), is a disease of epithelial barrier dysfunction. The predisposing factors remain under investigation and few commonalities between the skin and the esophagus are known. There are no surrogate markers for EoE risk. OBJECTIVE: We sought to assess for concurrent skin and esophageal dysfunction by evaluating ceramide (CER) levels in the skin of children with EoE. METHODS: We collected up to 21 skin tape strips (STSs) from the volar forearm of non-AD subjects with EoE (n = 21) and non-AD, non-EoE controls (n = 17) and evaluat

Evidence for ceramide induced cytotoxicity in retinal ganglion cells.

Ceramides are bioactive compounds that play important roles in regulating cellular responses to extracellular stimuli and stress. Previous studies have shown that ceramides contribute to retinal degeneration associated with ischemic and ocular hypertensive stress. Acid sphingomyelinase (ASMase) is one of the major enzymes responsible for the stress-induced generation of ceramides. The goals of this study are to investigate the effects of ceramides on retinal ganglion cells (RGCs) and of ASMase inhibition in ocular hypertensive mice. Induced pluripotent stem cell (iPSC)-derived RGCs and primary…

The Pathogenic and Therapeutic Implications of Ceramide Abnormalities in Atopic Dermatitis.

Ceramides play an essential role in forming a permeability barrier in the skin. Atopic dermatitis (AD) is a common chronic skin disease associated with skin barrier dysfunction and immunological abnormalities. In patients with AD, the amount and composition of ceramides in the stratum corneum are altered. This suggests that ceramide abnormalities are involved in the pathogenesis of AD. The mechanism underlying lipid abnormalities in AD has not yet been fully elucidated, but the involvement of Th2 and Th1 cytokines is implicated. Ceramide-dominant emollients have beneficial effects on skin barr…

Ceramide structure dictates glycosphingolipid nanodomain assembly and function.

Gangliosides in the outer leaflet of the plasma membrane of eukaryotic cells are essential for many cellular functions and pathogenic interactions. How gangliosides are dynamically organized and how they respond to ligand binding is poorly understood. Using fluorescence anisotropy imaging of synthetic, fluorescently labeled GM1 gangliosides incorporated into the plasma membrane of living cells, we found that GM1 with a fully saturated C16:0 acyl chain, but not with unsaturated C16:1 acyl chain, is actively clustered into nanodomains, which depends on membrane cholesterol, phosphatidylserine an

Lysosomal Ceramide Metabolism Disorders: Implications in Parkinson's Disease.

Ceramides are a family of bioactive lipids belonging to the class of sphingolipids. Sphingolipidoses are a group of inherited genetic diseases characterized by the unmetabolized sphingolipids and the consequent reduction of ceramide pool in lysosomes. Sphingolipidoses include several disorders as Sandhoff disease, Fabry disease, Gaucher disease, metachromatic leukodystrophy, Krabbe disease, Niemann Pick disease, Farber disease, and GM2 gangliosidosis. In sphingolipidosis, lysosomal lipid storage occurs in both the central nervous system and visceral tissues, and central nervous system patholog…

Ceramide-enriched membrane domains in infectious biology and development.

Ceramide has been shown to be critically involved in multiple biological processes, for instance induction of apoptosis after ligation of death receptors or application of gamma-irradiation or UV-A light, respectively, regulation of cell differentiation, control of tumor cell growth, infection of mammalian cells with pathogenic bacteria and viruses or the control of embryo and organ development to name a few examples. Ceramide molecules form distinct large domains in the cell membrane, which may serve to re-organize cellular receptors and signalling molecules. Thus, in many conditions, ceramid

Perturbation of sphingolipid metabolism and ceramide production in HIV-dementia.

Infection by the human immunodeficiency virus type 1 (HIV-1) often results in neurological dysfunction including HIV dementia (HIVD). Alterations in cytokine and redox balance are thought to play important roles in the pathogenesis of HIVD, but the specific mechanisms underlying neuronal dysfunction and death are unknown. Activation of cytokine receptors and oxidative stress can induce the production of ceramide from membrane sphingomyelin, and recent findings suggest that ceramide is an important mediator of a form of programmed cell death called apoptosis. We now report that levels of cerami
