The protective effect of melatonin against brain oxidative stress and hyperlocomotion in a rat model of mania induced by ouabain.

This study was designed to investigate the protective effect of melatonin in a preclinical animal model of mania induced by ouabain (OUA). Male Wistar rats were pretreated with melatonin (5 or 20mg/kg; intraperitoneal, i.p.) for seven days or with the mood stabilizer lithium chloride (positive control) (45 mg/kg, per oral, p.o.). One day after the last dose, animals received an intracerebroventricular (i.c.v.) injection of OUA (5μl, 10(-5)M), a Na(+)K(+)ATPase-inhibiting compound. Locomotor activity was assessed in the open-field test (OFT). The levels of reactive species (RS), protein carbony

Peroxidase-catalyzed bromination of tyrosine, thyroglobulin, and bovine serum albumin: comparison of thyroid peroxidase and lactoperoxidase.

A recent paper (Buchberger, W., 1988, J. Chromatogr. 432, 57) on lactoperoxidase-catalyzed bromination of tyrosine and thyroglobulin stated, without evidence, that thyroid peroxidase (TPO) is able to use bromide as a substrate. This was in disagreement with unpublished experiments previously performed in this laboratory, and we undertook, therefore, to examine this subject further. Highly purified porcine TPO was compared with lactoperoxidase (LPO) and chloroperoxidase (CPO) for ability to catalyze bromination of tyrosine, thyroglobulin, and bovine serum albumin (BSA). The incubation mixture c

Diverse microplastics reprogram rhizosphere processes and constrain photosynthetic performance in barley through coordinated changes in oxidative regulation, nutrient cycling, enzyme hotspots, and microbial organization.

This study examined short-term responses of barley to polyethylene and polyvinyl chloride microplastics in a controlled 60-day greenhouse rhizobox experiment with three replicates per treatment. Microplastics were applied at 0, 2, and 4% of dry soil mass, corresponding to high experimental loadings of 20,000 and 40,000 mg kg-1, respectively. Bulk and rhizosphere soils were evaluated separately using enzyme zymography, nutrient analysis, amplicon sequencing, microbial network analysis, Mantel tests, and structural equation modeling. Higher polyethylene exposure and both polyvinyl chloride treat

Catechin (epigallocatechin-3-gallate) supplement restores the oxidation: antioxidation balance through enhancing the total antioxidant capacity in Wistar rats with cadmium-induced oxidative stress.

Catechins are bioactive flavanols commonly found in the fruits and leaves of plants, particularly the fresh tea leaves. This experimental study aims to evaluate the antioxidant properties of epigallocatechin-3-gallate, one of the most prominent catechins, and its ability to mitigate cadmium-induced oxidative stress. Eighty rats were randomly assigned to four groups of 20: an untreated control group (group 1), a catechin-treated group (group 2), a cadmium-exposed group (group 3), and a cadmium-catechin group (group 4). Group 2 rats received daily oral doses of catechin at 300 mg/kg body weight,

Microalgae-based integrated treatment of tannery wastewater: emphasizing microbial synergy for sustainable remediation.

Tannery wastewater (TWW) presents a complex and toxic mixture of pollutants, including heavy metals such as chromium, cadmium, lead, and nickel, as well as organic compounds, surfactants, sulfides, chlorides, and dyes. Conventional treatment methods often lead to high operational costs, sludge generation, and disposal issues. This review critically examines the integration of microalgae-based systems with physical, chemical, and biological methods for advanced TWW remediation, emphasising mechanistic insights and consortia-driven treatment. Microalgae facilitate pollutant removal through bioso

The killing of newborn larvae of Trichinella spiralis by eosinophil peroxidase in vitro.

Helminth infections in mammals are characterized by a high level of eosinophils in parasitized tissues and blood, and it has recently been suggested that these cells have a direct parasiticidal effect. Newborn larvae of Trichinella spiralis can be killed within 20 min by incubation at room temperature in a cell-free system, including purified human eosinophil peroxidase (EPO), H2O2 and chloride at pH 5.5. Killing was measured by microscopic observation of the larvae. The larvicidal effect was dependent on each component of the EPO/H2O2/Cl- system and could be prevented by using SO4(2-) instead

Ethylene formation by polymorphonuclear leukocytes. Role of myeloperoxidase.

Ethylene formation from the thioethers, beta-methylthiopropionaldehyde (methional) and 2-keto-4-thiomethylbutyric acid by phagocytosing polymorphonuclear leukocytes (PMNs) was found to be largely dependent on myeloperoxidase (MPO). Conversion was less than 10% of normal when MPO-deficient PMNs were employed; formation by normal PMNs was inhibited by the peroxidase inhibitors, azide, and cyanide, and a model system consisting of MPO, H2O2, chloride (or bromide) and EDTA was found which shared many of the properties of the predominant PMN system. MPO-independent mechanisms of ethylene formation

Comparison of Morphological Signs of Neurodegenerative Changes and Expression of Neuroprotective Factors in Adult and Aged Wistar Rats in AlCl3-Induced Neurodegeneration.

Aluminum ions are an immunostimulating agent and a promoter of the production of ROS that play a key role in the pathogenesis of age-related diseases. The severity of morphological signs of neurodegeneration caused by chronic intake of AlCl3 aqueous solution (100 mg/kg, 60 days) and mRNA expression of genes encoding the neuroprotective factor BDNF and the antioxidant enzyme GPX4 were compared in adult and aged male Wistar rats. In comparison with the adult control rats, aged animals showed a more pronounced immunohistochemical reaction with antibodies to tau protein (pTau) in the entorhinal an

Electrochemical immunosensor for interferon-γ based on disposable ITO detector and HRP-antibody-conjugated nano gold as signal tag.

Tuberculosis is the most frequent cause of infection-related death worldwide. A new disposable electrochemical immunosensor with low cost and simple fabrication was proposed to detect interferon-γ (IFN-γ). Diallyldimethylammonium chloride (PDDA) and Au nanoparticle (AuNP) composite were used to provide an efficient biointerface, horseradish peroxidase (HRP)-labeled antibody-conjugated AuNP (HRP-Ab2-AuNP) bioconjugates were used as a novel signal tag. The large amounts of HRP on the signal tag can catalyze the oxidation of Hydroquinone (HQ) by H2O2, which can induce an amplified reductive curre

Immunocytochemical localization of metallothionein in the rat prostate gland.

Metallothionein (MT), which binds zinc and other metals, was localized within the rat prostatic complex at light and electron microscopic levels utilizing immunocytochemistry. Two groups of mature, male Sprague-Dawley rats were injected with cadmium chloride subcutaneously (2 mg/kg body weight) to induce the synthesis of MT, and were sacrificed 48 h postinjection. From the first group, prostatic tissue from the three lobes was prepared for light microscopy. The indirect peroxidase antiperoxidase procedure was used for MT localization using rabbit anti-MT as the primary antibody. The three lobe

Mechanisms of tumour cell destruction by PMA-activated human neutrophils.

Phorbol myristate acetate (PMA)-activated neutrophils were found to destroy B lymphoblast tumour cells (Raji) as determined by the 51Cr release assay. The target cell lysis was prevented by azide, suggesting the involvement of the myeloperoxidase enzyme. Catalase and cytochrome c caused a marked impairment of the neutrophil-mediated cytolysis, whereas superoxide dismutase significantly enhanced the target cell destruction. These data indicate that hydrogen peroxide plays a key role in the target cell injury; superoxide anion appears to be devoid of direct cytotoxic activity, despite its requir

Effects of pH and thiocyanate on hydrogen peroxide-induced evolution of molecular oxygen in human mixed saliva.

Hydrogen peroxide-induced evolution of molecular oxygen was measured with a Clark-type electrode in a buffered reaction mixture containing mixed whole or dialysed saliva. The optimum pH for oxygen evolution in mixed whole saliva was around 8. Oxygen evolution was also observed in dialysed saliva, suggesting that free SCN- is not essential. The optimum pH was around pH 6. Sodium thiocyanate inhibited the oxygen evolution under acidic conditions in dialysed saliva, increasing the K(m) of hydrogen peroxide and decreasing the Vmax. Ferric chloride (1 mM), a chelator of SCN-, also inhibited oxygen

Purification and properties of anionic glutathione S-transferase from bovine ciliary body.

An anionic glutathione S-transferase was purified from bovine ciliary body by DEAE-agarose chromatography and affinity chromatography on GSH-agarose and Orange A. The enzyme accounts for about 25% of total soluble glutathione S-transferase activity of the tissue. The purified enzyme has a molecular mass of about 50,000 Da and is composed of two identical subunits of about 25,000 Da. The enzyme has a pI of 5.8. The enzyme conjugates GSH with 1-chloro-2,4-dinitrobenzene, p-nitrobenzyl chloride, 1,2-epoxy-3-(p-nitrophenyl)propane, 1,2-dinitrobenzene and 3,4-dinitrobenzoic acid. The Km values for

The Effects of Co-administration of Lithium and Vitamin E on Hepatic Oxidative Status in Aged Male and Female Rats: Biochemical and Histomorphological Evaluations.

Lithium serves as an effective mood stabilizer for bipolar disorder, especially in older adults, but high doses or prolonged use may lead to toxicity. Here, we assessed the hepatic toxicity of lithium chloride (LiCl) in aged male and female rats and examined whether vitamin E as an antioxidant can mitigate lithium's toxic effects. Forty-eight Wistar rats aged 16-18 months were divided into four equal groups for each sex (n = 6 each): a placebo group receiving vehicle (almond oil), a vitamin E group receiving 100 mg/kg of vitamin E every other day, a lithium chloride (LiCl) group receiving 50 m

Substrate-dependent metabolomic signatures of myeloperoxidase activity in airway epithelial cells: Implications for early cystic fibrosis lung disease.

Myeloperoxidase (MPO) is released by neutrophils in inflamed tissues. MPO oxidizes chloride, bromide, and thiocyanate to produce hypochlorous acid (HOCl), hypobromous acid (HOBr), and hypothiocyanous acid (HOSCN), respectively. These oxidants are toxic to pathogens, but may also react with host cells to elicit biological activity and potential toxicity. In cystic fibrosis (CF) and related diseases, increased neutrophil inflammation leads to increased airway MPO and airway epithelial cell (AEC) exposure to its oxidants. In this study, we investigated how equal dose-rate exposures of MPO-derived
