Automated cation exchange microcolumn chromatography of haemoglobins has been modified for the analysis of haemoglobin A2. It provides the quantitative data of sufficient precision and specificity for the investigation of potential heterozygotes for beta-thalassaemia. Results have been compared with an established method of electrophoresis followed by densitometry of the eluted bands.

Urine and serum myoglobin have been separated on an anion-exchange column, packed by the slurry technique. Urine or serum was injected directly into the column and eluted isocratically with Tris buffer. Freshly prepared myoglobin from human muscle gives two peaks in the chromatogram. Upon storage, one peak slowly decreases while the other increases in size and has the same capacity factor as horse myoglobin. A thin-layer chromatographic method for the detection of sugars is described. The method can also be adapted for the use in biochemistry, pharmacology, and food chemistry. The solvent systems are: I, benzene/1-butanol/acetone/water (20 : 30 : 40 : 10, v/v); II, 1,2-dichloroethane/glacial acetic acid/methanol/water (50 : 25 : 15 : 10, v/v); III, boric acid 0.5%/1-butanol/2-propanol (20 : 30 : 50, v/v). Detection is developed by a diphenylamine aniline phosphoric acid reagent. Additionally an enzymatic method for the detection of some sugars is described.

This paper reviews the applications of high-pressure liquid chromatography (HPLC) to forensic problems, and discusses some of the developments that have taken place in the use of the technique in the Metropolitan Police Laboratory. Preparation of octadecyltrichlorosilane-modified silica is described and some of the chromatographic characteristics of this material are investigated. Applications of HPLC to the analysis of cannabis, opium alkaloids, amphetamine-related materials, LSD and polynuclear hydrocarbons are described.

Quantitative haemoglobin A(2) (HbA(2)) levels are used for the diagnosis of beta-thalassaemia. Preparation of hybrid nanocompositional chitosan/silica sorbent was carried out. It was shown that formation of gel in sol-gel process of hydrolytic polycondensation of tetraethoxysilane (TEOS) with including of chitosan consists of two stages. Suppression of crystallization of chitosan in obtained two-phase system and changes in IR spectra are evidenced of interactions between molecules of chitosan and silanol groups of silica network. The resulting hybrid chitosan/silica sorbent was tested by high-performance liquid chromatography (HPLC).
