Changes in the quantity of collagen type I in women with genuine stress incontinence.

The aim of this study, was to examine changes in the quantity of collagen type I in the pubocervical fascia of women with genuine stress incontinence (GSI), with and without pelvic relaxation. Seventy-eight patients participated in the study and they were divided into three groups that were comparable with respect to their age and parity. All the patients underwent filling cystometry and patients with detrusor instability were excluded from the study. Biopsies were obtained from the pubocervical fascia. The presence of collagen type I was determined with an immunohistochemical technique. The X…

Morphometric analysis of type I collagen fibrils in the osteoid of osteogenesis imperfecta.

Electron microscopy and morphometric measurements of bone osteoid collagen diameter from 42 osteogenesis imperfecta (OI) patients and 25 age- and site-matched controls were carried out. Although the mean diameter did not correlate well with the severity of the disease, it related well with the clinical types and revealed collagen fibrils of reduced diameter in the osteoid of all OI types. Thus, OI type II (the severest type) demonstrated the smallest diameter (45 nm), followed by OI type I (the mildest form) with a mean diameter of 57 nm. The diameter obtained for type III (67 nm) and type IV

Comparison of type I, type III and type VI collagen binding assays in diagnosis of von Willebrand disease.

BACKGROUND: von Willebrand factor (VWF) plays a key role in coagulation by tethering platelets to injured subendothelium through binding sites for collagen and platelet GPIb. Collagen binding assays (VWF:CB), however, are not part of the routine work-up for von Willebrand disease (VWD). OBJECTIVES: This study presents data on collagen binding for healthy controls and VWD subjects to compare three different collagens. PATIENTS/METHODS: VWF antigen (VWF:Ag), VWF ristocetin cofactor activity and VWF:CB with types I, III and VI collagen were examined for samples obtained from the Zimmerman Progr

Demonstration of antibodies to denatured type I and type II collagen in juvenile rheumatoid arthritis, Still's syndrome and controls by [14C] collagen radioimmunoassay.

From 88 sera of patients with juvenile rheumatoid arthritis 30.6% showed antibodies to denatured type I collagen an 31.8% antibodies to type II collagen, a percentage which corresponds to radioimmunoassay results in adult RA. Rheumatoid factors were demonstrated with Waaler-Rose test in 14.7% and with latex test in 6.8% of investigated patients. Results of investigations with collagen types I and II correlated in 73.7%. Whilst type I collagen antibodies were found with equal frequency in active and non-active stages, type II antibodies were twice as frequent in active stages as in non-active s

Effect of praziquantel on the distribution of interstitial collagen types I and III and basement membrane collagen types IV and V in murine hepatic schistosomiasis.

The effect of specific chemotherapy (praziquantel) on liver function tests and on the distribution of collagen types I, III, IV and V was studied by indirect immunofluorescence in Swiss albino mice infected with Schistosoma mansoni. Treatment was started at 7 and 12 weeks after infection. Groups of treated and non-treated mice were killed 14 and 20 weeks after infection. Reduction in the amount of collagen and improvement of liver function were observed, especially when treatment was initiated early (7 weeks after infection), while collagen type III almost disappeared during the period of obse…

Intractable vasculitis, resorptive osteolysis, and immunity to type I collagen in type VIII Ehlers-Danlos syndrome.

A unique patient with type VIII Ehlers-Danlos syndrome and cutaneous vasculitis, resorptive osteolysis, and cardiac valvular disease is described. Collagen analyses identified morphologic and physical abnormalities of type I collagen. The patient's T lymphocytes could be propagated in vitro with type I collagen and produced a 60-kd lymphokine that bound this protein. Cellular autoimmunity to type I collagen may be responsible for this patient's intractable clinical condition.

Type I and type III collagen synthesis and composition in the valve matrix in aortic valve stenosis.

Changes in the collagenous matrix may contribute to the pathogenesis and progression of human aortic valve stenosis (AS). To evaluate the significance of collagen I and III in the pathogenesis of AS, we studied their synthesis in diseased valves. Type I and type III collagen mRNA expression and the immunohistochemical localization of the collagen antigens were studied from 36 AS and 2 normal aortic valves. The concentrations of propeptides and telopeptide structure of type I (PINP, PICP, and ICTP) and those of III collagens (PIIINP and IIINTP) were measured by radioimmunoassays in soluble tiss

Levels of type I and type III collagen in the bile duct of rats infected with Fasciola hepatica.

Collagen types and their levels were compared between bile ducts from Fasciola infected rats and bile ducts from uninfected animals. Both collagen types I and III were shown to be increased in infected animals but, levels of type I increased less than type III. These results indicate that fascioliasis produces changes in the collagen composition of the bile duct that are similar to those produced in cirrhosis of the liver and other pathologic conditions including wound healing. Such observations suggest that a study of the chronology of collagen deposition in fascioliasis might provide informa

Effects of oral administration of tripeptides derived from type I collagen (collagen tripeptide) on atherosclerosis development in hypercholesterolemic rabbits.

Digestion of type I collagen with a collagenase-type protease yields a collagen tripeptide (Ctp) fraction comprising Gly-X-Y sequences that exhibit diverse biological activities. We previously demonstrated that Ctp inhibits the proliferation and migration of cultured aortic smooth muscle cells (SMCs) in vitro. These cells contribute to the pathogenesis of atherosclerosis and other cardiovascular diseases. In order to evaluate the effects of Ctp on atherosclerosis development in vivo, here we used the Kurosawa and Kusanagi-hypercholesterolemic (KHC) rabbit model of familial hypercholesterolemia

Microsphere System Coloading Type I Collagen and Dexamethasone Sodium Phosphate for Osteoarthritis Therapy.

Knee osteoarthritis (KOA) has no effective nonsurgical treatment. Intra-articular therapy constitutes an effective localized intervention; however, persistent lymphatic drainage and synovial vascular exchange lead to rapid drug elimination. Encapsulating bioactive agents into microsphere delivery platforms enables localized intra-articular administration, with their scaffold-based spherical architecture serving as pharmaceutical and cellular carriers or tissue engineering scaffolds, representing a highly promising therapeutic avenue for KOA. Herein, PLGA was employed to prepare microspheres an

Urinary collagen type I degradation products as common fibrosis biomarkers in chronic diseases.

BACKGROUND: Fibrosis, characterised by excessive accumulation of collagen type I (COL1), is a common feature of chronic diseases, including liver diseases (LDs), chronic kidney disease (CKD) and heart failure (HF). COL1 degradation products can be detected in urine by proteomics/ peptidomics analyses and may serve as non-invasive biomarkers of fibrosis. We aimed to identify a common molecular signature of fibrosis across these diseases that may ultimately guide interventions to slow disease progression and prevent organ damage. METHODS: Using capillary electrophoresis coupled to mass spectrom…

The type I collagen paradox in PDAC progression: microenvironmental protector turned tumor accomplice.

Type I collagen (Collagen I, Col I), a principal component of the tumor extracellular matrix (ECM) that accounts for 80% of total ECM collagen, has emerged as a crucial factor in tumor biology. Recent research has found that Col I has influenced tumor growth, invasion, migration, and prognosis by forming a physical barrier and interacting with components in the tumor microenvironment (TME). Pancreatic ductal adenocarcinoma (PDAC) is characterized by a prominent stromal microenvironment with remarkable cellular and spatial heterogeneity, which significantly impacts the biology of the disease an

Electron Beam-Modified Collagen Type I Fibers: Synthesis and Characterization of Mechanical Response.

Ten MeV electron beam treatment facilitates a biomimetic introduction of cross-links in collagenous biopolymer systems, modifying their viscoelastic properties, mechanical stability, and swelling behavior. For reconstituted collagen type I fibers, electron-induced cross-linking opens up new perspectives regarding future biomedical applications in terms of tissue and ligament engineering. We demonstrate how electron irradiation affects stiffness both in low-strain regimes and in postyield regimes of biocompatible reconstituted rat tail collagen type I fibers. Stress-strain tests show a dose-dep…

Cyclophilin B control of lysine post-translational modifications of skin type I collagen.

Covalent intermolecular cross-linking of collagen is essential for tissue stability. Recent studies have demonstrated that cyclophilin B (CypB), an endoplasmic reticulum (ER)-resident peptidyl-prolyl cis-trans isomerase, modulates lysine (Lys) hydroxylation of type I collagen impacting cross-linking chemistry. However, the extent of modulation, the molecular mechanism and the functional outcome in tissues are not well understood. Here, we report that, in CypB null (KO) mouse skin, two unusual collagen cross-links lacking Lys hydroxylation are formed while neither was detected in wild type (WT)

Lysophosphatidic acid mediates fibrosis in injured joints by regulating collagen type I biosynthesis.

OBJECTIVE: Articular cartilage is a highly specialized tissue which forms the surfaces in synovial joints. Full-thickness cartilage defects caused by trauma or microfracture surgery heal via the formation of fibrotic tissue characterized by a high content of collagen I (COL I) and subsequent poor mechanical properties. The goal of this study is to investigate the molecular mechanisms underlying fibrosis after joint injury. DESIGN: Rat knee joint models were used to mimic cartilage defects after acute injury. Immunohistochemistry was performed to detect proteins related to fibrosis. Human feta

Lysyl hydroxylase 3 glucosylates galactosylhydroxylysine residues in type I collagen in osteoblast culture.

Lysyl hydroxylase 3 (LH3), encoded by Plod3, is the multifunctional collagen-modifying enzyme possessing LH, hydroxylysine galactosyltransferase (GT), and galactosylhydroxylysine-glucosyltransferase (GGT) activities. Although an alteration in type I collagen glycosylation has been implicated in several osteogenic disorders, the role of LH3 in bone physiology has never been investigated. To elucidate the function of LH3 in bone type I collagen modifications, we used a short hairpin RNA technology in a mouse osteoblastic cell line, MC3T3-E1; generated single cell-derived clones stably suppressin
