Granulocyte macrophage colony-stimulating factor in adenomyosis and autologous endometrium.

OBJECTIVE: Granulocyte macrophage colony-stimulating factor (GM-CSF) has been related to macrophage recruitment and activation and has been identified in the human endometrium. We determined whether adenomyosis expresses GM-CSF, and if present, compared GM-CSF protein expression in adenomyosis with that in autologous endometrium. METHODS: We examined ectopic and eutopic endometrium from 16 premenopausal women who had hysterectomies for abnormal uterine bleeding, pelvic pain, or uterine prolapse. Serial sections of premenopausal uteri containing endometrium and adenomyosis were analyzed by imm

Serum levels of macrophage colony-stimulating factor in trophoblastic disease.

OBJECTIVES: The aims of this study were to measure levels of colony stimulating factor (CSF-1) in patients with trophoblastic disease, to determine whether such measurement may be useful to supplement measurement of the prognostically reliable human chorionic gonadotrophin (hCG), and to assess whether measurement of CSF-1 may be helpful in predicting requirement for chemotherapy in patients with hydatidiform mole. METHODS: Serial weekly serum samples were selected for CSF-1 assay from representative diagnostic groups of patients with trophoblastic disease: hydatidiform-mole with spontaneous r

Use of macrophage colony-stimulating factor in the treatment of fungal infections.

Management of fungal infections is a major medical problem. The risk of developing a fungal infection is higher for patients who are undergoing dose-intensive therapy, are immunocompromised, have neutropenia, are receiving prophylactic antibiotics, have other infections, have invasive catheters, or have a history of severe trauma or burns. Survival is decreased among patients who develop fungal infection in these situations. In view of the high morbidity and mortality associated with fungal infections in transplant recipients, cytokines that enhance cell function, such as macrophage colony-sti

Granulocyte colony stimulating factor therapy of experimental cryptococcal meningitis.

Cryptococcal meningitis was induced in mice using intracerebral injection of Cryptococcus neoformans. Beginning either 3 days before or 1 day after infection, mice were treated with human recombinant granulocyte colony stimulating factor (hGCSF). In high doses hGCSF reduced the brain tissue burden of C. neoformans but had no effect on survival. The effect of hGCSF was dependent on size of the infecting dose and time of administration. A large innocula of C. neoformans, or when hGCSF was initiated after infection, there was no added benefit. Some groups of mice also received low doses of flucon

New clinical applications of granulocyte-macrophage colony-stimulating factor.

The cytokine granulocyte-macrophage colony stimulating factor (GM-CSF) has a pleiotropic activity that influences not only proliferation and differentiation of stem cells but also regulates several other cells involved in acute and chronic inflammation, as well as in the immune response. During the past 5 years, investigations have been performed to treat pathological situations rather than cytostatic induced neutropenia. This review presents the available experience on healing cut-, burn-wounds, leg ulcers, skin-graft resolution as well as cytostatic and radiotherapy induced mucositis by admi

A generalized cutaneous reaction induced by granulocyte colony-stimulating factor.

BACKGROUND: The increasing use of recombinant forms of granulocyte and granulocyte-macrophage colony-stimulating factors (GCSF and GMCSF) for neutropenic conditions has resulted in reports of a variety of cutaneous reactions. OBJECTIVE: We studied the clinical and histologic findings in three patients who underwent bone marrow transplantation and subsequently had a cutaneous eruption associated with the use of GCSF. METHODS: Biopsy specimens taken at the height of the eruption were studied histologically and immunohistochemically. RESULTS: The patients had indurated, well-demarcated, occasi

A prolonged use of granulocyte colony stimulating factor in Felty's syndrome.

Current treatment for Felty's syndrome, a triad of rheumatoid arthritis, splenomegaly and neutropenia, is often unsuccessful. Felty's syndrome may be related to decreased production of hematopoietic growth factors. We treated a patient with Felty's syndrome, profound neutropenia and a history of multiple complicated hospitalization for severe infections, with granulocyte colony stimulating factor (GCSF) for 18 months. After initiation of GCSF, the patient's neutrophil count has remained in the normal range for 18 months. After 2 easily treated infections at the start of therapy, she had only o

Colony-stimulating factor 1 in the human response to neonatal listeriosis.

Immunity to Listeria monocytogenes, an important pathogen in human neonatal sepsis, is mediated by mononuclear phagocytes, which are regulated by the hematopoietic growth factor colony-stimulating factor 1. Neonates with listeriosis had higher circulating colony-stimulating factor 1 levels and subsequent monocyte counts than those of both noninfected newborns and newborns infected with nonlisterial organisms.

Granulocyte-colony stimulating factor therapy in drug-induced agranulocytosis.

Drug-induced agranulocytosis in the outpatient setting is a rare but potentially fatal adverse effect of many classes of medications. Five patients with this disorder presented to Yale-New Haven (Conn) Hospital during 1990 through 1992. Three patients treated with granulocyte-colony stimulating factor and two patients treated with routine care were studied for relevant clinical outcomes. Treatment with granulocyte-colony stimulating factor was associated with a shorter duration of neutropenia and a decreased length of hospital stay, consistent with recent case reports. Despite the high cost of

Expression of the colony-stimulating factor 1 receptor in B lymphocytes.

The FMS proto-oncogene encodes for the colony-stimulating factor 1 receptor (CSF-1R), whose expression within the haematopoietic system has previously been thought to be restricted to cells of the mononuclear phagocyte lineage. We have studied the expression of the CSF-1R in peripheral blood mononuclear cells by indirect immunofluorescence and flow cytometry. FMS expression was detected on both monocytes and B lymphocytes from all samples analysed, including 14 haematologically normal individuals and 31 patients (23 in remission following cytotoxic therapy for lymphoma, six with B-cell chronic

Case report: role of granulocyte colony stimulating factor in radiotherapy.

Two patients who received granulocyte colony stimulating factor (G-CSF) before or during radiotherapy are reviewed. Both patients developed granulocytopenia after multi-agent chemotherapy. One patient had breast cancer with multiple distant metastasis and received palliative radiotherapy to the spine and hip joint. The other patient had locally advanced anal carcinoma and received concurrent 5-fluorouracil, mitomycin, and radiation therapy. Both patients completed the planned course of radiotherapy without interruption, after administration of G-CSF.

Structural analysis of the receptors for granulocyte colony-stimulating factor on neutrophils.

We investigated granulocyte colony-stimulating factor (G-CSF) receptors on neutrophils from three patients with chronic myelogenous leukemia (CML) in the chronic phase, in comparison with four normal volunteers. Because we experienced some difficulties in radioiodinating intact recombinant human G-CSF, we developed a new derivative of human G-CSF termed YPY-G-CSF. It was easy to iodinate this protein using the lactoperoxidase method because of two additional tyrosine residues, and its radioactivity was higher than that previously reported. The biological activity of YPY-G-CSF as G-CSF was full

Induction of monocyte migration by recombinant macrophage colony-stimulating factor.

Human recombinant macrophage-CSF (M-CSF) induced migration across polycarbonate or nitrocellulose filters of human peripheral blood monocytes. Checkerboard analysis of M-CSF-induced migration, performed by seeding different cytokine concentrations above and below the filter, revealed that the locomotory response involved chemotaxis, though some gradient-independent augmentation of migration occurred. Polymixin B did not affect M-CSF chemotaxis and M-CSF was active on monocytes from the LPS-unresponsive mouse strain C3H/HeJ. These findings rule out a contribution of minute endotoxin contaminati

Human urinary megakaryocyte colony-stimulating factor in thrombopoietic disorders.

The level of urinary Meg-CSF activity in patients with various thrombopoietic disorders was studied. Five out of eight patients with idiopathic thrombocytopenic purpura had megakaryocytic hyperplasia in the marrow and increased Meg-CSF activity in the urine. Urinary Meg-CSF activity in patients with polycythaemia vera and essential thrombocythaemia was normal. There was a significant inverse correlation between urinary Meg-CSF activity and peripheral blood platelet count but not bone marrow megakaryocyte mass. There was a significant increase of urinary Meg-CSF activity during the period of th

Androgen-induced production of colony-stimulating factor (CSF) and colony-inhibitory factor (CIF) in the submandibular gland in female mice.

Both testosterone (T) and 5 alpha-dihydrotestosterone (DHT) significantly increased colony-stimulating factor (CSF) as well as colony-inhibitory factor (CIF) in the submandibular gland (SMG) in female mice. The CIF activity was completely inactivated by heating at 60 degrees C for 15 min, while the CSF activity was stable against the heat treatment. It was found that the CSFs obtained from the androgen-treated female mice and from normal male mice behaved identically in chromatography on hydroxylapatite and Ultro-gel AcA 34 columns. The potency of DHT to increase CSF activity was about 10 time

Functional characterization of colony-stimulating factors in cytokine modulation and phagocytic cell activation in Japanese flounder.

Colony-stimulating factors (CSFs), including macrophage CSF (MCSF) and granulocyte CSF (GCSF), play central roles in the regulation and activation of phagocytic cells in vertebrates. However, their functions and regulatory mechanisms in teleosts remain partially understood. In this study, the functional characteristics of MCSF and GCSF were analyzed in Japanese flounder (Paralichthys olivaceus) to examine their tissue distribution, evolutionary relationships, and immunoregulatory roles in peripheral blood leukocytes (PBLs). MCSF and GCSF were highly expressed in the spleen and gill, supporting

Secretion of colony-stimulating factors by T cell clones. Role in adoptive protection against Listeria monocytogenes.

CSF have been postulated to be important mediators of host defenses. The current studies were undertaken to investigate the production of CSF by Listeria-specific, T cell clones and to assess the participation of CSF in anti-listerial host resistance. Listeria-specific L3T4+, Lyt-2- T cell clones were isolated and expanded by standard techniques. The clones themselves protected mice from listerial challenge when injected intravenously, and supernatants generated from Ag-stimulated clones were protective. In order to define factors important in the protection, supernatants from the clones were
