Effect of incubation temperature on the diagnostic sensitivity of the glanders complement fixation test.

The complement fixation test (CFT) is the only serological test prescribed by the World Organisation for Animal Health (OIE) for the diagnosis of glanders in international trading of equids. However, false-positive reactions have caused financial losses to the animal owners in the past, and false-negative tests have resulted in the introduction of glanders into healthy equine populations in previously glanders-free areas. Both warm (incubation at 37°C for 1 h) and cold (overnight incubation at 4°C) procedures are recommended by the OIE for serodiagnosis of glanders. In a comparison of the sens

Heated histoplasmin as a control for non-specificity in the complement fixation test for histoplasmosis.

We present data in support of the use of a heated histoplasmin control for the complement fixation (CF) test for histoplasmosis to assist in the detection of the presence of h or m antibody.

Detection of arbovirus antibodies in avian sera by the complement fixation-inhibition test.

The complement fixation-inhibition (CFI) test was described for the detection of antibodies to arboviruses in bird sera. The CFI antibody present in bird sera inhibited the standard complement-fixation reaction of a reference complement-fixing antigen-antibody pair. Using reference antigens (St. Louis encephalitis, eastern equine encephalomyelitis, western equine encephalomyelitis, and yellow fever) prepared from infected mouse brains and reference antisera prepared in rabbits or horses, reproducible CFI antibody titers were obtained in artificially immunized chickens. Time-course studies on t

THE COMPLEMENT FIXATION TEST IN THE DIAGNOSIS OF VIRUS INFECTIONS OF THE CENTRAL NERVOUS SYSTEM.

A specific complement fixation test can be obtained in various central nervous system virus infections by using as antigens emulsions of infected brain tissue, freezing and thawing the brain emulsion, and then centrifuging it in an angle head centrifuge at 3500 R.P.M. for 1 hour. The method has proved reliable in the case of rabies, St. Louis encephalitis, Japanese B encephalitis, lymphocytic choriomeningitis, Eastern equine encephalomyelitis, Western equine encephalomyelitis, louping ill, and spontaneous encephalomyelitis of mice (Theiler's disease). The specificity of the reaction, regardles

Comparative evaluation of commercial Premier EIA and microimmunodiffusion and complement fixation tests for Coccidioides immitis antibodies.

A total of 409 serum and cerebrospinal fluid specimens from human subjects with proven coccidioidomycosis, with other infections, or with no apparent illness were tested for antibodies to Coccidioides immitis by the Premier EIA (Meridian Diagnostics, Inc., Cincinnati, Ohio), which tests for immunoglobulin G (IgG) and IgM responses to coccidioidal antigens, and by the conventional complement fixation (CF) or immunodiffusion (ID) assays for antibodies corresponding to those detected by the tube precipitin (TP) or CF tests. Of the 409 specimens, 47 were from persons with confirmed coccidioidomyco

Antibodies and the Aberdeen typhoid outbreak of 1964. II. Coombs', complement fixation and fimbrial agglutination tests.

In a previous paper (Brodie, 1977) the value of the Widal test in the diagnosis of typhoid fever was shown to be limited. Evaluation of possible alternative tests showed that: (1) the sensitivity of the anti-human globulin (Coombs') test appeared greater than that of the agglutination test but the length of time (48 h) before results were available rendered it of little value in rapid diagnosis: (2) the complement fixation test offered no particular help towards diagnosis; (3) immunized and non-immunized typhoid fever patients developed fimbrial antibodies, as also did immunized healthy indivi

Serologic response of Babesia equi-infected horses as measured by complement-fixation and indirect fluorescent antibody tests.

Both the complement-fixation test (CFT) and the indirect fluorescent antibody test (IFAT) were conducted on weekly serum samples from nine Arab geldings for 28 days before and 256 days after their exposure to Babesia equi of European origin. On an average the IFAT became positive 8 days before the CFT and showed higher relative serum titer increases. Both test procedures successfully detected infection and neither showed an appreciable drop in titer during this time frame, with the exception of the CFT, which showed a transient drop immediately following treatment with imidocarb. A test conduc

Evaluation of the complement fixation and indirect immunofluorescence tests in the early diagnosis of primary Q fever.

A comparison was made of the performance of the commonly used complement fixation test and the more recently developed indirect immunofluorescence test in the early diagnosis of Q fever. The 303 sera tested were from 181 patients who contracted Q fever during an outbreak in Switzerland in 1983. Specific IgM antibodies were detected by the immunofluorescence test in 53% and 89% of sera obtained during the first and second week respectively after onset of illness. With the complement fixation test, the diagnosis could not be made until the second week of illness. The immunofluorescence test prov
