Temporal and spatial characterization of nicotinic acetylcholine receptor expression in Lepeophtheirus salmonis.

Nicotinic acetylcholine receptors (nAChRs) are key mediators of synaptic transmission in invertebrates and major targets of pesticide action. In early life stages of marine Copepoda, such as the parasitic salmon louse (Lepeophtheirus salmonis), the expression dynamics and tissue localization of nAChRs remain poorly characterized. Here, we investigate the expression of nAChR-related transcripts across the life cycle of L. salmonis, and spatial distribution of expression in the early copepodid life stage. Using RNA sequencing of pooled specimens from eleven groups (nauplius I to adults), we iden

Metabolization of emamectin benzoate into desmethyl emamectin benzoate in spiked marine sediments.

Emamectin benzoate (EMB) (4″deoxy- 4″-epi-methylaminoavermectin) is a pesticide developed to control pests on various crops, and in forestry. It is also used in salmon aquaculture to control sea lice infestations as an in-feed therapeutant. Little is known about EMB metabolites and potential associated toxicities in marine sediments. In this study, we used natural marine sediments (sand and mud) fortified at an EMB concentration of 60 ppb (wet weight). Results show an almost immediate transformation of EMB to Desmethyl EMB (DES) with no increased rates of metabolization when stored sediment sa

First record of association of copepods with highly venomous box jellyfish Chironex, with description of new species of Paramacrochiron (Cyclopoida: Macrochironidae).

Paramacrochiron chironecicola n. sp. (Copepoda: Cyclopoida: Macrochironidae) is described from the highly venomous box jellyfish Chironex sp. collected from Malampaya Sound, Palawan Island, The Philippines. This is the first record of copepods associated with cubozoan medusae, although other cnidarian groups such scyphozoans, hydrozoans, and anthozoans are common hosts for symbiotic copepods. The infection sites were on the subumbrella, pedalium, and rhopalium, but also rarely on the adradial furrow. The new species is distinguished from other congeners by a combination of the following featur

Novel gene isolated from Caligus rogercresseyi: a promising target for vaccine development against sea lice.

Sea lice (Copepoda, Caligidae) are the most widely distributed marine pathogens in the salmon industry in the last 30 years. Caligus rogercresseyi is the most important species affecting Chile's salmon industry. Vaccines against caligid copepods have the potential to be a cost-effective means of controlling the infestation and avoid many of the disadvantages of medicine treatments. However, research in the development of such vaccines has begun only recently and approaches used thus far have met with little or no success. In the present study, we characterized a novel gene (denoted as my32) fr

Immunoprophylactic potential of parasite-derived antigens for controlling Lernaea cyprinacea infection in Ctenopharyngodon idella.

Lernaea cyprinacea, an invasive ectoparasitic copepod, poses a significant threat to freshwater aquaculture due to its high pathogenicity and association with severe epizootics leading to mass fish mortalities. The present study evaluated the immunogenic potential of parasite-derived antigens in Ctenopharyngodon idella (grass carp) as a immunoprophylactic strategy against L. cyprinacea infection. Crude antigens isolated from L. cyprinacea were administered intraperitoneally to the experimental group, whereas the control group received phosphate-buffered saline (PBS), and a naturally infected g

First evidence of the induction of domoic acid production in Pseudo-nitzschia australis by the copepod Temora longicornis from the French coast.

Diatoms of the genus Pseudo-nitzschia are widespread in marine waters. Some of them can produce the toxin domoic acid (DA) which can be responsible for amnesic shellfish poisoning (ASP) when transferred into the food web. These ASP events are of major concern, due to their ecological and socio-economic repercussions, particularly on the shellfish industry. Many studies have focused on the influence of abiotic factors on DA induction, less on the role of biotic interactions. Recently, the presence of predators has been shown to increase DA production in several Pseudo-nitzschia species, in part

Secretion of extracellular vesicles during ontogeny of the tapeworm Schistocephalus solidus.

We provide the first ultrastructural evidence of the secretion of extracellular vesicles (EVs) across all parasitic stages of the tapeworm Schistocephalus solidus (Müller, 1776) (Cestoda: Diphyllobothriidea) using a laboratory life cycle model. We confirmed the presence of EV-like bodies in all stages examined, including the hexacanth, procercoids in the copepod, Macrocyclops albidus (Jurine, 1820), plerocercoids from the body cavity of the three-spined stickleback, Gasterosteus aculeatus Linnaeus, and adults cultivated in artificial medium. In addition, we provide description of novel tegumen

Effect of cleaner fish on sea lice in Norwegian salmon aquaculture: a national scale data analysis.

The salmon aquaculture industry has adopted the use of invertivorous 'cleaner fishes' (CF) for biological control of sea louse infestations on farmed salmon. At present, ~50 million CF are used annually in Norway alone, with variable success in experimental and industrial contexts. We used a national scale database of louse counts, delousing treatments and CF stocking events on Norwegian salmon farms to test for evidence of CF efficacy at 488 sites that completed a grow-out cycle within 2016-2018. Our analysis revealed that sites using more CF over the duration of a grow-out cycle did not have

Modelling sea lice control by lumpfish on Atlantic salmon farms: interactions with mate limitation, temperature and treatment rules.

Atlantic salmon farming is one of the largest aquaculture sectors in the world. A major impact on farm economics, fish welfare and, potentially, nearby wild salmonid populations, is the sea louse ectoparasite Lepeophtheirus salmonis. Sea louse infestations are most often controlled through application of chemicals, but in most farming regions, sea lice have evolved resistance to the small set of available chemicals. Therefore, alternative treatment methodologies are becoming more widely used. One increasingly common alternative treatment involves the co-culture of farmed salmon with cleaner fi

Association between sea lice (Lepeophtheirus salmonis) infestation on Atlantic salmon farms and wild Pacific salmon in Muchalat Inlet, Canada.

Growth in salmon aquaculture over the past two decades has raised concerns regarding the potential impacts of the industry on neighboring ecosystems and wild fish productivity. Despite limited evidence, sea lice have been identified as a major cause for the decline in some wild Pacific salmon populations on the west coast of Canada. We used sea lice count and management data from farmed and wild salmon, collected over 10 years (2007-2016) in the Muchalat Inlet region of Canada, to evaluate the association between sea lice recorded on salmon farms with the infestation levels on wild out-migrati

Effect of an Otic Milbemycin Oxime Formulation on Tegastes acroporanus Infesting Corals.

The copepod Tegastes acroporanus is a notorious pest of captive corals in the genus Acropora. In recent years, infestations of T. acroporanus have become widespread among public aquaria and coral propagation facilities and have been largely controlled with the extra-label use of milbemycin oxime formulations (Carl 2008). Many of these drug formulations (which were intended for dogs) have been discontinued by their manufacturers in favor of multidrug products, many of which are unsuitable for corals, forcing experimentation with alternatives. This report provides the first data on populations o

Influence of dissolved organic matter character on mercury incorporation by planktonic organisms: an experimental study using oligotrophic water from Patagonian lakes.

Ligands present in dissolved organic matter (DOM) form complexes with inorganic divalent mercury (Hg2+) affecting its bioavailability in pelagic food webs. This investigation addresses the influence of a natural gradient of DOM present in Patagonian lakes on the bioaccumulation of Hg2+ (the prevailing mercury species in the water column of these lakes) by the algae Cryptomonas erosa and the zooplankters Brachionus calyciflorus and Boeckella antiqua. Hg2+ accumulation was studied through laboratory experiments using natural water of four oligotrophic Patagonian lakes amended witht'97Hg2+. The b

Ectoparasite fauna of the lebranche mullet, Mugil liza, from a coastal lagoon in the Southwestern Atlantic.

Coastal lagoons are among the most productive aquatic ecosystems worldwide and are widely used by mugilid fishes. Mugil liza stands out as one of the main resources of lagoon fisheries in southern and southeastern Brazil. This study aimed to identify the species composing the ectoparasite fauna of M. liza from the Maricá Lagoon, Rio de Janeiro State, Brazil. A total of 44 specimens of M. liza were examined, of which 95.4% were parasitized by at least one ectoparasite species. Six ectoparasite species were recorded. The copepod Ergasilus caraguatatubensis was the most representative species, wi

Salmon lice in the Pacific Ocean show evidence of evolved resistance to parasiticide treatment.

Parasitic salmon lice (Lepeophtheirus salmonis) threaten the economic and ecological sustainability of salmon farming, and their evolved resistance to treatment with emamectin benzoate (EMB) has been a major problem for salmon farming in the Atlantic Ocean. In contrast, the Pacific Ocean, where wild salmon are far more abundant, has not seen widespread evolution of EMB-resistant lice. Here, we use EMB bioassays and counts of lice on farms from the Broughton Archipelago, Canada-a core region of salmon farming in the Pacific-to show that EMB sensitivity has dramatically decreased since 2010, con

Small, charged proteins in salmon louse (Lepeophtheirus salmonis) secretions modulate Atlantic salmon (Salmo salar) immune responses and coagulation.

Little is known about glandular proteins secreted from the skin- and blood-feeding ectoparasite salmon louse (Lepeophtheirus salmonis). The labial gland has ducts extending into the oral cavity of the lice, and the present study aimed to identify novel genes expressed by this gland type and to investigate their role in modulation of host parameters at the lice feeding site. Five genes associated with labial gland function were identified and named Lepeophteirus salmonis labial gland protein (LsLGP) 1-4 and 1 like (LsLGP1L). All LsLGPs were predicted to be small charged secreted proteins not en

Sea lice Lepeophtheirus spinifer, Tuxophorus sp. and Caligus sp. infections on wild-caught queenfish Scomberoides commersonnianus from northern Australia.

Studies of ectoparasites of wild-caught queenfish Scomberoides commersonnianus from several areas in northern Australia were reviewed to investigate relationships between parasite burdens, environmental conditions and external lesions. A sample of 27 queenfish captured near a dredge spoil disposal site in Gladstone Harbour, Queensland, Australia, in January 2012 was anomalous, with a high percentage of fish (66.6%) exhibiting grossly visible skin lesions including foci of erythema and petechial haemorrhages, particularly on the pectoral girdle and ventrolateral surfaces. Coinfection takes its toll: Sea lice override the protective effects of vaccination against a bacterial pathogen in Atlantic salmon.

Vaccination is considered crucial for disease prevention and fish health in the global salmon farming industry. Nevertheless, some aspects, such as the efficacy of vaccines, can be largely circumvented during natural coinfections. Sea lice are ectoparasitic copepods that can occur with a high prevalence in the field, are frequently found in co-infection with other pathogens, and are highly detrimental to fish health. The aim of this case-control study was to evaluate the interaction between the detrimental effects of coinfection and the protective effects of vaccination in fish. We used the in

Projected impacts of 21st century climate change on diapause in Calanus finmarchicus.

Diapause plays a key role in the life cycle of high latitude zooplankton. During diapause, animals avoid starving in winter by living in deep waters where metabolism is lower and met by lipid reserves. Global warming is therefore expected to shorten the maximum potential diapause duration by increasing metabolic rates and by reducing body size and lipid reserves. This will alter the phenology of zooplankton, impact higher trophic levels and disrupt biological carbon pumps. Here, we project the impacts of climate change on the key North Atlantic copepod Calanus finmarchicus under IPCC RCP 8.5.
