Light-Emitting Diode Array with Optical Linear Detector Enables High-Throughput Differential Single-Cell Dielectrophoretic Analysis.

This paper presents a lens-free imaging approach utilizing an array of light sources, capable of measuring the dielectric properties of many particles simultaneously. This method employs coplanar electrodes to induce velocity changes in flowing particles through dielectrophoretic forces, allowing the inference of individual particle properties from differential velocity changes. Both positive and negative forces are detectable. The light source utilized in this system is composed of LEDs with a wavelength of 470 nm, while detection is performed using a 256-element optical array detector. Measu

Nature as blueprint: Global phenotype engineering of CHO production cells based on a multi-omics comparison with plasma cells.

Especially for the production of artificial, difficult to express molecules a further development of the CHO production cell line is required to keep pace with the continuously increasing demands. However, the identification of novel targets for cell line engineering to improve CHO cells is a time and cost intensive process. Since plasma cells are evolutionary optimized for a high antibody expression in mammals, we performed a comprehensive multi-omics comparison between CHO and plasma cells to exploit optimized cellular production traits. Comparing the transcriptome, proteome, miRNome, surfac

Exploring metabolic responses and pathway changes in CHO-K1 cells under varied aeration conditions and copper supplementations using 1 H NMR-based metabolomics.

The optimization of bioprocess for CHO cell culture involves careful consideration of factors such as nutrient consumption, metabolic byproduct accumulation, cell growth, and monoclonal antibody (mAb) production. Valuable insights can be obtained by understanding cellular physiology to ensure robust and efficient bioprocess. This study aims to improve our understanding of the CHO-K1 cell metabolism using 1 H NMR-based metabolomics. Initially, the variations in culture performance and metabolic profiles under varied aeration conditions and copper supplementations were thoroughly examined. Furth

Structural and functional exploration of three newly identified coagulation factor IX mutations in Chinese hemophilia B patients.

AIM: To structurally and functionally characterize three newly identified F9 missense mutations, C268Y, I316F, and G413V, in Chinese hemophilia B patients. METHODS: FIX mutants were expressed in vitro by transient transfection of Chinese hamster ovary (CHO) cells. One-stage activated partial thromboplastin time (APTT) based assay and enzyme-linked immunosorbent assay (ELISA) were used to measure the coagulation activity and antigen level of FIX in conditioned medium. Western blot analysis was also used to evaluate interference of the mutations with synthesis and secretion of FIX. A structural

Enhanced cell growth, production, and mAb quality produced in Chinese hamster ovary-K1 cells by supplementing polyamine in the media.

Polyamines such as putrescine (PUT), spermidine (SPD), and spermine (SPM) are amine group-containing biomolecules that regulate multiple intracellular functions such as proliferation, differentiation, and stress response in mammalian cells. Although these biomolecules can be generated intracellularly, lack of polyamine-synthesizing activity has occasionally been reported in a few mammalian cell lines such as Chinese hamster ovary (CHO)-K1; thus, polyamine supplementation in serum-free media is required to support cell growth and production. In the present study, the effects of biogenic polyami

N-glycosylation modulates enzymatic activity of Trypanosoma congolense trans-sialidase.

Trypanosomes cause the devastating disease trypanosomiasis, in which the action of trans-sialidase (TS) enzymes harbored on their surface is a key virulence factor. TS enzymes are N-glycosylated, but the biological functions of their glycans have remained elusive. In this study, we investigated the influence of N-glycans on the enzymatic activity and structural stability of TconTS1, a recombinant TS from the African parasite Trypanosoma congolense. We expressed the enzyme in Chinese hamster ovary Lec1 cells, which produce high-mannose type N-glycans similar to the TS N-glycosylation pattern in

Synthesis of neutral ether lipid monoalkyl-diacylglycerol by lipid acyltransferases.

In mammals, ether lipids exert a wide spectrum of signaling and structural functions, such as stimulation of immune responses, anti-tumor activities, and enhancement of sperm functions. Abnormal accumulation of monoalkyl-diacylglycerol (MADAG) was found in Wolman's disease, a human genetic disorder defined by a deficiency in lysosomal acid lipase. In the current study, we found that among the nine recombinant human lipid acyltransferases examined, acyl-CoA:diacylglycerol acyltransferase (DGAT)1, DGAT2, acyl-CoA:monoacylglycerol acyltransferase (MGAT)2, MGAT3, acyl-CoA:wax-alcohol acyltransfera

Substrate-induced unfolding of protein disulfide isomerase displaces the cholera toxin A1 subunit from its holotoxin.

To generate a cytopathic effect, the catalytic A1 subunit of cholera toxin (CT) must be separated from the rest of the toxin. Protein disulfide isomerase (PDI) is thought to mediate CT disassembly by acting as a redox-driven chaperone that actively unfolds the CTA1 subunit. Here, we show that PDI itself unfolds upon contact with CTA1. The substrate-induced unfolding of PDI provides a novel molecular mechanism for holotoxin disassembly: we postulate the expanded hydrodynamic radius of unfolded PDI acts as a wedge to dislodge reduced CTA1 from its holotoxin. The oxidoreductase activity of PDI wa

Nanoparticle cytotoxicity depends on intracellular solubility: comparison of stabilized copper metal and degradable copper oxide nanoparticles.

Metal nanoparticles have distinctly different chemical and physical properties than currently investigated oxides. Since pure metallic nanoparticles are igniting at air, carbon stabilized copper nanoparticles were used as representative material for this class. Using copper as a representative example, we compare the cytotoxicity of copper metal nanoparticles stabilized by a carbon layer to copper oxide nanoparticles using two different cell lines. Keeping the copper exposure dose constant, the two forms of copper showed a distinctly different response. Whilst copper oxide had already been rep

Polyclonal antibody-mediated mitotic inhibition in Chinese hamster ovary cells.

The nucleus of a mammalian cell undergoes profound reorganization when the cell enters mitosis and a number of proteins involved at various levels of the cell cycle have been characterized. The presence of mitotic-specific proteins has been reported and their roles are important in understanding the mechanics of cell division. The ability of antibodies to recognize mitotic protein antigens and further inhibit mitosis is potentially valuable in their role as therapeutic and diagnostic agents in cancer therapy. In this study, we have aimed to analyze proteins isolated from mitotic cells of Chine

[Phenomenon of delayed disruption of telomeric links between chromosomes in polykaryocytes formed from human-Chinese hamster hybrid cells].

The clones MOM-8-1 and MOM-8-3 of human--Chinese hamster cell hybrids were used for induction of the phenomenon of delayed disruption of the telomeric links between chromosomes. Colcemide (0.08 microgram/ml) and 5-bromodeoxyuridine (20 micrograms/ml) were present in cell culture for 30 hours. The dicentrics were observed in tetraploid, hypotetraploid and hypodiploid metaphases. No differences between the clones were found. The hybrid cells were the second object in which the phenomenon under discussion was reproduced.

Recombinant erythropoietin expression elevates by ubiquitous chromatin opening element (UCOE) in CHO DG44 cells.

The growing demand for biopharmaceuticals has increased the need for efficient and cost-effective recombinant protein production platforms. Transcriptional gene silencing after random genomic integration of expression vectors remains a major limitation in mammalian expression systems and often reduces protein yield. Incorporation of ubiquitous chromatin opening elements (UCOEs) into expression cassettes has emerged as a promising strategy to mitigate position effects and enhance transgene expression. This study examined the effect of a UCOE-containing expression system on erythropoietin (EPO)

Streamlined stable CHO cell line development using droplet microfluidics with image-based monoclonality assessment.

Rapid generation of monoclonal CHO cell lines remains a key bottleneck in mammalian cell line development (CLD), often requiring months of iterative screening and cloning. Here, we present an integrated droplet microfluidic workflow for high-throughput screening and isolation of antibody-producing ExpiCHO-S cells using a FRET-based secretion assay. The FRET assay was developed and optimized for picodroplet-based screening by evaluating probe configuration, donor-to-acceptor ratio, antibody concentration range, and signal saturation behavior. The platform enables picodroplet-based detection of

Antiapoptosis Engineering for Improved Protein Production from CHO Cells.

Improving the time integral of viable cell concentration by overcoming cell death, namely apoptosis, is one of the most widely used strategies for the efficient production of therapeutic proteins. By establishing stable cell lines that overexpress antiapoptotic genes or downregulate proapoptotic genes, the final product yields can be enhanced as cells become more resistant to environmental stresses. From the selection of high-expressing clones to verification of antiapoptotic activity, the method to construct a stable antiapoptotic cell line is discussed in this chapter.

Assessing the Adjuvant Potential of Chinese Hamster Ovary Host Cell Proteins Using an In Vitro Dendritic Cell Assay.

Host cell proteins (HCPs) are process-related impurities of therapeutic protein production and may affect product quality or patient safety. In clinical trials, certain HCPs (e.g., PLBL2 or CCL2) that co-purify with the therapeutic protein have been associated with immune reactions in patients. In this study, we examined the adjuvant potential of six commonly detected HCPs from CHO cells (PRDX1, S100A4, PLBL2, CCL2, CLU, and YWHAE) using an in vitro dendritic cell (DC) maturation assay. Recombinant HCPs were expressed in CHO cells to mimic manufacturing conditions. PRDX1, S100A4, and PLBL2 cau

Differential polyamine metabolism in CHO cell lines: Insights into cell growth and antibody quality.

Chinese hamster ovary (CHO) cell lines are widely utilized host cell lines in cell culture bioprocessing. Although they originated from a common ancestor, accumulated genetic mutations have led to significant heterogeneity in their behavior under specific conditions. This study investigates the cell line-specific impact of polyamine (PUT; putrescine) withdrawal on the growth, metabolism, and antibody production among three CHO clones derived from different parental cell lines: CHO-K1, CHO-S, and CHO-DG44. CHO-K1 cells strongly depended on external polyamines, showing a 77 % reduction in viable

HPRT Mutation Assay for Chinese Hamster Ovary Cells.

Gene mutations are a hallmark of cancer. Although mutations can occur due to various endogenous and extracellular stress factors daily, detecting them is not straightforward. The HPRT gene is one of the housekeeping genes and is essential for the nucleotide salvage pathway. However, it is not strictly necessary since the de novo pathway can compensate for nucleotides synthesis. By using 6-thioguanine, a cytotoxic purine analog, as a selection agent, we can eliminate HPRT-proficient cells and quantify mutation frequency. Moreover, molecular techniques can provide further analysis of these mutat

A transposon-based transient transfection system in CHO-K1 cells enables quality prediction of stable cell line proteins.

In biologics drug discovery, transient protein expression is widely used to rapidly produce biologics, thereby accelerating the identification of lead candidates. However, the accuracy and consistency of predicting further product quality in large-scale production needs to be considered, especially with respect to physicochemical properties and posttranslational modifications. With this in mind, a transient expression system utilizing Chinese hamster ovary K1 (CHO-K1) has been established, which integrates high expression capability with quality profiles similar to those of the protein produce

Deciphering the determinants of recombinant protein expression across the human secretome.

Protein secretion is an essential process of mammalian cells. In biomanufacturing, this process can be optimized to enhance production yields and biotherapeutic quality. While cell line engineering and bioprocess optimization have yielded high protein titers for some recombinant proteins, many remain difficult to express. Here, we investigated factors influencing protein expression in Chinese hamster ovary (CHO) cells, expressing 2,135 Human Secretome Project proteins. While the abundance of mRNA from recombinant proteins explained less than 1% of observed variation in secretion titers, analys
