Electron-microscopic study of gamma-aminobutyric acid immunoreactivity in the chief cells of the mouse carotid body.

The present study shows gamma-aminobutyric acid (GABA) immunoreactivity in almost all chief cells of the mouse carotid body. GABA immunoreaction products were distributed diffusely in the cytoplasm as well as in the dense-cored granules. These findings suggest that GABA may be localized in the cytoplasm and granules, and may be released from the cells.

Stress granules and organelles: coordinating cellular responses in health and disease.

Membrane-bound organelles and membraneless organelles (MLOs) coordinate various biological processes within eukaryotic cells. Among these, stress granules (SGs) are significant cytoplasmic MLOs that form in response to cellular stress, exhibiting liquid-like properties alongside stable substructures. SGs interact with diverse organelles, thereby influencing cellular pathways that are critical in both health and disease contexts. This review discusses the interplay between SGs and organelles and explores the methodologies employed to analyze interactions between SGs and other MLOs. Furthermore,

Granule changes of human and murine endocrine cells in the gastrointestinal epithelia are characteristic of piecemeal degranulation.

Piecemeal degranulation is a unique pattern of cell secretion that consists of a slow release of granule contents from cytoplasmic secretory granules, which leaves empty chambers that do not fuse with each other or with the plasma membrane. To our knowledge, no cell types other than mast cells, basophils, and eosinophils have been reported in the literature to show morphological features of piecemeal degranulation. In the present study we provide evidence for ultrastructural morphologies characteristic of piecemeal degranulation in entero-endocrine cells of the human and murine gastrointestina

Cytoplasmic components of natural killer cells limit the growth of Cryptococcus neoformans.

Murine natural killer (NK) cell-mediated inhibition of growth of a yeast-like target cell, Cryptococcus neoformans, was completely abrogated by blocking the effector cell secretory process with monensin. Therefore, further studies were performed to determine the ability of various cytoplasmic fractions of NK cells to mediate inhibition of cryptococcal growth. Percoll-fractionated homogenates of rat LGL tumor cells demonstrated that the granule-containing fractions plus three additional sets of less dense cytoplasmic fractions displayed anti-cryptococcal activity; whereas only the cytoplasmic g

Electron microscopic observation of erythema nodosum with the eosinophilic infiltration. A case report.

The presence of eosinophils localized within an inflammatory skin lesion of a patient with erythema nodosum has been studied by electron mic … atous space between the collagen bundles. A few intact eosinophils that showed minor alterations of cytoplasmic granules were also present. The significance of these findings, which may enhance endothelial permeability, is discussed.

Fine structure of an oat cell carcinoma of the lung associated with ectopic ACTH syndrome.

Electron-microscopic examination of an oat-cell carcinoma associated with the ectopic ACTH syndrome demonstrated characteristic cytoplasmic granules of 60-240 nm. diameter, consisting of a dense central core separated by a clear halo from an outer investing membrane. Comparison with previously examined non-secretory oat-cell carcinomas showed the granules to be more numerous in the present case. They are considered to represent secretory activity in the tumour.

Preferential G3BP1-induced condensation of DNA-RNA mixtures from stress granules over RNA or double-stranded DNA.

G3BP1 is a regulator of stress granule assembly capable of driving condensation of RNA in vitro. Recently, we purified stress granule cores from yeast and mammalian cells, and found that in addition to RNA and proteins, they contain large amounts of circular double-stranded DNA (dsDNA). Since G3BP1 has been implicated in DNA-RNA heteroduplex resolution, we compared the ability of G3BP1 to drive condensation of cytoplasmic RNA, circular cytoplasmic dsDNA, and total nucleic acids (comprising both, circular dsDNA and RNA) extracted from purified stress granule cores of HEK293T cells. Imaging unde

Ultrastructure of neurosecretory cells in the pars intercerebralis of Rhodnius prolixus (Hemiptera).

Six neuron types are distinguished in the pars intercerebralis of the starved fifth instar of Rhodnius prolixus. All neuron types contain electron dense secretory granules derived from Golgi complexes which are of characteristic size and morphology in each type. The neuron types are not thought to represent stages in a secretory cycle. The variety of neuron types described is related to that revealed by staining sections of the same cells with paraldehyde fuchsin. Active synthesis of neurosecretory granules continues throughout starvation and the lysosomal system appears to be involved in the

Aging RNA granule dynamics in neurodegeneration.

Disordered RNA-binding proteins and repetitive RNA sequences are the main genetic causes of several neurodegenerative diseases, including amyotrophic lateral sclerosis and Huntington's disease. Importantly, these components also seed the formation of cytoplasmic liquid-like granules, like stress granules and P bodies. Emerging evidence demonstrates that healthy granules formed via liquid-liquid phase separation can mature into solid- or gel-like inclusions that persist within the cell. These solidified inclusions are a precursor to the aggregates identified in patients, demonstrating that dysr

DDX17 is involved in DNA damage repair and modifies FUS toxicity in an RGG-domain dependent manner.

Mutations in the RNA binding protein, Fused in Sarcoma (FUS), lead to amyotrophic lateral sclerosis (ALS), the most frequent form of motor neuron disease. Cytoplasmic aggregation and defective DNA repair machinery are etiologically linked to mutant FUS-associated ALS. Although FUS is involved in numerous aspects of RNA processing, little is understood about the pathophysiological mechanisms of mutant FUS. Here, we employed RNA-sequencing technology in Drosophila brains expressing FUS to identify significantly altered genes and pathways involved in FUS-mediated neurodegeneration. We observed th

Stress granules in the spinal muscular atrophy and amyotrophic lateral sclerosis: The correlation and promising therapy.

Increasing genetic and biochemical evidence has broadened our view of the pathomechanisms that lead to Spinal muscular atrophy (SMA) and Amyotrophic lateral sclerosis (ALS), two fatal neurodegenerative diseases with similar symptoms and causes. Stress granules are dynamic cytosolic storage hubs for mRNAs in response to stress exposures, that are evolutionarily conserved cytoplasmic RNA granules in somatic cells. A lot of previous studies have shown that the impaired stress granules are crucial events in SMA/ALS pathogenesis. In this review, we described the key stress granules related RNA bind

Ultrastructural immunocytochemical characterization of the thyrotroph in rat and human pituitaries.

The beta chain of thyroid stimulating hormone (TSH) was localized in normal rat and human pituitaries with the unlabeled antibody--peroxidase-antiperoxidase complex technique and antisera to rat or bovine TSH beta. Araldite embedded tissues, immunoreactivity of TSH was best preserved after fixation with 1% glutaraldehyde or picric acid formaldehyde. In the TSH cells, the immunocytochemical stain was located on granules. However, there was a variation in staining of individual granules, and among the population of granules. Rat TSH cells were ovoid or angular to stellate, and contained granules

The ultrastructure of epithelioid cells in the avian (Gallus domesticus) carotid body.

10 clinically healthy white leghorn males were utilised for this investigation. Apart from toluidine blue-pyronin-stained Araldite sections, uranyl acetate and lead monoxide staining revealed three types of epithelioid cells. Nuclear and cytoplasmic contents, especially three types of granules, conveniently designated as q, r and p granules, are described in detail and tabulated. Technical difficulties in obtaining carotid body tissue as well as clear-stained sections for ultastructural studies are mentioned.

Electron microscopy elucidates eosinophil degranulation patterns in patients with eosinophilic esophagitis.

BACKGROUND: In patients with eosinophilic esophagitis (EoE), eosinophils accumulate and release granule proteins onto esophageal epithelium. However, little is understood about the mechanism of eosinophil degranulation. OBJECTIVE: To determine and quantify eosinophil degranulation patterns, we studied esophageal biopsy specimens from both the proximal and distal esophagi of 9 randomly selected patients with EoE. METHODS: The specimens were fixed in glutaraldehyde, embedded, sectioned, and imaged by means of transmission electron microscopy. Eosinophils and their granules were identified by t

Intracellular topography of glycine-extended pro-gastrin-processing intermediates in human antral mucosa: an electron-microscopic immunocytochemical study.

To identify and characterize the subcellular topography of glycine-extended pro-gastrin-processing intermediates (G-Gly) in human antral mucosa, we performed an electron microscopic immunocytochemical study using region-specific antisera generated against the synthetic peptide, Tyr-Gly-Trp-Met-Asp-Phe-Gly (GL7), and C-terminal-specific anti-gastrin antisera. As has been previously reported, G-cells contained both electron-dense and electron-lucent granules, with a range of intermediate forms. Gastrin immunoreactivity was demonstrated in almost all granules of each type, whereas anti-GL7 antise

A method for the ultrastructural demonstration of non-specific esterase in human blood and lymphoid tissue.

Using the substrate 2-naphthylthiol acetate (NTA), we developed a reproducible method of demonstrating a non-specific esterase while retaining nuclear and cytoplasmic details at the ultrastructural level. The NTA esterase had a distribution and pattern of staining similar to those of esterases demonstrable at the light microscopic level by the alpha-naphthyl acetate or naphthol AS-D acetate esterase reaction. The NTA esterase appeared as intensely electron-dense granules of varying size and shape in the cytoplasm. The granules were most abundant in the cells of the histiomonocytic series. The
