Effects of chemical straighteners on the hair shaft and scalp.

BACKGROUND: The effects of chemical straighteners on the scalp and hair shaft are not fully known, although such substances are widely used. Hair straightening became popular in Brazil with the use of formaldehyde and its derivatives, despite the prohibition by the current legislation. OBJECTIVE: To identify changes in hair shaft and scalp caused by the use of chemical straighteners. METHODS: A search was performed using keywords in three databases from 03/16/2020 to 05/20/2020, with publications between the years 2000 to 2020. After applying the inclusion and exclusion criteria, 33 articles

Screening and analysis of 940 organic micro-pollutants in river sediments in Vietnam using an automated identification and quantification database system for GC-MS.

In order to obtain a detailed picture of pollution by organic micro-pollutants in Vietnamese rivers, 940 semi-volatile organic compounds in river sediments collected from four major cities were examined by a comprehensive gas chromatography-mass spectrometry-database. The number of detected chemicals at each site ranged from 49 to 158 (median 96 out of 940) with 185 analytes detected at least once in the survey. The substances detected with high frequency (over 80%) and high concentrations were n-alkanes, phthalates, sterols and PAHs. For most substances, sediments from metropolitan areas (Han

Development and validation of a quantitative structure-activity relationship for chronic narcosis to fish.

Vertebrate testing under the European Union's regulation on Registration, Evaluation, Authorisation and Restriction of Chemical substances (REACH) is discouraged, and the use of alternative nontesting approaches such as quantitative structure-activity relationships (QSARs) is encouraged. However, robust QSARs predicting chronic ecotoxicity of organic compounds to fish are not available. The Ecological Structure Activity Relationships (ECOSAR) Class Program is a computerized predictive system that estimates the acute and chronic toxicity of organic compounds for several chemical classes based o

Prioritizing Chemical Candidates from Non-targeted Analysis Using Metadata, Spectral Similarity, and Hazard Scoring within INTERPRET NTA.

New approach methodologies like non-targeted analysis (NTA) are increasingly relevant for identifying and monitoring emerging contaminants. Utilizing high-resolution mass spectrometry (HRMS), NTA methods can detect and annotate chemicals without prior knowledge. Yet, NTA results often have associated uncertainties owing, in part, to a lack of standardized methods. To address this challenge, the US Environmental Protection Agency (US EPA) developed the Interface for Processing, Reviewing, and Translating NTA data ("INTERPRET NTA") to support NTA research within its Office of Research and Develo

Phexpo: a package for bidirectional enrichment analysis of phenotypes and chemicals.

Phenotypes are the result of the complex interplay between environmental and genetic factors. To better understand the interactions between chemical compounds and human phenotypes, and further exposome research we have developed "phexpo," a tool to perform and explore bidirectional chemical and phenotype interactions using enrichment analyses. Phexpo utilizes gene annotations from 2 curated public repositories, the Comparative Toxicogenomics Database and the Human Phenotype Ontology. We have applied phexpo in 3 case studies linking: (1) individual chemicals (a drug, warfarin, and an industrial

Unravelling drug-induced hepatic steatosis: Clinical sub-phenotypes, outcome prediction, and identification of high-concern drugs and hazardous chemical attributes.

BACKGROUND: Drug-induced liver injury (DILI) is a rare but serious adverse drug reaction with a wide range of clinical presentations. While three major DILI phenotypes-hepatocellular, cholestatic, and mixed-are well established, other injury patterns such as steatosis-associated DILI (DIS) are harder to identify and remain underexplored. Existing limited evidence suggests that steatosis is frequent among DILI patients, yet this subtype and its causative drugs have not been systematically characterized in large patient cohorts. GOALS: This study aims to fill this knowledge gap by collecting in

Cysteinome: The first comprehensive database for proteins with targetable cysteine and their covalent inhibitors.

The covalent modification of intrinsically nucleophilic cysteine in proteins is crucial for diverse biochemical events. Bioinformatics approaches may prove useful in the design and discovery of covalent molecules targeting the cysteine in proteins to tune their functions and activities. Herein, we describe the Cysteinome, the first online database that provides a rich resource for the display, search and analysis of structure, function and related annotation for proteins with targetable cysteine as well as their covalent modulators. To this end, Cysteinome compiles 462 proteins with targetable

Correlation of the structures of agricultural fungicides to gene expression in Saccharomyces cerevisiae upon exposure to toxic doses.

Correlations between the chemical structures of agricultural fungicides and mRNA expression levels following exposure of Saccharomyces cerevisiae to toxic doses of thiuram, zineb, maneb, TPN, and PCP were examined. Structurally, thiuram, zineb, and maneb are dithiocarbamate fungicides, whereas TPN and PCP are not. To characterize chemical toxicity, genes expression was classified according to the functional groups used by the MIPS database. However, no correlations between the classification scheme and chemical structures were found. Hierarchical clustering of gene expression profiles was perf

Elucidation of endogenous and exogenous chemicals in maternal serum using high-resolution mass spectrometry.

The increasing exposure to environmental chemicals calls for comprehensive non-targeted analysis to detect unrecognized substances in human samples. We examined human serum samples to classify compounds as endogenous or exogenous using public databases and to explore the relationships between exposure markers and metabolic patterns. Serum samples from 84 pregnant women at 32 weeks gestation were analyzed using LC-QToFMS. Using the PubChemLite for Exposomics database, we annotated and classified 106 compounds (51 endogenous, 55 exogenous). The compound patterns were analyzed using three dimensi

High resolution mass spectrometry based profiling of diet-related deoxyribonucleic acid adducts.

Exposure of DNA to endo- and exogenous DNA binding chemicals can result in the formation of DNA adducts and is believed to be the first step in chemically induced carcinogenesis. DNA adductomics is a relatively new field of research which studies the formation of known and unknown DNA adducts in DNA due to exposure to genotoxic chemicals. In this study, a new UHPLC-HRMS(/MS)-based DNA adduct detection method was developed and validated. Four targeted DNA adducts, which all have been linked to dietary genotoxicity, were included in the described method; O(6)-methylguanine (O(6)-MeG), O(6)-carbo

Developing structure-activity relationships for the prediction of hepatotoxicity.

Drug-induced liver injury is a major issue of concern and has led to the withdrawal of a significant number of marketed drugs. An understanding of structure-activity relationships (SARs) of chemicals can make a significant contribution to the identification of potential toxic effects early in the drug development process and aid in avoiding such problems. This process can be supported by the use of existing toxicity data and mechanistic understanding of the biological processes for related compounds. In the published literature, this information is often spread across diverse sources and can b

Pesticide exposure monitoring databases in applied risk analysis.

Faced with the need to evaluate under what conditions chemicals can be used with "reasonable certainty of no harm" to workers and consumers, industry and government agencies have embraced quantitative risk analysis as a science-based approach for product development, regulatory evaluations, and associated risk management decision making. Beginning in the 1990s, a variety of industry-sponsored task forces have been formed to develop exposure-related data to support safety evaluations for pesticide chemicals used in agricultural, industrial, institutional, residential, and other settings. Human

Chemical Peels in Skin Cancer: A Review.

Background: Chemexfoliation is widely used to reverse signs of photodamage. Although photodamage can eventually lead to skin cancer, it remains unclear whether chemical peels also affect photocarcinogenesis. Moreover, concerns about the systemic and/or cutaneous toxicity of peeling agents have already arisen. Objective: This review sought primarily to summarize the data available on the effects of chemical peels on ultraviolet-induced skin carcinogenesis, focusing particular attention on actinic keratoses and cutaneous field cancerization. In addition, considerations about the systemic and/or

FKBP5 blockade may provide a new horizon for the treatment of stress-associated disorders: An in-silico study.

OBJECTIVE: We searched for, from the FDA (Food and Drug Administration-USA)-approved drugs, inhibitors of FKBP5 with tolerable adverse effect profiles (eg, mild headache, sedation, etc.) and with the ability to cross the blood brain barrier (BBB), using bio-informatics tools (in-silico). This may pave the road for designing clinical trials of such drugs in patients with functional seizures (FS) and other stress-associated disorders. METHODS: Several databases were used to find all the approved drugs that potentially have interactions with FKBP51 protein [ie, CTD gene-chemical interaction sect

Novel clinical phenotypes, drug categorization, and outcome prediction in drug-induced cholestasis: Analysis of a database of 432 patients developed by literature review and machine learning support.

BACKGROUND: Serum transaminases, alkaline phosphatase and bilirubin are common parameters used for DILI diagnosis, classification, and prognosis. However, the relevance of clinical examination, histopathology and drug chemical properties have not been fully investigated. As cholestasis is a frequent and complex DILI manifestation, our goal was to investigate the relevance of clinical features and drug properties to stratify drug-induced cholestasis (DIC) patients, and to develop a prognosis model to identify patients at risk and high-concern drugs. METHODS: DIC-related articles were searched

Chemical profiling and classification of illicit heroin by principal component analysis, calculation of inter sample correlation and artificial neural networks.

Artificial neural networks (ANNs) were utilised to validate illicit drug classification in the profiling method used at "Institut de Police Scientifique" of the University of Lausanne (IPS). This method established links between samples using a combination of principal component analysis (PCA) and calculation of a correlation value between samples. Heroin seizures sent to the IPS laboratory were analysed using gas chromatography (GC) to separate the major alkaloids present in illicit heroin. Statistical analysis was then performed on 3371 samples. Initially, PCA was performed as a preliminary
