Cellular senescence in the dental pulp and its implications for endodontics: a scoping review.

INTRODUCTION: Cellular senescence compromises pulp vitality and may negatively affect vital pulp therapy and regenerative endodontics. Exposure to dental materials may accelerate this process, highlighting the need for safer biomaterials and novel therapeutic strategies. OBJECTIVES: To map and analyze the scientific evidence on cellular senescence in dental pulp, focusing on triggers, biological consequences, and implications for endodontic therapies. MATERIALS AND METHODS: This scoping review was conducted following PRISMA-ScR guidelines (OSF registration: https://doi.org/10.17605/OSF.IO/5ARJ

Pulpal studies: biocompatibility or effectiveness of marginal seal?

Standards presently used to evaluate the biologic compatibility of dental restorative materials are examined and their inherent shortcomings are noted in this report. Studies citing the alleged effects of dental materials on the pulp are reviewed and compared to new theories, which suggest that the principal cause of pulpal inflammation is bacterial infiltration. Thus, studies that are not able to exclude bacteria as a variable cannot be used to draw conclusions as to the cause of inflammatory changes. This paper presents a proposal to alter the current biocompatibility testing methods to meth

Inflammatory root resorption in primary molars: prevalence and associated factors.

This study aimed at determining the prevalence of inflammatory root resorption and associated factors in 1068 primary mandibular molars in 453 children 3 to 12 years of age. Age, dental history and medical history were recorded using a questionnaire administered to the children's parents/caregivers. Previously trained and calibrated examiners assessed radiographic images of the primary molars by direct observation, with the aid of a viewing box. Root resorption (physiological or inflammatory), dental crown status (healthy, carious with no pulp involvement, carious with pulp involvement and evi

Tertiary Dentin Barrier Formation: A Comparison Between the Effects of Two Calcium Silicate Based Materials.

Pulp capping is a vital pulp therapy that aims to prolong the life of a tooth by protecting it after pulp exposure occurred. Pulp capping biomaterials are intended to induce odontoblasts to deposit a natural tertiary dentin barrier to protect the pulp-dentin complex. Two pulp capping agents with calcium silicates in their composition, but with different mechanisms of the setting reaction were tested in vivo with the main objective of comparing their effect on the pulp-dentin complex. The specific aim was to evaluate the preservation of pulp vitality following in vivo direct and indirect pulp c

Cholinergic Nerve Differentiation of Mesenchymal Stem Cells Derived from Long-Term Cryopreserved Human Dental Pulp In Vitro and Analysis of Their Motor Nerve Regeneration Potential In Vivo.

The reduction of choline acetyltransferase, caused by the loss of cholinergic neurons, leads to the absence of acetylcholine (Ach), which is related to motor nerve degeneration. The aims of the present study were to evaluate the in vitro cholinergic nerve differentiation potential of mesenchymal stem cells from cryopreserved human dental pulp (hDPSCs-cryo) and to analyze the scale of in vivo motor nerve regeneration. The hDPSCs-cryo were isolated and cultured from cryopreserved dental pulp tissues, and thereafter differentiated into cholinergic neurons using tricyclodecane-9-yl-xanthogenate (D

Detecting calcified pulp stones in patients with periodontal diseases using digital panoramic and periapical radiographies.

BACKGROUND/PURPOSE: Pulp stones are discrete calcified masses appearing in the dental pulp of healthy, disease and unerupted teeth. They are presented freely within the pulp tissue or attached and embedded in any part of coronal and radicular dentin. Here, the purposes of this study were to identify the presence of pulp stones in periodontal patients using digital panoramic and periapical radiographies, and to determine the association with gender, age, tooth type and dental arch. MATERIALS AND METHODS: This is a retrospective study on selected records of 465 dental samples obtained between J

Non-Psychoactive Cannabinoid Modulation of Nociception and Inflammation Associated with a Rat Model of Pulpitis.

Despite advancements in dental pain management, one of the most common reasons for emergency dental care is orofacial pain. Our study aimed to determine the effects of non-psychoactive Cannabis constituents in the treatment of dental pain and related inflammation. We tested the therapeutic potential of two non-psychoactive Cannabis constituents, cannabidiol (CBD) and β-caryophyllene (β-CP), in a rodent model of orofacial pain associated with pulp exposure. Sham or left mandibular molar pulp exposures were performed on Sprague Dawley rats treated with either vehicle, the phytocannabinoid CBD (5

A Correlation between Clinical Classification of Dental Pulp and Periapical Diseases with its Patho Physiology and Pain Pathway.

BACKGROUND: Dental pain due to pulpal involvement is difficult to diagnose due to the apparent inaccessibility of pulp to the clinical tests, indistinct symptoms, and referred toothache originating from the periodontal tissues. Though we have various clinical classification systems to categorize pulpal diseases, we are yet biased about the exact pathophysiology and pain pathway associated with it. Dental pulp has a complex physiology, and so is its pathophysiology. AIMS & OBJECTIVES: To concisely reviews the basic understanding of the pathophysiology of pulp, pain pathway, and its correlation

Expression Profiling of S100 Proteins in Healthy and Irreversibly Inflamed Human Dental Pulps.

INTRODUCTION: Several S100 proteins have been shown to play an important role in the innate immune response to infection and in regenerative processes. However, they have scarcely been investigated during inflammation of the dental pulp. Therefore, in this study, we performed gene expression profiling of S100 proteins in healthy and inflamed human dental pulps. METHODS: Tissue samples of human dental pulps were used, including 15 clinically diagnosed as symptomatic irreversible pulpitis (SIP), 7 as asymptomatic irreversible pulpitis (AIP), and 19 as healthy pulp (HP). S100 gene expression lev

Relationship between the difference in electric pulp test values and the diagnostic type of pulpitis.

BACKGROUND: According to the diagnosis criteria of the American Association of Endodontists (AAE), sensitive responses to cold and/or heat tests of suspected teeth compared with those of control teeth can be used for the diagnosis of pulpitis, but the role of electric pulp test (EPT) is not mentioned. It is believed that EPT has some limitations in determining the vitality of the pulp. The aim of this study was to explore the association between the difference in EPT values and the differential diagnoses of reversible pulpitis (RP) and symptomatic irreversible pulpitis (SIRP) caused by dental
