NMJ-related diseases beyond the congenital myasthenic syndromes.

Neuromuscular junctions (NMJs) are a special type of chemical synapse that transmits electrical stimuli from motor neurons (MNs) to their innervating skeletal muscle to induce a motor response. They are an ideal model for the study of synapses, given their manageable size and easy accessibility. Alterations in their morphology or function lead to neuromuscular disorders, such as the congenital myasthenic syndromes, which are caused by mutations in proteins located in the NMJ. In this review, we highlight novel potential candidate genes that may cause or modify NMJs-related pathologies in human

Combinatorial approaches increasing neuronal activity accelerate recovery after spinal cord injury.

Combinatorial approaches targeting multiple aspects of spinal cord injury (SCI) pathophysiology are needed to maximize functional recovery. We hypothesized that enhancing neuronal activity-by strengthening corticospinal synapses through Hebbian stimulation and increasing neuronal transmission with 4-aminopyridine (4-AP), a potassium blocker-could accelerate locomotor recovery in individuals with chronic SCI. Participants were randomly assigned to receive 10 mg of 4-AP or placebo, where both groups followed with 60 min of Hebbian stimulation targeting corticospinal-motor neuronal synapses suppl

Intracellular magnesium-dependent modulation of gap junction channels formed by neuronal connexin36.

Gap junction (GJ) channels composed of Connexin36 (Cx36) are widely expressed in the mammalian CNS and form electrical synapses between neurons. Here we describe a novel modulatory mechanism of Cx36 GJ channels dependent on intracellular free magnesium ([Mg(2+)]i). We examined junctional conductance (gj) and its dependence on transjunctional voltage (Vj) at different [Mg(2+)]i in cultures of HeLa or N2A cells expressing Cx36. We found that Cx36 GJs are partially inhibited at resting [Mg(2+)]i. Thus, gj can be augmented or reduced by lowering or increasing [Mg(2+)]i, respectively. Similar chang…

Protein kinase C involvement in the acetylcholine release reduction induced by amyloid-beta(25-35) aggregates on neuromuscular synapses.

Using intracellular recording of the diaphragm muscle of adult rats, we have investigated the short-term functional effects of amyloid-beta (Abeta(25-35) peptide aggregates on the modulation of acetylcholine (ACh) release and the involvement of protein kinase C (PKC). The non-aggregated form of this peptide does not change the evoked and spontaneous transmitter release parameters on the neuromuscular synapse. However, the aggregated form of Abeta(25-35) acutely interferes with evoked quantal ACh release (approximately 40% reduction) when synaptic activity in the ex vivo neuromuscular preparati

Functional and ultrastructural neuroanatomy of interactive intratectal/tectonigral mesencephalic opioid inhibitory links and nigrotectal GABAergic pathways: involvement of GABAA and mu1-opioid receptors in the modulation of panic-like reactions elicited by electrical stimulation of the dorsal midbrain.

In the present study, the functional neuroanatomy of nigrotectal-tectonigral pathways as well as the effects of central administration of opioid antagonists on aversive stimuli-induced responses elicited by electrical stimulation of the midbrain tectum were determined. Central microinjections of naloxonazine, a selective mu(1)-opiod receptor antagonist, in the mesencephalic tectum (MT) caused a significant increase in the escape thresholds elicited by local electrical stimulation. Furthermore, either naltrexone or naloxonazine microinjected in the substantia nigra, pars reticulata (SNpr), caus

[Multidisciplinary therapy for focal spasticity treatment].

For treating focal dystonias and spastic syndromes local intramuscular injections of botulinum toxin type A (BTA) are the method of choice. The therapy aims to reduce disabling dystonic and spastic movement disorders, ease care and personal hygiene, reduce pain, and improve quality of life. With respect to functional improvement from and accuracy of BTA-injection, treatment results in spasticity often profits from electrical or sonographical guidance of BTA injection in addition to active time localized training methods (physio- and occupational therapy). Since we know that only active synapse

Cell body position of Drosophila Moonwalker Descending Neurons regulates locomotor circuit function.

Both mammalian and invertebrate nervous systems contain densely packed neuronal cell bodies in stereotyped locations, which express cell sur … l body clustering, which blocks initiation of backward walking. When the cell bodies are clustered, electrical synapses form between MDN cell bodies. The gap junction protein Innexin 8 (also known as Shaking B) maintains a more depolarized resting membrane potential and allows synchronous firing of MDNs. Together, these electrophysiological properties are necessary t …

Recent Advances in the Synthesis and Application of Tellurium Semiconductors.

Tellurium (Te), an attractive p-type van der Waals semiconductor, has been considered a promising candidate in electrical applications due to its unique one-dimensional chiral atomic-helical-chain structure, tunable bandgap, and ultrahigh hole mobility. This review summarizes recent advances in the controlled synthesis of Te semiconductor nanostructures, including one-dimensional tellurium nanowires and two-dimensional tellurene in the form of nanosheets and thin films. We further highlight emerging electrical applications of Te in field-effect transistors, logic circuits, photodetectors, memr

High-Frequency Electrical Stimulation Elicits Distinct Response Modes Via Synaptic Interplay.

High-Frequency electrical stimulation (HFS) has gained prominence as a powerful neuromodulation technique with multiple therapeutic interventions, yet its mechanisms of action and interaction with synaptic circuitry remain unclear. In this study, we examined how 2 kHz HFS interacts with excitatory and inhibitory signaling. We targeted retinal ganglion neurons as they preserve intact synaptic circuits and single-cell recordings via whole-cell patch clamping are readily achievable, making them an ideal model for investigating HFS-induced modulation in physiologically relevant conditions. Artificial Synapse Based on Black Phosphorus/SnS2 Heterostructure Transistor for Neuromorphic Computing with High Accuracy.

Artificial synaptic devices have garnered significant attention for overcoming the limitations of conventional von Neumann architecture in neuromorphic computing. However, the reported nonlinear weight update of artificial synapses prevents a hardware neural network from achieving the same high training and learning accuracy levels as those attained by a soft neural work. Despite the large-area fabrication constraints, two-dimensional (2D) materials that have tunable electronic band alignment and optoelectronic properties have emerged as promising candidates for artificial synapses. Here, we d

Electrical stimulation of mesencephalic noradrenergic pathway: effects on luteinizing hormone levels in blood of ovariectomized and ovariectomized, steroid-primed rats.

This study examined the effect of electrical stimulation of the dorsal mesencephalic tegmentum (DMT) region on blood LH levels in long term ovariectomized (OVX), pentobarbital-anesthetized rats, with the aim of activating the principal ascending noradrenergic (NE) bundle. In OVX unprimed rats, electrical stimulation (with parameters of 100 Hz, 0.5-msec effective biphasic pulses, 150-200 microA, and 15 sec on/off for up to 1.5 h) of the DMT region inside the ascending NE bundle either completely or partially inhibited the pulsatile pattern of blood LH levels characteristic of OVX animals; stimu

In vivo AAV9-Myo7a gene rescue restores hearing and cholinergic efferent innervation in inner hair cells.

In the mammalian cochlea, sensory hair cells are crucial for the transduction of acoustic stimuli into electrical signals, which are then relayed to the central auditory pathway via spiral ganglion neuron (SGN) afferent dendrites. The SGN output is directly modulated by inhibitory cholinergic axodendritic synapses from the efferent fibers originating in the superior olivary complex. When the adult cochlea is subjected to noxious stimuli or aging, the efferent system undergoes major rewiring, such that it reestablishes direct axosomatic contacts with the inner hair cells (IHCs), which occur onl

The Mood Stabilizer Lithium Slows Down Synaptic Vesicle Cycling at Glutamatergic Synapses.

Lithium is a mood stabilizer broadly used to prevent and treat symptoms of mania and depression in people with bipolar disorder (BD). Little is known, however, about its mode of action. Here, we analyzed the impact of lithium on synaptic vesicle (SV) cycling at presynaptic terminals releasing glutamate, a neurotransmitter previously implicated in BD and other neuropsychiatric conditions. We used the pHluorin-based synaptic tracer vGpH and a fully automated image processing pipeline to quantify the effect of lithium on both SV exocytosis and endocytosis in hippocampal neurons. We found that lit

The Corticofugal Effects of Auditory Cortex Microstimulation on Auditory Nerve and Superior Olivary Complex Responses Are Mediated via Alpha-9 Nicotinic Receptor Subunit.

BACKGROUND AND OBJECTIVE: The auditory efferent system is a complex network of descending pathways, which mainly originate in the primary auditory cortex and are directed to several auditory subcortical nuclei. These descending pathways are connected to olivocochlear neurons, which in turn make synapses with auditory nerve neurons and outer hair cells (OHC) of the cochlea. The olivocochlear function can be studied using contralateral acoustic stimulation, which suppresses auditory nerve and cochlear responses. In the present work, we tested the proposal that the corticofugal effects that modul

Differential short-term changes in GABAergic or glycinergic synaptic efficacy on rat hypoglossal motoneurons.

Using whole cell patch-clamp recording from hypoglossal motoneurons of a neonatal rat brain slice preparation, we investigated short-term changes in synaptic transmission mediated by GABA or glycine. In 1.5 mM extracellular Ca(2+) [Ca(2+)](o), pharmacologically isolated GABAergic or glycinergic currents were elicited by electrical stimulation of the reticular formation. At low stimulation frequency, glycinergic currents were larger and faster than GABAergic ones. GABAergic currents were strongly facilitated by pulse trains at 5 or 10 Hz without apparent depression. This phenomenon persisted af

Functional evaluation of biological neurotoxins in networked cultures of stem cell-derived central nervous system neurons.

Therapeutic and mechanistic studies of the presynaptically targeted clostridial neurotoxins (CNTs) have been limited by the need for a scalable, cell-based model that produces functioning synapses and undergoes physiological responses to intoxication. Here we describe a simple and robust method to efficiently differentiate murine embryonic stem cells (ESCs) into defined lineages of synaptically active, networked neurons. Following an 8 day differentiation protocol, mouse embryonic stem cell-derived neurons (ESNs) rapidly express and compartmentalize neurotypic proteins, form neuronal morpholog

Rehabilitation following brain damage: some neurophysiological mechanisms. The effects of repetitive stimulation in recovery from damage to the central nervous system.

There is a growing body of evidence that the central nervous system (CNS), even in the adult animal, is capable of adaptation and reorganization not only as a result of partial damage to the CNS but also in response to stimulation. Environmental stimulation produces changes including expansion of visual cortex, increases in dendritic branching, glia and cholinesterase. Environmental stimulation also produces behavioural changes. Experimental electrical stimulation produces changes in synapse size, synaptic vesicle change, dendritic branching and changes in synaptic transmission. In man, repeti
