Effects of Embryo Production Method and Culture Medium on Embryonic Development in Red-Rumped Agouti.

Advances in Assisted Reproductive Technologies (ARTs) help overcome reproductive barriers. However, a comprehensive understanding of embryonic development is crucial for their success. In vitro fertilization (IVF) and artificial oocyte activation (AOA) are embryo production methods commonly used. Previous studies have reported low developmental success with these methods, possibly due to the culture medium used. Therefore, we aimed to optimize ARTs in red-rumped agouti by evaluating the influence of production method and culture medium on embryonic development. Oocytes were matured in vitro an

Immunohistochemical localization of nanog and Oct4 in stem cell compartments of human sacrococcygeal teratomas.

AIMS: To study the range of differentiation and presence of cells positive for stem cell markers in 20 sacrococcygeal teratomas (SCTs) which were consecutively operated on between 1990 and 2000 in the Department of Paediatric Surgery in Tübingen, Germany. METHODS AND RESULTS: Preserved paraffin-embedded material was re-evaluated. In addition to tissues of various organs, caudal organ structures not described before were identified, such as colon with pancreas originating from colonic crypts, Fallopian tube and vaginal epithelia. The derivation of the latter was confirmed by Müllerian duct spe

Placental Inflammation Leads to Abnormal Embryonic Heart Development.

BACKGROUND: Placental heart development and embryonic heart development occur in parallel, and these organs have been proposed to exert reciprocal regulation during gestation. Poor placentation has been associated with congenital heart disease, an important cause of infant mortality. However, the mechanisms by which altered placental development can lead to congenital heart disease remain unresolved. METHODS: In this study, we use an in vivo neutrophil-driven placental inflammation model through antibody depletion of maternal circulating neutrophils at key stages during time-mated murine preg

Early embryogenesis in CHDFIDD mouse model reveals facial clefts and altered cranial neurogenesis.

CDK13-related disorder, also known as congenital heart defects, dysmorphic facial features and intellectual developmental disorder (CHDFIDD) is associated with mutations in the CDK13 gene encoding transcription-regulating cyclin-dependent kinase 13 (CDK13). Here, we focused on the development of craniofacial structures and analyzed early embryonic stages in CHDFIDD mouse models, with one model comprising a hypomorphic mutation in Cdk13 and exhibiting cleft lip/palate, and another model comprising knockout of Cdk13, featuring a stronger phenotype including midfacial cleft. Cdk13 was found to be

Expert Fusion Network for Automated Blastocyst Morphology and IVF Decision Support.

Conventional morphology-based grading of blastocysts is subjective and inconsistent. Although artificial intelligence and deep learning methods have im proved performance, they continue to struggle with generalization across imaging domains, low-contrast structures, and multi-scale feature integration, which limits clinical applicability and may inflate performance estimates. We propose the Expert Fusion Network (EFN), an AI-driven framework designed to address these challenges in blastocyst assessment. EFN automates the classification and segmentation of the key embryonic structures: the trop…

Time-Lapse Imaging and Morphometric Analysis of Tracheal Development in Drosophila.

Detailed and quantitative analyses of the cellular events underlying the formation of specific organs or tissues is essential to understand the general mechanisms of morphogenesis and pattern formation. Observation of live tissues or whole-mount fixed specimens has emerged as the method of choice for identifying and quantifying specific cellular and tissular structures within the organism. In both cases, cell and subcellular structure identification and good quality image acquisition for these analyses are essential. Many markers for live imaging and fixed tissue are now available for detectin

VACTERL association with double-chambered left ventricle: A rare occurrence.

VACTERL association is a non-random association of birth defects of unknown etiology derived from structures of embryonic mesoderm. The common cardiac defects seen with VACTERL association are ventricular septal defects, atrial septal defects, and tetralogy of Fallot. We present a 2-year-old child with VACTERL association in whom we detected double-chambered left ventricle on transthoracic echocardiography.

Foregut mesenchyme contributes cells to pancreatic acini during embryonic development in a chick-quail chimera model.

To understand causes of developmental abnormalities of the pancreas, it is essential to understand its normal embryonic development. Current understanding of the development of pancreatic exocrine tissue is that it develops solely from embryonic epithelium, while the role of the surrounding mesenchyme is to signal to this epithelium and form connective tissue. Recent work in our laboratory has shown that pancreatic bud mesenchyme can contribute cells to islets during embryonic development. However, no published studies have investigated in detail whether mesenchyme contributes cells to the exo

Nicotine-induced embryonic malformations mediated by apoptosis from increasing intracellular calcium and oxidative stress.

BACKGROUND: Tobacco smoking by women during pregnancy increases the risk of congenital birth defects in the infants. Among the smoke products, nicotine is believed to be the major teratogenic factor that perturbs embryonic development. However, the role of nicotine in embryonic malformations has not been addressed, and the mechanisms by which nicotine affects embryonic development remain to be delineated. METHODS: To investigate the effects of nicotine on early embryogenesis, murine embryos at embryonic day (E) 8.5 were dissected out of the uteri, cultured in a roller bottle system, and treat

Isoleucyl-tRNA synthetase 1 is essential for embryogenesis in mice.

Isoleucyl-tRNA synthetase 1 (IARS1) is essential for protein synthesis. Although IARS1 mutations cause growth and metabolic disorders in humans, its role in embryonic development remains unclear. We generated Iars1 knockout (KO) mice and observed complete absence of homozygous KOs among 44 pups, indicating embryonic lethality. A χ2 test confirmed significant deviation from Mendelian ratios. At E12.5, multiple resorbed implantation sites were observed, and histology at E9.5 showed embryos lacking recognizable structures. Evidence of implantation (decidual swellings with residual trophoblast and

Semilobar Holoprosencephaly with Neurogenic Hypernatraemia: Two new cases.

Holoprosencephaly (HPE) is a developmental defect of the embryonic forebrain and midface. It is due to the non-cleavage of the embryonic forebrain into two cerebral hemispheres and the incomplete development of the paramedian structures. The overall prevalence is 1.31 per 10,000 births. The aetiology could be genetic, environmental, or both. HPE is classified into alobar, semilobar, and lobar subtypes based on the degree of separation of the cerebral hemispheres. We report two new cases of semilobar HPE with neurogenic hypernatraemia. Lack of thirst and hypodypsia associated with chronic hyper

Pax 1, a member of a paired box homologous murine gene family, is expressed in segmented structures during development.

The mouse genome contains at least three copies of sequences homologous to the "paired box", a conserved domain in several Drosophila segmentation genes of the pair-rule and segment polarity classes. Overlapping phages were isolated from two different genomic libraries using the Drosophila gooseberry distal paired box as a probe. The hybridizing sequences are highly homologous to the conserved Drosophila paired box sequences. A single 3.1 kb Pax 1 (paired box gene) transcript was detected during embryonic development, whereas no transcripts were detected in adult tissues. Detailed in situ hybr

Preparation of Mouse Embryonic Eye Sections for Routine Histology.

Eye development is a complex process that begins during the first stages of embryonic development; during its formation, a series of cell proliferation, migration, and differentiation events occur. A frequently used laboratory method is tissue staining with two dyes known as hematoxylin-eosin (H&E). H&E staining allows us to observe the arrangement of the cell layers and the formation of characteristic structures. It is fundamental for research in developmental biology since it facilitates a comprehensive analysis of the formation of organs and systems. In this chapter, we will describe the pro

A cross-tissue physicochemical causal chain underlying vertebrate mandibular morphogenesis.

Understanding morphogenesis increasingly requires attention to physical aspects. In complex embryonic structures comprising multiple adjacent tissues, morphogenetic change in one tissue can impose geometric constraints that shape another tissue. However, such cross-tissue physical coordination remains largely unexplored. Using zebrafish jaw development, we show that dynamic folding of the oral ectoderm drives the elongation and fusion of mandibular primordia by constraining and guiding mesenchymal cell motion. This folding is initiated by Shh signaling, mainly from the adjacent neural tube, wh…

Nkx2-5(+)islet1(+) mesenchymal precursors generate distinct spleen stromal cell subsets and participate in restoring stromal network integrity.

Secondary lymphoid organ stromal cells comprise different subsets whose origins remain unknown. Herein, we exploit a genetic lineage-tracing approach to show that splenic fibroblastic reticular cells (FRCs), follicular dendritic cells (FDCs), marginal reticular cells (MRCs), and mural cells, but not endothelial cells, originate from embryonic mesenchymal progenitors of the Nkx2-5(+)Islet1(+) lineage. This lineage include embryonic mesenchymal cells with lymphoid tissue organizer (LTo) activity capable also of supporting ectopic lymphoid-like structures and a subset of resident spleen stromal c

Retinoic acid affects craniofacial patterning by changing Fgf8 expression in the pharyngeal ectoderm.

Retinoic acid signaling plays important roles in establishing normal patterning and cellular differentiation during embryonic development. In this study, we show that single administration of retinoic acid at embryonic day 8.5 causes homeotic transformation of the lower jaw into upper jaw-like structures. This homeosis was preceded by downregulation of Fgf8 and Sprouty expression in the proximal domain of the first pharyngeal arch. Downregulation of mesenchymal genes such as Dlx5, Hand2, Tbx1 and Pitx2 was also observed. The oropharynx in retinoic acid-treated embryos was severely constricted.

Quantified growth of the human embryonic heart.

The size and growth patterns of the components of the human embryonic heart have remained largely undefined. To provide these data, three-dimensional heart models were generated from immunohistochemically stained sections of ten human embryonic hearts ranging from Carnegie stage 10 to 23. Fifty-eight key structures were annotated and volumetrically assessed. Sizes of the septal foramina and atrioventricular canal opening were also measured. The heart grows exponentially throughout embryonic development. There was consistently less left than right atrial myocardium, and less right than left ven

Epidermolysis bullosa and embryonic lethality in mice lacking the multi-PDZ domain protein GRIP1.

Glutamate receptor-interacting protein 1 (GRIP1) is an adaptor protein composed of seven PDZ (postsynaptic density-95/Discs large/zona occludens-1) domains, capable of mediating diverse protein-protein interactions. GRIP1 has been implicated in the regulation of neuronal synaptic function, but its physiologic roles have not been defined in vivo. We find that elimination of murine GRIP1 results in embryonic lethality. GRIP1(-/-) embryos develop abnormalities of the dermo-epidermal junction, resulting in extensive skin blistering around day 12 of embryonic life. Ultra-structural characterization
