THE MECHANISM OF THE EMETIC ACTION OF SODIUM SALICYLATE.

In the dog, the emetic ED50 for sodium salicylate was 256+/-113 mg/kg for the intravenous route with a mean latency of 10.4+/-3.96 min, and for the oral route 228+/-68 mg/kg with a mean latency of 18.2+/-2.63 min. Values are means with standard errors. Ablation of the emetic chemoreceptor trigger-zone gave complete protection against the emetic action of 300 mg of sodium salicylate intravenously but only partial protection against the emetic action of the same dose of sodium salicylate orally. Either the intravenous salicylate acted on the trigger-zone or ablation of the trigger-zone interfere

Anti-emetics for cancer chemotherapy-induced emesis: Potential of alternative delivery systems.

Currently, the most commonly used routes of administration of antiemetics in chemotherapeutic regimens are oral and intravenous. Patient compliance and thus efficacy of conventional drug schedules and formulations are often impaired by difficulties associated with oral or intravenous uptake of the administered chemotherapy. Alternative or new drug delivery systems should overcome these problems by improving patient compliance. Several new drug delivery systems are available and development of these new systems is ongoing, in particular to meet delivery requirements of modern biological therape…

Cardio-respiratory effects of paraquat with and without emetics on Wistar rats.

We investigated the effects of paraquat products both containing and devoid of emetics on the cardio-respiratory functions of Wister rats. Anesthetized rats were administered intravenously 0.46 ml/kg Gramoxone, a commercial product containing 24% paraquat and an emetic. Paraquat equivalence equalled 110 mg/kg body weight. ECG and respiratory curves were recorded with a polygraph. Fifteen seconds after injection, A-V block and bradycardia occurred and the heart stopped beating soon after. Though the heart was severely affected, the respiratory curve differed very little from pre-injection value…

Use of taste repellants and emetics to prevent accidental poisoning of dogs.

Twelve taste repellents and 3 oral emetics were tested. The taste repellents were capsaicin, capsicum, oleoresin, sucrose octaacetate, quinine tonic, quassia wood extract, vanillamide, horseradish extract, caffeine, pepperoni enhancer, acorn extract, and commercially available bitter and hot flavors. The emetics tested were: antimony potassium tartrate, apomorphine, and copper sulfate. Intake of a 20% sucrose solution by Beagles was significantly depressed by addition of vanillamide at concentrations greater than 0.001%, by capsicum and capsaicin at concentrations greater than 0.01%, and by ho…

Centrally acting emetics. 10. Rigid dopamine congeners derived from octahydrobenzo[f]quinoline.

In a study of conformational requirements for certain dopaminergic agonist molecules, a series of conformationally predictable dopamine congeners related to cis- and trans-octahydrobenzo[f]quinoline was prepared. The complexity and equivocal character of the reduction of variously substituted 4-methyl-1,2,3,4,5,6-hexahydrobenzo[f]quinolines were demonstrated and studied. It was shown that several literature methods for reduction of these systems were in error regarding the stereochemical nature of the product(s). It has been concluded that geometrically specific and predictable reductions of t

Rapid identification of emetic Bacillus cereus by immunochromatography.

The immunochromatographic assay, which targets a marker protein co-expressed during the synthesis of cereulide by an emetic Bacillus cereus strain, was used for easily, rapidly and specifically identifying the emetic strains among B. cereus strains from various materials associated with food poisonings. All 50 of the emetic strains showed a positive reaction to the assay, but all 50 diarrheal strains had a negative reaction. The bacterial counts of 108 cfu/ml in enrichment broth and 109 cfu/ml in food-containing enrichment were required for the identification of emetic B. cereus. The present a

Assessing the emetic potential of PDE4 inhibitors in rats.

1. Type 4 phosphodiesterase (PDE4) inhibitors mimic the pharmacological actions of alpha(2)-adrenoceptor antagonists. This has been postulated as the mechanism by which PDE4 inhibitors induce emesis and was also demonstrated by their ability to reverse xylazine/ketamine-induced anaesthesia. We further characterized this latter effect since it appears to reflect the emetic potential of PDE4 inhibitors. 2. Selective inhibitors of PDE 1, 2, 3, 4 and 5 were studied in rats, on the duration of anaesthesia induced by the combination of xylazine (10 mg kg(-1), i.m.) and ketamine (10 mg kg(-1), i.m.).

The influence of cyclizine and perphenazine on the emetic effect of meptazinol.

The effectiveness of 50 mg cyclizine and 2.5 mg perphenazine against the emetic sequelae of 100 mg meptazinol were studied in a randomized d … ng nausea or vomiting overall. Perphenazine, 2.5 mg, showed no useful anti-emetic effect. Both anti-emetics increased the soporific effect of premedication at the 90-min interval. Subjects receiving perphenazine experienced significantly more dizziness than those of other groups.
