The role of myofibroblasts in the pathogenesis of Dupuytren's contracture was investigated by light and electron microscopic histochemical methods. Dupuytren's myofibroblasts contain an intracellular contractile mechanism that is driven by the dephosphorylation of adenosine triphosphate. Our study of calcium adenosinetriphosphatase (ATPase) activities verifies that the site of this energy system is on the myofilaments of the myofibroblasts. The degree of ATPase activity, as determined by cell counts, appeared to correlate with the residual contracture as predicted by the Legge and McFarlane Ou

An active Arthus reaction induced in the rabbit cornea leads to scleral disease resembling that in humans. Collagen damage was located in the corneal stroma at the site of the immune reaction. In the electron microscope, some regions of corneal stroma, without loss of lamellar integrity, demonstrated the apparent swelling of individual, scattered collagen fibrils such that a helical arrangement of component microfibrils was evident. In areas showing more extensive damage, fibril fragmentation and solubilization was associated with the aggregation of fibrous banded material. These observations

Opticin, a novel member of the leucine-rich repeat (LRR) family, has been reported to bind to collagen fibrils. Many members of the LRR family of extracellular matrix proteins have been reported to bind to fibrillar collagen and regulate the diameter of collagen fibrils and lateral fusion between fibrils. Collagen fibrils are important for the maintenance of the vitreous body in the eye and growth plate cartilage of joints. Oculo-skeletal dysplasia (OSD) is a heterogeneous group of heritable genetic disorders affecting humans and a few breeds of dogs. Labrador retrievers and Samoyeds affected

Epidermolysis bullosa (EB) in a group of genodermatoses characterized by the fragility of skin. Previous studies on the dystrophic (scarring) forms of EB have suggested abnormalities in anchoring fibrils, morphologically recognizable attachment structures that provide stability to the association of the cutaneous basement membrane to the underlying dermis. Since type VII collagen is the major component of the anchoring fibrils, we examined the genetic linkage of dominant dystrophic EB (EBDD) and the type VII collagen gene (COL7A1) locus, which we have recently mapped to chromosome 3p, in three

Bone is mineralized when hydroxyapatite crystals derived from calcium ions and inorganic phosphate (Pi) grow along collagen fibrils in the extracellular matrix. Mineralization is initiated by nucleation of those crystals. Mature osteoblasts secrete matrix vesicles into osteoid, which contain growing hydroxyapatite crystal seeds. After rupture of the lipid bilayer of those vesicles, crystals continue to grow as a mineralized nodule and adhere to collagen fibrils. It remains controversial whether nucleation occurs mainly in matrix vesicles or also extra-vesicularly around collagen fibrils. Miner

Collagen XIV is a fibril-associated collagen with interrupted triple helices (FACIT) that helps regulate collagen fibril spacing, alignment, and extracellular matrix organization. Its role in corneal biomechanics remains incompletely understood. Here, we investigated murine wild-type (WT) and collagen XIV-deficient (Col14a1-/-) corneas using complementary optical elastography modalities: Brillouin microscopy and air-coupled ultrasound optical coherence elastography (ACUS-OCE). Brillouin microscopy generated high-resolution maps of Brillouin frequency shift in WT and Col14a1-/- corneas (N = 4 p

This article details a quantitative method to measure the D-periodic spacing of type I collagen fibrils using atomic force microscopy coupled with analysis using a two-dimensional fast fourier transform approach. Instrument calibration, data sampling and data analysis are discussed and comparisons of the data to the complementary methods of electron microscopy and X-ray scattering are made. Examples of the application of this new approach to the analysis of type I collagen morphology in disease models of estrogen depletion and osteogenesis imperfecta (OI) are provided. We demonstrate that it i

The mechanical properties of connective tissues are tailored to their specific function, and changes can lead to dysfunction and pathology. In most mammalian tissues the mechanical environment is governed by the micro- and nano-scale structure of collagen and its interaction with other tissue components, however these hierarchical properties remain poorly understood. In this study we use the human cornea as a model system to characterise and quantify the dominant deformation mechanisms of connective tissue in response to cyclic loads of physiological magnitude. Synchronised biomechanical testi

ADAM10 is a transmembrane metalloprotease that regulates diverse signalling functions via the shedding of membrane protein ectodomains, and is implicated in tumour development, including glioblastoma multiforme (GBM) and gastrointestinal (GI) cancers, where high ADAM10 expression is associated with poor prognosis. We assessed the role of ADAM10 by gene knockout (KO) in U251 GBM cells, and its effects on protein shedding and protein expression on cell proliferation and on the growth of tumour xenografts in mice. The growth of tumours was severely delayed, relative to modest effects on prolifera

In Crohn's disease, extracellular matrix (ECM) dysfunction and loss of tissue integrity are key pathological features. Fibril-associated collagens with interrupted triple helices (FACIT) are known to regulate ECM assembly and mediate biophysical cues in their microenvironment but remain largely understudied. This study aimed to investigate a novel biomarker reflecting remodelling of the FACIT collagen type XXII (PRO-C22) and explore its association with differing phenotypes of CD and treatment outcomes. Two cohorts of patients with CD were included in the study [cohort 1 (n = 65), cohort 2 (n …

Hepatic stellate cells (HSCs) in the perisinusoidal space are surrounded by hepatocytes, liver sinusoidal endothelial cells, Kupffer cells, and other resident immune cells. In the normal liver, HSCs communicate with these cells to maintain normal liver functions. However, after chronic liver injury, injured hepatocytes release several proinflammatory mediators, reactive oxygen species, and damage-associated molecular patterns into the perisinusoidal space. Consequently, such alteration activates quiescent HSCs to acquire a myofibroblast-like phenotype and express high amounts of transforming g

In mammals, the periodontal ligament (PDL) is a highly specialized tissue which facilitates tooth eruption and lends mechanical support to the tooth once in occlusion. The PDL extracellular matrix fibers play a major role in such functions. During its development, the spatial arrangement of the PDL extracellular matrix undergoes rapid changes. So that it could be determined whether the structural alteration in the PDL is associated with changes in the expression of collagenous proteins with different functional properties, the transcriptional patterns of collagens I and XII were examined. The

Type VI collagen was immunolocalized in several soft connective tissues at the light and electron microscopic level. Positive labeling was found in all tissues examined, periodontal ligament, gingiva, mesenterium and periosteum. The labeled structures could be divided into 2 categories: microfibrils intermingling with collagen fibrils, and those that formed bundles (oxytalan fibres and elastin-associated microfibrils). Control sections incubated with antibody preabsorbed to purified type VI collagen, or with non-immune antibody, proved to be negative. Our observations indicate that the structu

Data increasingly indicates that many anterior cruciate ligament (ACL) injuries are likely due to tissue damage accumulation and the gradual decrease in ACL strength and stiffness over time. Previous ex vivo experiments found molecular-level ACL damage (collagen unraveling) to be associated with increased tissue compliance at the collagen fibril-level. However, it remains unclear if this biomechanical effect is present in vivo, or what effect other modulators of tissue mechanics (e.g., glycosaminoglycans (GAGs)) have in fatigued-damaged ACL matrices. Herein, we investigated the in vivo relatio

Fibroblasts from normal and progressive systemic sclerosis involved skin more closely resemble in vivo cells when cultured in a collagen gel matrix, than do fibroblasts cultured on plastic. Ultrastructural studies show that the cytoskeleton and secretory organelles from these cells are better developed in the presence of a collagen gel. Fibronectin, glycosaminoglycans and collagen fibrils are produced by the cells and deposited in the preformed matrix. Collagen synthesis is greater in the scleroderma derived fibroblasts for all culture conditions with increased deposition in the matrix. Total

A Ruffini corpuscle was identified in the dense reticular dermis of the human scalp from a patient with alopecia areata. The corpuscle measured approximately 50 micrometer in diameter. One afferent myelinated axon with a diameter of 4-6 micrometer supplies the corpuscle. Branched axon terminals and the associated Schwann cells tightly envelop parallel bundles of collagen fibrils. Axon terminals evidenced focal swellings, and small finger-like protrusions projected into the endoneural connective tissue. The terminals are characterized by the presence of abundant mitochondria, numerous vesicles,

Second harmonic generation (SHG) microscopy is a powerful tool in assessing collagen structure, especially with respect to differentiating the respective architectures of normal and diseased tissues. An under-explored area is exploiting SHG to determine the sub-resolution aspects of the collagen fibril size, polarity, and packing (∼50-100 nm diameters). Due to the phase-matching and associated coherence of SHG, these structural aspects are encoded in the wavelength dependence of the spatial emission and relative conversion efficiency, denoted the creation attributes. As a means to extract this

PURPOSE: Deficiencies in enzymes involved in proteoglycan (PG) turnover underlie a number of rare mucopolysaccharidoses (MPS), investigations of which can considerably aid understanding of the roles of PGs in corneal matrix biology. Here, the authors analyze novel pathologic changes in MPS VII (Sly syndrome) to determine the nature of PG-collagen associations in stromal ultrastructure.
METHODS: Transmission electron microscopy and electron tomography were used to investigate PG-collagen architectures and interactions in a cornea obtained at keratoplasty from a 22-year-old man with MPS VII, wh

The authors studied the posterior attachment of the ciliary muscle in seven young (3-10 yr) and five old (26-34 yr) rhesus monkeys by light microscopy and electron microscopy. Posterior attachment of the muscle bundles consisted of elastic tendons, exclusively. The elastic tendons were continuous with the elastic lamina of Bruch's membrane and were also connected by smaller elastic fibers to an elastic meshwork that surrounds the pars plana vessels. In some areas, the tendons formed focal contacts with the endothelial cells. The authors found that in old eyes, the tendons and the elastic fiber
