Effect of female sex hormone supplementation and withdrawal on gastrointestinal and colonic transit in postmenopausal women.

Females are disproportionately affected by constipation, which is often aggravated during pregnancy. Bowel function also changes during the luteal phase of the menstrual cycle. The aim was to compare the effects of acute administration of female sex steroids on gastric emptying, small bowel transit and colonic transit in healthy postmenopausal subjects. A second aim was to determine whether withdrawal of the hormones was associated with a change in transit. Forty-nine postmenopausal females were randomized to receive for 7 days 400 mg day(-1) micronized progesterone, 0.2 mg day(-1) oestradiol,

Inhibition of tetrahydroisoxazolo-pyridin-3-ol and muscimol and its mechanism on gastrointestinal transit in mice.

The effect of tetrahydroisoxazolo-pyridin-3-ol (THIP) and muscimol on gastrointestinal transit and a possible mechanism for this effect were studied using the charcoal meal test in mice. These agents inhibited transit dose relatedly. The inhibitory effect was completely reversed by yohimbine and phentolamine. A minor degree of antagonism by bicuculline was observed. There were an additive response in morphine-pretreated animals and partial antagonism in naloxone-pretreated animals. Propranolol failed to modify the inhibitory effect. The findings suggest a predominantly alpha 2-receptor-mediate…

Scintigraphic study of the gastrointestinal transit of a liquid meal in patients with chronic Chagas' disease.

Gastric emptying and small bowel transit of a liquid meal (isotonic dextrose) were assessed by a scintigraphic technique in 16 patients with Chagas' disease involving the esophagus and/or the colon, including one case with mega-jejunum, and in 10 control subjects. Initial gastric emptying was faster in the Chagas' disease group than in controls (gastric retention at 15 min, median and (range): 52% (15-86) vs 71% (43-97), P < 0.01) although there was no significant difference in total gastric emptying (T1/2: 20 min (4-132) vs 31 min (13-280), P > 0.05). Both the front (time to reach the proxima

Evaluation of gastrointestinal transit times and pH in healthy cats using a continuous pH monitoring system.

OBJECTIVES: The aim of this study was to characterize gastrointestinal (GI) transit times and pH in healthy cats. METHODS: GI transit times and pH were measured in six healthy, colony-housed, purpose-bred spayed female cats using a continuous, non-invasive pH monitoring system in a sequential order design. For the first period ('pre-feeding'), food was withheld for 20 h, followed by oral administration of a pH capsule. Five hours post-capsule administration, cats were meal-fed by offering them their daily allowance of food for 1 h. For the second period ('post-feeding'), food was withheld for

Cerebral dopamine neurotrophic factor is essential for enteric neuronal development, maintenance, and regulation of gastrointestinal transit.

Cerebral dopamine neurotrophic factor (CDNF) is expressed in the brain and is neuroprotective. We have previously shown that CDNF is also ex … re reflected functionally in delayed gastric emptying, slowed colonic motility, and prolonged total gastrointestinal transit. CDNF is expressed selectively in isolated enteric neural crest-derived cells (ENCDC), which also express the dopamine-related transcription factor Foxa2. Addition of CDNF to ENCDC promotes development of dopaminergic neurons; moreover, survival of …

Whole gut transit scintigraphy in the clinical evaluation of patients with upper and lower gastrointestinal symptoms.

OBJECTIVE: In a single noninvasive, quantitative test, whole gut transit scintigraphy (WGTS) measures gastric emptying (GE), small bowel transit (SBT), and colonic transit (CT). The aim of this study was to investigate the clinical utility of WGTS in patients with functional gastrointestinal (GI) symptoms. METHODS: A total of 108 patients with either dyspeptic upper GI symptoms (n = 35) or constipation (n = 73) underwent WGTS. Patients consumed a meal of 99 m-Tc egg sandwich with 111-In DTPA in water. They were imaged every 30 min for 6 h to measure GE and SBT, and at 24, 48, and 72 h to meas

Hyperglycaemia as a factor affecting kappa-opiate agonist-induced inhibition of the gastrointestinal transit in mice.

The effects of highly selective kappa-opiate agonists were assessed on the gastrointestinal motility in normoglycaemic and hyperglycaemic conditions in mice. Chronic hyperglycaemia was induced by streptozocin injection (200 mg kg-1 i.p.), 7-8 days before the experiment. Acute hyperglycaemia was induced by glucose injection (5 g kg-1 i.p.) at the time of opiate administration. The kappa-opiate agonists, U-50488H and U-69593 (1, 3 and 10 mg kg-1) were injected (i.p.) just before the charcoal meal. The animals were killed 45 min later and the distance travelled by the test meal was measured. In t

Action of T-2 toxin on gastrointestinal transit in mice: protective effect of an argillaceous compound.

Using sodium [51Cr]chromate as radiolabeled marker, gastrointestinal transit of a milk test meal was determined in mice receiving for 4 days T-2 toxin (1 mg/kg per day per os) alone or with a clay, smectite (2 g/kg per day), given according to four different procedures. Gastric emptying and small intestinal transit were significantly accelerated after the 1st T-2 administration and during the 4 days of treatment. When smectite was given together with the toxin with or without pretreatment by smectite alone for 2 days, the T-2 induced disturbances in gastrointestinal transit remained unchanged.…

Transition of care in pediatric surgical patients with complex gastrointestinal disease.

Pediatric surgeons provide care for infants and children with a wide variety of conditions throughout the body. Many of these conditions are congenital or occur very early in life, and for this reason, providing continuity of care for these patients into adulthood is an emerging challenge. In the gastrointestinal tract, congenital and acquired conditions are now associated with excellent long-term prognosis; however, little guidance on long-term care exists. The aim of this article is to discuss aspects that are important to transitioning care of pediatric surgical patients with complex gastro

[Transition in chronic gastrointestinal diseases. From pediatric to adult care].

Technological advances and the globalization of knowledge have led to a considerable increase in the number of patients with chronic gastrointestinal disease who transition from pediatric to adult care during one of the most vulnerable life stages: adolescence. The Transition Working Group of the Gastroenterology Committee of the Sociedad Argentina de Pediatría conducted an exhaustive literature search and summoned leading specialists in the most frequent chronic pathologies from all over the country to unify criteria based on evidence and experience. As a result, a series of recommendations a

Effects of two different acute and subchronic stressors on gastrointestinal transit in the rat: A radiographic analysis.

BACKGROUND: The reaction to stress is an adaptive response necessary for survival. When stressors are repeated, the organism adapts, although these adaptive responses can become dysregulated and result in disease, causing gastrointestinal (GI) disorders. Radiographic methods allow the non-invasive study of how a given factor affects GI transit in the same animal at different time points. These methods have never been applied to study the consequences of stress on GI motor function and their dependency on time and stimulus. Therefore, our aim was to characterize, using radiographic techniques,

Inhibition of gastrointestinal transit due to surgical trauma or peritoneal irritation is reduced in capsaicin-treated rats.

Treatment of newborn rats with capsaicin (0.16 mmol/kg) is known to cause a permanent degeneration of mainly unmyelinated afferent neurons. In this study, postoperative ileus and ileus produced by peritoneal irritation with iodine were investigated in adult rats treated with capsaicin as neonates. It was found that in capsaicin-treated rats both forms of ileus, measured as inhibition of gastrointestinal transit, were significantly reduced as compared with vehicle-treated control animals. Adrenoceptor blockade in untreated rats reduced the ileus in response to peritoneal irritation to approxima…

Contrast-enhanced multispectral optoacoustic tomography for the assessment of the gastrointestinal transit in patients with cystic fibrosis.

Cystic fibrosis (CF) affects the gastrointestinal tract, but assessing gastrointestinal transit usually requires invasive procedures or exposure to ionizing radiation. Contrast-enhanced multispectral optoacoustic tomography (CE-MSOT) offers a novel, non-invasive, and radiation-free approach to assess gastrointestinal function by orally administered dyes. In this clinical pilot-study five patients with cystic fibrosis and four healthy volunteers received CE-MSOT before and 6-times hourly after a standardized breakfast with Indocyanin green (ICG) as dye. The gastric antrum, terminal ileum and si…
