Sequential histopathology of cavitary liver abscess. Formation induced by axenically grown Entamoeba histolytica.

Multiple hamster liver passage of Entamoeba histolytica trophozoites with intervening recovery into axenic culture caused increased virulence as measured by increase in the size of the lesion produced. Lesions produced by amebae that had not been liver-passaged did not persist; however, multiply liver-passaged substrains produced large, fluid-filled abscesses one month to six weeks after inoculation. Six days after inoculation, lesions consisted of multiple granulomas, lymphocytes, and E histolytica trophozoites. Large, fluid-filled abscesses produced by liver-passaged substrains lacked the gr

Sarcocystis fayeri sp. n. from the horse.

Hearts, diaphragms, esophagi, and spinal cords from 266 horses were obtained at slaughter in Creston, Ohio. Tissues were examined microscopically for Sarcocystis in sections, digested in trypsin to obtain bradyzoites, and fed to 10 dogs and 10 cats. Intramuscular cysts were found in selections of two hearts from 57 horses and four esophagi from 107 horses. The cysts were up to 900 micron long and up to 70 micron wide. The cyst wall was 1 to 2 micron thick and cross-striated. The enclosed bradyzoites were banana-shaped, 15 to 20 by 20 to 3 micron, and contained several PAS-positive granules. Br

Intestinal colonization of germ-free mice with indole-producing E. coli modulates the central and peripheral endocannabinoidome.

Colonization of germ-free (GF) mice with indole-producing E. coli exacerbates anxiety- and depression-like behaviors. Lack and subsequent reintroduction of gut microbiota in mice alter in opposing ways the brain and intestinal levels of the lipid mediators of the expanded endocannabinoid system, or endocannabinoidome (eCBome), which controls affective behaviors. By using LC/MS-MS-based targeted lipidomics and qPCR, we investigated the effect of GF mouse colonization with E. coli strains capable (wild-type, I+) or not (knock-out, I-) of producing indole, on the brain and intestinal eCBome. Protocol for performing traumatic spinal cord injury surgery on mice in germ-free isolators and assessing injury recovery and pathophysiology.

An altered gut microbiome affects recovery from traumatic spinal cord injury (SCI). Here, we present a protocol for performing SCI survival surgery on mice in germ-free isolators. Specifically, we describe steps for colonization, preparing surgical tools, performing surgery, and post-surgical care. We then outline procedures for behavioral assessment, followed by collecting and processing tissue to assess injury recovery and pathophysiology. This protocol can be easily adapted to study the gut microbiome's influence on other forms of central nervous system injury.

Colesevelam Reduces Ethanol-Induced Liver Steatosis in Humanized Gnotobiotic Mice.

Alcohol-related liver disease is associated with intestinal dysbiosis. Functional changes in the microbiota affect bile acid metabolism and result in elevated serum bile acids in patients with alcohol-related liver disease. The aim of this study was to identify the potential role of the bile acid sequestrant colesevelam in a humanized mouse model of ethanol-induced liver disease. We colonized germ-free (GF) C57BL/6 mice with feces from patients with alcoholic hepatitis and subjected humanized mice to the chronic-binge ethanol feeding model. Ethanol-fed gnotobiotic mice treated with colesevelam

Control of viruses infecting grapevine.

Grapevine is a high value vegetatively propagated fruit crop that suffers from numerous viruses, including some that seriously affect the profitability of vineyards. Nowadays, 64 viruses belonging to different genera and families have been reported in grapevines and new virus species will likely be described in the future. Three viral diseases namely leafroll, rugose wood, and infectious degeneration are of major economic importance worldwide. The viruses associated with these diseases are transmitted by mealybugs, scale and soft scale insects, or dagger nematodes. Here, we review control meas

Pathogenicity of Actinobacillus minor, Actinobacillus indolicus and Actinobacillus porcinus strains for gnotobiotic piglets.

The purpose of the study was to evaluate the clinical significance of Actinobacillus minor, Actinobacillus porcinus and Actinobacillus indolicus strains in gnotobiotic piglets. Twenty-two 6-h-old Caesarean-delivered and colostrum-deprived piglets were intranasally and orally inoculated with 2 x 10(6) colony-forming units of an A. minor (group 2; n = 9), A. indolicus (group 3; n = 5), or A. porcinus (group 4; n = 8) strain. Six other piglets were inoculated in the same way with phosphate-buffered saline solution and used as controls (group 1). All pigs were observed for clinical signs and recta

The role of intestinal microflora in the formation of the methylthio adduct metabolites of paracetamol. Studies in neomycin-pretreated and germ-free mice.

The contribution of the gastrointestinal microflora to the formation of methylthio adducts from paracetamol has been studied by comparing the fate of this drug in conventional mice with that in germ-free and neomycin-treated animals. In both germ-free and neomycin-treated mice there was a highly significant reduction in the urinary excretion of 3-methylthioparacetamol, its glucuronic acid and sulfate conjugates and its sulfoxide, with no other systemic alteration to the overall fate of the drug. These data are consistent with the gut flora playing a major role in the C-S cleavage of paracetamo

Intestinal deconjugation of bilirubin in germfree and conventional rats.

The content of conjugated bilirubin (CB) was about the same in small intestinal contents from germfree (GF) and conventional (CONV) rats. Caecal contents from GF rats contained considerably more CB than from CONV rats. The results indicated that the caecum is the major site of microbial deconjugation of CB in the CONV rat. Separation of azopigments prepared from small intestinal contents of GF and CONV rats revealed similar patterns. Similar separation patterns were also observed with azopigments prepared from caecal contents and faeces of GF rats. Some chemical properties of CB prepared from

[Use of the gnotobiotic rat for determination of the in vivo activity of Bifidobacterium beta-galactosidase].

Under the conditions of in vitro cultivation, the height of the beta-galactosidase activity of Bifidobacterium spec. is essentially influenced by the composition of the culture medium. The use of gnotobiotic (germ-free and monoassociated with Bifidobacterium longum) rats permitted to differentiate in the chyme between beta-galactosidase activities of mucosal and microbial origin. In germ-free animals, the chyme in the small intestine and the colon contains nearly 10-20% of the activity measured in the mucosa (in each case expressed as g on a wet-weight basis). Monoassociation with B. longum do
