Efficacy of postexposure therapy against glanders in mice.

Burkholderia mallei, the causative agent of glanders, is a CDC Tier 1 Select Agent for which there is no preventive vaccine and antibiotic therapy is difficult. In this study, we show that a combination of vaccination using killed cellular vaccine and therapy using moxifloxacin, azithromycin, or sulfamethoxazole-trimethoprim can protect BALB/c mice from lethal infection even when given 5 days after infectious challenge. Vaccination only, or antibiotic therapy only, was not efficacious. Although antibiotics evaluated experimentally can protect when given before or 1 day after challenge, this ti…

The hamster model of intraperitoneal Burkholderia mallei (glanders).

Thirty-one female Syrian hamsters (Mesocricetus auratus) were inoculated intraperitoneally with a lethal dose of Burkholderia mallei (Budape … ously. The results of the study indicate that the Syrian hamster is a useful small animal model for glanders.

From glanders to globulins: A study in comparative medicine.

The anti-globulin test was described in 1945, and ever since has been synonymous with the lead author, Robin Coombs, a young veterinary surgeon, at that time embarking on a career in immunological research. This was marked by a number of important contributions in the field, including the description and categorisation of hypersensitivity reactions, co-authored with Philip Gell. Together they wrote the classical text, Clinical Aspects of Immunology, which has been updated and republished over the ensuing 50 years. Although Robin Coombs is best remembered for his contributions to medical immuno

Present and future therapeutic strategies for melioidosis and glanders.

Burkholderia pseudomallei and Burkholderia mallei are the causative agents of melioidosis and glanders, respectively. Both Gram-negative pathogens are endemic in many parts of the world. Although natural acquisition of these pathogens is rare in the majority of countries, these bacteria have recently gained much interest because of their potential as bioterrorism agents. In modern times, their potential destructive impact on public health has escalated owing to the ability of these pathogens to cause opportunistic infections in diabetic and perhaps otherwise immunocompromised people, two growi…

[Rayer's studies on the contagion of glanders (1837-1843)].

P. Rayer (1795-1867) had never thoroughly published his experimental studies on the contagion of glanders. His recently un-earthed hand written papers allow us to depict his experimental approach and its results. He was not the first who transmitted glanders from a patient to horses or donkeys. But he did it systematically with glander secretions from acute and chronic cases. Whatever was the disease of the donors the transmitted forms were unpredictably either chronic or acute. His conclusion was that the two forms were two symptomatic aspects of a unique disease. Clinically dormant states we…

Epidemiological Situation of Glanders in the State of Pará, Brazil.

Glanders is an anthropozoonosis caused by the bacteria Burkholderia mallei, affecting mainly equids. It has been eradicated in North America, Australia, and Western Europe, but continues to occur sporadically in countries in Asia, Africa, the Middle East, and South America. Its notification is mandatory by the World Organization for Animal Health. After 30 years, the disease reappeared in Brazil in 1999 and, thereafter, 1,413 outbreaks have been reported. However, the epidemiological situation of the disease in the country is not adequately known. Thus, 2718 animals from 654 properties in the …

Efficacy of finafloxacin in a murine model of inhalational glanders.

Burkholderia mallei, the causative agent of glanders, is principally a disease of equines, although it can also infect humans and is categorized by the U.S. Centers for Disease Control and Prevention as a category B biological agent. Human cases of glanders are rare and thus there is limited information on treatment. It is therefore recommended that cases are treated with the same therapies as used for melioidosis, which for prophylaxis, is co-trimoxazole (trimethoprim/sulfamethoxazole) or co-amoxiclav (amoxicillin/clavulanic acid). In this study, the fluoroquinolone finafloxacin was compared …

Emerging role of biologics for the treatment of melioidosis and glanders.

Introduction: Two important pathogenic species within the genus Burkholderia, namely Burkholderia pseudomallei (Bpm) and Burkholderia mallei (Bm), are the causative agents of the life-threatening diseases melioidosis and glanders, respectively. Due to their high mortality rate and potential for aerosolization, they have gained interest as potential biothreat agents and are classified as Tier 1 Select Agents.Areas covered: The manuscript provides an overview of the literature covering the efforts taken in the last 10 years to develop new therapeutics measures against both Bpm and Bm, with atten…

Humoral immune responses in a human case of glanders.

Within 2 months of acquiring glanders, a patient developed 8-, 16-, and 4-fold increases, respectively, in specific IgA, IgG, and IgM serological titers against Burkholderia mallei. Within 14 months of infection, the titers decreased to the baseline. Serum from this patient was also highly reactive against Burkholderia pseudomallei whole cells. Burkholderia mallei whole cells did not react with sera from patients with other diseases. Therefore, an assay using a B. mallei cellular diagnostic antigen may be useful for the serodiagnosis of glanders.

Use of a Western blot technique for the serodiagnosis of glanders.

BACKGROUND: The in vivo diagnosis of glanders relies on the highly sensitive complement fixation test (CFT). Frequently observed false positive results are troublesome for veterinary authorities and cause financial losses to animal owners. Consequently, there is an urgent need to develop a test with high specificity. Hence, a Western blot assay making use of a partly purified lipopolysaccaride (LPS) containing antigen of three Burkholderia mallei strains was developed. The test was validated investigating a comprehensive set of positive and negative sera obtained from horses and mules from end…

Early methods for the surveillance and control of glanders in Europe.

From the many existing documents on the history of glanders, it is possible to detail the practical measures adopted for disease surveillance and control from antiquity until the 19th century, principally in European countries. Surveillance is based on clinical diagnosis, post-mortem examination, animal inoculation and knowledge of the conditions under which infection occurred: aetiology, pathogenesis, susceptible species, virulent material, mode of infection, incubation period, etc. The historical data are assembled and compared, with comments on each of these points. Control is based on the …

[In vitro development of fluoroquinolone resistance in the glanders pathogen].

To improve the scheme for the therapy of malleus, the possible development of fluoroquinolone resistance in the cells of Pseudomonas mallei was studied. The frequency of the mutations determining the resistance to oxolinic acid, norfloxacin, enoxacin, ofloxacin and ciprofloxacin was detected. The spectrum of the cross resistance in the strains was investigated. The influence of the resistance mutations on the culture virulence was studied. Recommendations on the rational use of fluoroquinolones in the treatment of malleus are presented.
