Guinea pig maximization test: Podophyllum lignans.

The guinea pig maximization test was used to study the sensitizing potential of Podophyllum lignans; 0.5% of each drug in propylene glycol was used for induction and 0.01%, 0.05%, 0.1% and 0.5% preparations for topical challenge. The 0.1% and 0.5% preparations caused non-allergic toxic reactions associated with the cutaneous destructive influence of these drugs; allergic responses did not occur.

Fluorophotometry. II. Streptozocin-treated guinea pigs.

Normal and streptozocin-treated guinea pigs were examined with a fluorophotometer one hour after receiving an intravenous injection of fluorescein sodium. The level of fluorescence in the vitreous was significantly greater in animals showing marked glucosuria than in normal animals or those showing little glucosuria. The excessive fluorescence of the vitreous is similar to that reported by others in diabetic humans and rats but is found in the guinea pig, an animal known normally to have no retinal vasculature.

Comparison of guinea pig sensitization methods.

Up to now, different methods in guinea pig sensitization were used, to assess contact sensitization potential of low molecular weight chemicals. In order to compare the efficiency of the epicutaneous, intradermal and combined sensitization methods, two stronger sensitizers, the water soluble chlormethylimidazoline hydrochloride (CMI) and the lipid soluble 1,3-dinitrochlorobenzene (DNCB) were used for the experiments. Eleven modified sensitization methods were performed. In a first trial each substance was tested on 10 animals. In a further study those techniques which had produced the best res…

Cytomegalovirus infection in weanling guinea pigs.

Cytomegalovirus (CMV) infection of weanling guinea pigs was studied after subcutaneous and intracerebral inoculation of virus. No clinical illness appeared by day 24 after subcutaneous inoculation, even though animals developed persisting infection in many tissues, with marked variation in histogogic expression. Virus was obtained in higher titer from homogenates of salivary gland and thymus tissue. In other organs, including the brain, virus was found only be cocultivation of trypsinized liver tissue cells with guinea pig embryo cells. Appearance of virus in tissue culture was sometimes delay…

Liver disease in Papua New Guinea.

This paper gives, in detail, the causes of either liver disease or hepatomegaly in 100 patients, mostly adults, admitted to the medical wards of Angau Memorial Hospital, Lae, during 1968 and 1969. The major findings included liver cell carcinoma, cirrhosis (often with chronic active hepatitis), tropical splenomegaly, pericholangitis and hepatitis. There were 27 with miscellaneous findings including ten with normal, or almost normal, livers despite the definite enlargement. Patients with liver cell carcinoma presented late in the course of their illness and had a poor prognosis. Others, with pe

Transmembrane potentials in guinea-pig hepatocytes.

1. In pieces of guinea-pig liver with an intact capsule, the mean resting potential (E) is -49.8 mV, close to the value for in vivo hepatocytes and significantly higher than a similar preparation of rat liver. E is considerably less than the calculated K equilibrium potential of about -90 mV.2. Changes in [K(+)](o) in the range of 0-10 mM produce almost no change in E. At [K(+)](o) above 20 mM, a tenfold increase produces only a 33 mV depolarization. The results with changing [K(+)](o) are the same in Cl(-) free solutions (isethionate substitution) or when the [K(+)](o)[Cl(-)](o) product is ke…

Epidemiological studies on guinea-worm infection.

Dracontiasis is one of the most easily preventable of the tropical parasitic diseases, yet in India the infection is still present in large numbers of people and causes extreme degrees of morbidity and incapacitation. The 10 000 inhabitants of 4 villages in South India were interviewed and the prevalence of guinea-worm infection was found to vary from 11% to 54%; the prevalence increased with the proportion of the population using step-wells. Questioning revealed that infections with more than one worm at a time had occurred in 707 out of 1759 infected persons, that reinfections were common and…

Naturally occuring histoplasmosis in guinea pigs.

Histoplasmosis naturally occurring in laboratory guinea pigs is described in its clinical, necropsy, histological and mycological aspects. The animals if adult show a chronic disease with progressive emaciation and lameness of the hind legs. The young below three months of age died in 2 to 4 weeks presenting ruffled fur, great dorsal curvature and sometimes closed eyelids and catarrhal conjunctivitis. At necropsy the principal lesions were ulcerative gastritis, hemorrhagic and catarrhal enteritis, enlarged spleen and mesenteric lymph nodes. Sometimes the liver, lungs, mediastinal lymph nodes an…

Guinea worm disease--the final chapter?

The global campaign for the eradication of dracunculiasis has succeeded in reducing the incidence of guinea worm disease by >99% in the past 20 years. This has been accomplished by the provision of safe drinking water supplies, filtration of drinking water, chemical control of intermediate hosts, case containment and, crucially, local health-education initiatives.

Health research in Papua New Guinea.

The Papua New Guinea Institute of Medical Research (PNGIMR) is one of the most respected health research institutions in the developing world, and its studies of the local health problems of PNG have consistently had international relevance. This article examines the structural and philosophical factors that have enabled the success of the PNGIMR, and presents the PNGIMR approach to research as a potential model for other disease-endemic countries. An overview of PNGIMR research into malaria and filariasis is given with selected examples as an introduction to a Trends in Parasitology series on…
