Nipah virus (NiV) is a highly pathogenic zoonotic virus that causes severe encephalitis and respiratory diseases and has a high mortality rate in humans (>40%). Epidemiological studies on various fruit bat species, which are natural reservoirs of the virus, have shown that NiV is widely distributed throughout Southeast Asia. Therefore, there is an urgent need to develop effective NiV vaccines. In this study, we generated recombinant vaccinia viruses expressing the NiV glycoprotein (G) or fusion (F) protein using the LC16m8 strain, and examined their antigenicity and ability to induce immunity.

Nipah virus is a relatively newly discovered emerging virus on the WHO list of priority pathogens which has the potential to cause outbreaks with high fatality rates. Whilst progress is being made in the development of animal models for evaluating vaccines and therapies, some of the more fundamental data on Nipah virus are lacking. We performed studies to generate novel information on the aerosol survival of Nipah virus and to look at the efficacy of two common disinfectants. We also performed studies to evaluate the inactivation of Nipah virus by using neutral buffered formalin. Nipah virus w

Nipah virus (NiV) is a highly virulent zoonotic pathogen capable of triggering severe outbreaks with pandemic potential. This review consolidates current knowledge on its biology, transmission, pathogenesis, and clinical presentation, alongside recent progress in diagnostics, therapeutics, and vaccine development. Human encroachment into wildlife habitats has intensified interactions with fruit bat reservoirs, driving recurrent spillover events in South and Southeast Asia. Advances in structural virology have elucidated the mechanisms of viral entry, particularly the interaction of the G (atta

OBJECTIVES: Nipah and Hendra are deadly zoonotic diseases with pandemic potential. To date, no human vaccine or monoclonal antibody (mAb) has been licensed to prevent disease caused by these pathogens. The aim of this scoping review was to identify and describe all Phase I, II, and III clinical trials of vaccine candidates or mAbs candidates designed to prevent Nipah and Hendra in humans and to compare the characteristics of the vaccine candidates to characteristics outlined in the Target Product Profile drafted by the World Health Organisation as part of the WHO Research & Development Bluepri

Nipah virus (NiV) causes a highly lethal disease in humans who present with acute respiratory or neurological signs. No vaccines against NiV have been approved to date. Here, we report on the clinical impact of a novel NiV-derived nonspreading replicon particle lacking the fusion (F) protein gene (NiVΔF) as a vaccine in three small animal models of disease. A broad antibody response was detected that included immunoglobulin G (IgG) and IgA subtypes with demonstrable Fc-mediated effector function targeting multiple viral antigens. Single-dose intranasal vaccination up to 3 days before challenge

Nipah virus (NiV), a highly pathogenic Henipavirus in humans, has been responsible for annual outbreaks in recent years. Experiments involving live NiV are highly restricted to biosafety level 4 (BSL-4) laboratories, which impedes NiV research. In this study, we developed transcription and replication-competent NiV-like particles (trVLP-NiV) lacking N, P, and L genes. This trVLP-NiV exhibited the ability to infect and continuously passage in cells ectopically expressing N, P, and L proteins while maintaining stable genetic characteristics. Moreover, the trVLP-NiV displayed a favourable safety …

BACKGROUND: Ticks are the second most significant vector of human pathogens worldwide, with 911 documented species globally and a broad distributed across China. Currently, over 160 tick-borne viruses (TBVs) have been identified, several of which pose substantial threats to human health, such as Dabie bandavirus, Jingmen tick virus, Alongshan virus, Songling virus, Beiji nairovirus, and Langya henipavirus, raising increasing global attention. Despite their significance, the diversity of TBVs in Beijing remains poorly characterized.

Nipah virus (NiV) and Hendra virus (HeV) are newly emerging dangerous zoonotic pathogens of the Henipavirus genus of the Paramyxoviridae family. NiV and HeV (HNVs) which are transmitted by bats cause acute respiratory disease and fatal encephalitis in humans. To date, as there is a lack of antiviral drugs or effective antiviral therapies, the development of vaccines against those two viruses is of primary importance, and the immunogen design is crucial to the success of vaccines. In this study, the full-length protein (G), the ectodomain (Ge) and the head domain (Gs) of NiV attachment glycopro…

Prediction and management of zoonotic spillover requires an understanding of infection dynamics within reservoir host populations. Spillover risk is commonly inferred from infection prevalence based on detection of viral genomic material, yet detection alone does not indicate the presence of infectious virus or a sufficient dose for transmission. We undertook a comprehensive investigation of Hendra virus shedding in its primary reservoir, Pteropus bats, analyzing quantitative PCR with reverse transcription (RT-qPCR) data from 6151 pooled urine samples collected across five sites over 3 years.

Nipah virus is a priority pathogen that is receiving increasing attention among scientists and in work on epidemic preparedness. Despite this trend, there has been almost no bioethical work examining ethical considerations surrounding the epidemiology, prevention, and treatment of Nipah virus or research that has already begun into animal and human vaccines. In this paper, we advance the case for further work on Nipah virus disease in public health ethics due to the distinct issues it raises concerning communication about the modes of transmission, the burdens of public health surveillance, th

The emergence of the novel henipavirus, Langya virus, received global attention after the virus sickened over three dozen people in China. There is heightened concern that henipaviruses, as respiratory pathogens, could spark another pandemic, most notably the deadly Nipah virus (NiV). NiV causes near-annual outbreaks in Bangladesh and India and induces a highly fatal respiratory disease and encephalitis in humans. No licensed countermeasures against this pathogen exist. An ideal NiV vaccine would confer both fast-acting and long-lived protection. Recently, we reported the generation of a recom…

Nipah virus (NiV) is a highly pathogenic, recently emerged paramyxovirus that has been responsible for sporadic outbreaks of respiratory and encephalitic disease in Southeast Asia. High case fatality rates have also been associated with recent outbreaks in Malaysia and Bangladesh. Although over two billion people currently live in regions in which NiV is endemic or in which the Pteropus fruit bat reservoir is commonly found, there is no approved vaccine to protect against NiV disease. This report examines the feasibility and current efforts to develop a NiV vaccine including potential hurdles

Climate change and biodiversity loss are global environmental crises driving emerging infectious diseases, including bat-borne pathogens of public health significance: Ebola, Marburg, Nipah, Hendra, classic rabies virus, and Australian bat lyssavirus, as well as the fungus Histoplasma This review presents the evidence on the mechanisms of transmission, impact, and prevention and management strategies relevant to each of these pathogens. Transmission pathways vary between pathogens but often involve exposure to body fluids or excreta of a bat or other infected animal or human. The diseases asso

Nipah virus (NiV) is a highly virulent zoonotic virus classified as a priority pathogen and biohazard. In 2014, an outbreak of NiV-like disease in the Province of Sultan Kudarat, Mindanao, Philippines resulted in a 53% case fatality rate. Here, we identified wildlife bat hosts of NiV by conducting monthly serological surveillance of flying foxes and other native frugivorous bat species across Luzon. We estimated 13.92% NiV seroprevalence in native flying foxes. We also detected NiV neutralizing activity in some flying fox sera and identified factors such as age and seasonality as drivers of hi

BACKGROUND: Detected first in humans in 1998, Nipah virus (NiV) has produced regular outbreaks with high mortality rates in Southeast Asia. Outbreaks are often driven by human-to-human transmission, posing an infection risk to both the wider community and healthcare workers who care for NiV cases. Infection prevention and control (IPC) practices are key to preventing amplification of disease outbreaks and protecting both healthcare workers and other patients, but significant evidence gaps exist in the identification of specific modes of NiV transmission that occur in outbreaks and the effectiv

Nipah virus (NiV) is an emerging bat-borne zoonotic virus that can be transmitted to humans and other animals through infected bats or contaminated foods. The disease is highly lethal in humans (40%-75%) and has the potential for human-to-human transmission. Currently, there are no approved treatments or vaccines for NiV infection in humans or animals. Consequently, there is a pressing need for a highly sensitive, precise, and visually detectable assay to enable early intervention and mitigate the transmission of NiV infection. Here, we report a single-copy sensitive, field-deployable, one-pot

Nipah (Nee-pa) viral disease is a zoonotic infection caused by Nipah virus (NiV), a paramyxovirus belonging to the genus Henipavirus of the family Paramyxoviridae. It is a biosafety level-4 pathogen, which is transmitted by specific types of fruit bats, mainly Pteropus spp. which are natural reservoir host. The disease was reported for the first time from the Kampung Sungai Nipah village of Malaysia in 1998. Human-to-human transmission also occurs. Outbreaks have been reported also from other countries in South and Southeast Asia. Phylogenetic analysis affirmed the circulation of two major cla…
