Integration of Laccase-like Nanozymes and Sensing Strategies for Food Quality and Safety Monitoring.

Laccase-like nanozymes (LLNs) are a class of functional nanomaterials inspired by natural laccases. Due to their excellent catalytic performance, structural tunability, and superior stability, LLNs have emerged as promising tools for food quality and safety monitoring. This review establishes a critical structure-activity-application framework for LLNs, correlating bioinspired and structural engineering strategies, with the regulation of active sites, electronic structures, and catalytic microenvironments, and, ultimately, with their analytical performance in food matrices. Key insights are pr…

Laccase-Catalyzed Biotransformation of Dihydrostilbenes and Phenanthrenes: A Route to Novel Natural Product Derivatives.

Biotransformation is an eco-friendly and effective strategy for enhancing the chemical diversity of natural products for biological screening. Among various biotransformation methods, laccase-mediated transformations have successfully yielded bioactive compounds across multiple chemical classes. In this study, laccases from Trametes versicolor were used to catalyze the biotransformation of the dihydrostilbene gigantol and the phenanthrenes flavidin and coleonin, leading to the formation of a series of structurally complex derivatives. The selected substrates were subjected to laccase-mediated …

Functional characterization of fungal laccases via siRNA silencing enables targeted biodegradation.

Edible fungi serve as efficient bioconverters, transforming lignocellulosic wastes into valuable fungal proteins by extracellular enzyme systems (e.g., laccases). However, the functional characterization of these lignocellulolytic enzymes is still a major challenge, limiting the strategic application of fungal systems in targeted biodegradation. Here, we developed a versatile, non-transformative siRNA delivery platform for rapid gene function analysis in the edible fungus Pleurotus ostreatus. Using spray-induced (SIGS) in mycelia, injection-induced (IIGS) in fruiting bodies, and co-culture-ind…

Optimized production of laccase from Pseudomonas stutzeri and its biodegradation of lignin in biomass.

Laccases are multi-copper oxidases that play an important role in the biodegradation of phenolic compounds, lignin, dye, and wastes. Here, we report the screening of potential laccase-producing indigenous bacterial isolates and subsequent optimization of laccase production using crop residues as cheap supplementary energy sources. Among 16 bacterial isolates, seven were selected based on the appearance of reddish-brown bacterial colonies and guaiacol oxidation assay after 10 days of incubation at 37 °C. The maximum laccase activity (2.755 U/mL) was observed for bacterial isolate WR2. Response …

Biochemical Characterization of Laccase from Spirulina CPCC-695 and Their Role in Estrone Degradation.

The addition of exogenous endocrine disrupting compounds (EDCs) like estrone, in the food chain through the aquatic system, disrupts steroid biosynthesis and metabolism by altering either the genomic or non-genomic pathway that eventually results in various diseases. Thus, bioremediation of these compounds is urgently required to prevent their addition and persistence in the environment. Enzymatic degradation has proven to be a knight in shining armour as it is safe and generates no toxic products. The multicopper oxidases (E.C. 1.10.3.2 benzenediol: oxygen oxidoreductase), laccase with the po…

Tris-Copper Nanozyme as a Novel Laccase Mimic for the Detection and Degradation of Phenolic Compounds.

Phenolic compounds are one of the main organic pollutants in the environment that can seriously affect ecosystems, even at very low concentrations. Due to the resistance of phenolic compounds to microorganisms, conventional biological treatment methods face challenges in effectively addressing this pollution problem. In this study, a novel laccase mimic (Tris-Cu nanozyme) is prepared using a simple and rapid synthesis strategy based on the coordination of copper ions and amino groups in Tris(hydroxymethyl)aminomethane (Tris). It is found that the Tris-Cu nanozyme exhibits good catalytic activi…

Structural and biochemical insights into an engineered high-redox potential laccase overproduced in Aspergillus.

Fungal laccases have great potential as biocatalysts oxidizing a variety of aromatic compounds using oxygen as co-substrate. Here, the crystal structure of 7D5 laccase (PDB 6H5Y), developed in Saccharomyces cerevisiae and overproduced in Aspergillus oryzae, is compared with that of the wild type produced by basidiomycete PM1 (Coriolopsis sp.), PDB 5ANH. SAXS showed both enzymes form monomers in solution, 7D5 laccase with a more oblate geometric structure due to heavier and more heterogeneous glycosylation. The enzyme presents superior catalytic constants towards all tested substrates, with no …

[Determination of polyphenolic complex in wines by electrochemical methods and using the enzymes tyrosinase and laccase].

Several red wines were studied to find a correlation between physicochemical parameters characterizing the antioxidant status of wine and total content of phenols in samples. The content of dissolved oxygen (its value varied from 0.75 to 3.28 mg/ml), pH (3.10-3.63), redox potential (-186 to -106 mV), mass concentration of free and total sulfur dioxide (10-30 and 36-200 mg/dm3, respectively), absorption spectra, and total phenol content were determined. The wines fell into two main groups-with a relatively low (1850-2050 mg/dm3) and high (2300-2900 mg/dm3) contents of polyphenols. It was demons

Recent Progress and Future Prospects of Laccase Immobilization on MOF Supports for Industrial Applications.

Laccase is a multicopper oxidoreductase enzyme that can oxidize organics such as phenolic compounds. Laccases appear to be unstable at room temperature, and their conformation often changes in a strongly acidic or alkaline environment, making them less effective. Therefore, rationally linking enzymes with supports can effectively improve the stability and reusability of native enzymes and add important industrial value. However, in the process of immobilization, many factors may lead to a decrease in enzymatic activity. Therefore, the selection of a suitable support can ensure the activity and…

Recent advances in laccase activity assays: A crucial challenge for applications on complex substrates.

Despite being one of the first enzymes discovered in 1883, the determination of laccase activity remains a scientific challenge, and a barrier to the full use of laccase as a biocatalyst. Indeed, laccase, an oxidase of the blue multi-copper oxidases family, has a wide range of substrates including substituted phenols, aromatic amines and lignin-related compounds. Its one-electron mechanism requires only oxygen and releases water as a reaction product. These characteristics make laccase a biocatalyst of interest in many fields of applications including pulp and paper industry, biorefineries, fo…

[Monogenic variants in Laccase domain-containing 1 (LACC1) as the cause of juvenile arthritis].

Monogenic mutations in laccase domain-containing 1 (LACC1) are associated with clinical pictures that mimic severe courses of polyarticular or systemic juvenile idiopathic arthritis. The diseases are characterized by an early onset during the first year of life, a familial clustering and a high inflammatory activity. The courses are mostly difficult to influence and often lead to sequelae. In this article four cases from two families are presented in which the homozygous mutation p.T276fs* in LACC1 was detected. The children initially suffered from polyarticular or systemic forms of juvenile a…

Kinetics and mechanisms for sulfamethoxazole transformation in the phenolic acid-laccase (Trametes versicolor) system.

Oxidation of phenolic acids (PCs) by laccase could produce various kinds of reactive oxygen species (ROS), which is expected to have substantial impact on the transformation of antibiotics like sulfamethoxazole (SMX) in soil and aquatic environments. In this study, the formation of semiquinones radical (SQ●-), superoxide anion radical (O2●-), hydrogen peroxide (H2O2), hydroxyl radical (●OH), and singlet oxygen (1O2) in a laccase-gallic acid (GA) reaction system was confirmed. Meanwhile, GA would be transformed to its monomeric quinone and quinones of di- and tri-polymers. Transformation of SMX…

Inhibition mechanisms of Fe2+/Fe3+ and Mn2+ on fungal laccase-enabled bisphenol a polyreaction.

Bisphenol A (BPA) is regarded as an endocrine disruptor associated with negative health effects in animals and humans. Laccase from white-rot fungus can enable BPA oxidation and auto-polymerization to circumvent its biotoxicity, but the work concerning the effect mechanisms of divalent and trivalent metal ions (MIs) on BPA polyreaction have rarely been reported. Herein, Trametes versicolor laccase-started BPA conversion within 1 h followed pseudo-first order kinetics, and the rate constant (kprcs) and half-life were respectively 0.61 h-1 and 1.14 h. The presence of Ca2+, Mg2+, Cu2+, Pb2+, Cd2+…

Coupling of natural organic matter-metal binding and laccase-catalyzed oxidation of tetrabromobisphenol A.

Laccases are a group of copper-containing oxidase enzymes found in aquatic and terrestrial environment. They can catalyze one-electron oxidation of phenolic compounds to radical intermediates using molecular oxygen as the electron accepter. The radical intermediates can subsequently couple to each other to form dimers. In this study, we investigated the kinetics of tetrabromobisphenol A (TBBPA) transformation in laccase-catalyzed oxidation process. It was revealed that the removal of TBBPA was first order to the concentrations of both substrate and laccase. Natural organic matter (NOM) inhibit…

A new laccase-mediator system facing the biodegradation challenge: Insight into the NSAIDs removal.

Non-steroidal anti-inflammatory drugs (NSAIDs) are widely found pollutants in the aquatic environment and the currently available treatments for their removal are usually associated with some drawbacks. The aim of this research was to apply a laccase-mediator system for the degradation of some commonly used NSAIDs, namely diclofenac (DCF), naproxen (NAP) and ketoprofen (KP). The biocatalyst was obtained by direct immobilization on chitosan beads of a periodate-oxided laccase from Trametes versicolor. A preliminary study aimed to optimize DCF degradation in the presence of 2,2-azinobis (3-ethyl…

Impact of simultaneous retention of micropollutants and laccase on micropollutant degradation in enzymatic membrane bioreactor.

This study systematically compares the performance of ultrafiltration (UF) and nanofiltration (NF) based enzymatic membrane bioreactors (EMB … in NF-EMBR as compared to UF-EMBR, which could be attributed to the prolonged contact time between laccase and micropollutants following their effective retention by the NF membrane. A small decline in permeate flux was observed during EMBR operation. However, the flux could be recovered by flushing the membrane with permeate.
