Comparative genomics of three non-hematophagous leeches (Whitmania spp.) with emphasis on antithrombotic biomolecules.

Leeches are well known for blood-feeding habits and are widely used for medicinal purposes as they secrete various antithrombotic substances. However, some leeches exhibit non-hematophagous habits and their significance for medicinal use is controversial. Here we provide the chromosome-level genomes of two non-hematophagous leeches, Whitmania acranulata and Whitmania laevis, and, in combination with previous results from Whitmania pigra, we compared these genomes with an emphasis on antithrombotic biomolecules. All three species had the same chromosome number of 11. The genome size, repeat sit…

Management of herpes zoster in Ayurveda through medicinal leeches and other composite Ayurveda Treatment.

Herpes zoster commonly known as shingle's caused by varicella zoster virus (VZV). Recent studies have shown that exosomes play important role in pathogenesis as well as spread of herpes. Leeches when applied over affected part of herpes sucks the blood and there by interferes with extracellular communication through these exosomes and their by showing pain reduction, reduction in burning and arrest progression of disease. This technique is cost effective and easy to apply. In Ayurveda herpes can be correlated with Pittaj visarpa. Principle of treatment is Raktmokshan (~Bloodletting) and Pradeh…

Time to Change Theory; Medical Leech from a Molecular Medicine Perspective Leech Salivary Proteins Playing a Potential Role in Medicine.

Followed by developing modern medicine, leeches did not have extensive use as before; however, in the late 19th century, they were still used in most countries all over the world. Thus far, leeches were utilized in treating various diseases like skin disorders, arthritis, and cancer. In Egypt, using leeches for treatment dates back to early 1500 BC. A medical leech's salivary glands involve over 100 bioactive proteins and the salivary gland secretion contains bacteriostatic, analgesic, and anticoagulation influences; with resolving activity, it causes microcirculation disorders elimination, re…

Beyond the epistaxis: voluntary nasal leech (Dinobdella ferox) infestation revealed the leech behaviours and the host symptoms through the parasitic period.

Dinobdella ferox is the most reported and studied leech species parasitizing mammalian mucus membranes. However, our knowledge of D. ferox during parasitizing and its effects on hosts has remained limited. To investigate the behaviour of D. ferox in the parasitic period and the subsequent host symptoms, the author was infested voluntarily with three juvenile leeches under regularly-conducted examinations of complete blood count (CBC) and nasal endoscopy (NE). After the first head protrusion of the parasitizing leech at the host nostril, the occurrence of bleeding-related symptoms decreased and…

Characterization of the natural peptidome of four leeches by integrated proteogenomics and pseudotargeted peptidomics.

Animal-derived drugs are an indispensable part of folk medicine worldwide. However, their chemical constituents are poorly approached, which leads to the low level of the quality standard system of animal-derived drugs and further causes a chaotic market. Natural peptides are ubiquitous throughout the organism, especially in animal-derived drugs. Thus, in this study, we used multi-source leeches, including Hirudo nipponica (HN), Whitmania pigra (WP), Whitmania acranulata (WA), and Poecilobdella manillensis (PM), as a model. A strategy integrating proteogenomics and novel pseudotargeted peptido…

Distribution and morphology of nociceptive cells in the CNS of three species of leeches.

The present study describes the segmental variation in the distribution and morphology of nociceptive neurons (N cells) in the central nervous system of the leech. N cells of midbody ganglia can be segregated into lateral and medial types. We show that monoclonal antibodies specific for N cells can distinguish between the two populations. The monoclonal antibodies were used to map the complete distribution of the cells along the nervous cord. There are two pairs of the medial and lateral nociceptive neurons in the midbody ganglia, one pair of the medial type in the sex ganglia (5 and 6), and a

Mechanisms and Research Progress of Leech in Diabetic Ulcer Fibrosis.

Delayed wound healing is a major complication of diabetes, with its core pathological process being pathological fibrosis driven by persistent inflammation, metabolic disorders and vascular dysfunction under hyperglycemia. This process involves pathological retention of inflammatory cells, abnormal expression of pro-fibrotic factors, and excessive deposition of extracellular matrix. As a traditional Chinese medicine, leech possesses multiple pharmacological effects including anticoagulation, anti-inflammation and improvement of microcirculation, exerting its therapeutic roles through regulatin

Hirudins and fenestrins of the African medicinal leech Asiaticobdella fenestrata.

African hematophagous leeches are largely underinvestigated both in terms of their biodiversity and the characterization of the repertoire of bioactive components of their salivary gland secretions. Asiaticobdella (formerly Aliolimnatis) fenestrata Moore, 1939, also referred to as the African medicinal leech, is hence a promising candidate for the identification and functional characterization of putative anticoagulants that may serve as inspirations for the development of new or the enhancement of existing drugs for medical purposes. A. fenestrata inhabits flooded plains of southern African r…

Ocular infestation by a juvenile leech, Myxobdella sinanensis in Japan.

PURPOSE: The case of ocular infestation by a leech is rare. We reported that Myxobdella sinanensis infests conjunctiva. OBSERVATIONS: A 5-year-old girl presented with blood clots in the inner corner of the left eye, and a history bloody eye discharge and bloody tears for 5 days. She was prescribed 0.5% levofloxacin ophthalmic drops for conjunctival damage. However, her parent watched a worm moving in her conjunctiva while taking a bath. She presented again the same day, and a worm was found in the left eye of the lower conjunctival fornix and was adsorbed to the inner corner. We removed a wor

An Unusual Cause of Recurrent Epistaxis: Nasal Leech Infestation.

Though a common symptom, epistaxis should never be overlooked as the implications may be consequential. Nasal leech infestation represents an unusual etiology of recurrent epistaxis. The leeches release enzymes to anaesthetize the attachment site to facilitate the feeding process. Given the inconspicuous area of infestation and the absence of pain, there is often a delayed treatment. A detailed history taking and physical examination remain paramount in the assessment of recurrent epistaxis.

Characterization of a monoacylglycerol lipase in the medicinal leech, Hirudo verbana.

Endocannabinoids are a class of lipid neuromodulators found throughout the animal kingdom. Among the endocannabinoids, 2-arachydonoyl glycerol (2-AG) is the most prevalent endocannabinoid and monoacylglycerol lipase (MAGL) is a serine hydrolase primarily responsible for metabolizing 2-AG in mammals. In the medicinal leech, Hirudo verbana, 2-AG has been found to be an important and multi-functional modulator of synaptic transmission and behavior. However, very little is known about the molecular components of its synthesis and degradation. In this study we have identified cDNA in Hirudo that en

An Unusual Cause of Bleeding on the Floor of Mouth: Leech Infestation.

Leech infestation is a very rare phenomenon in humans. It mostly occurs in humans when rural untreated water is drunk or while swimming in streams or lakes. When leeches adhere to the mucous membrane, they ingest blood. Thus, they can sometimes cause severe anemia that may require blood transfusion. We report a case that was referred to emergency service with bleeding in the floor of the mouth. A 10-year-old child was referred to the emergency service of a city hospital with a complaint of swelling in the floor of the mouth and spitting of blood. The patient was promptly taken to the operating…

An unusual cause of dysphonia with hemoptysis: a laryngeal live leech.

INTRODUCTION: Foreign bodies in the upper airway are one of the most challenging otolaryngology emergencies and have various presentations depending on their physical properties and location. Leeches are blood-sucking hermaphroditic worms that vary in color, length, and shape. They usually reside in fresh-water streams and lakes. When rural untreated water is drunk, leeches may localize in the nose, pharynx, and esophagus, or rarely in the larynx. CASE REPORT: This case is a man who was referred to our otolaryngology clinic with a complaint of hemoptysis and mild respiratory distress. The pat…

Rectal bleeding due to leech bite: a case report.

We present herein a case with a four-day history of fresh rectal bleeding due to leech bite. The cause was found to be a leech in the rectum by anoscope. This pathological condition is extremely rare in urban areas. Leech endoparasitism, although rare, may cause serious, even lethal, complications. Suspicion of leech infestation should be kept in mind when faced with intermittent or severe rectal bleeding in humans, and should be investigated surgically as with all other foreign bodies. In the current study, we present a patient who admitted with rectal bleeding due to leech bite. The leech wa

Aquatic leech as a rare cause of respiratory distress and hemoptysis.

This paper presents a 73 years old male with three weeks history of intermittent hemoptysis, dysphagia, dyspnea, stridor, and suffocations sensation. By means of fibrotic bronchoscopic examination, the cause was found to be a leech in the glottis. It was removed by injection of 4 ml lidocaine 2%. Infestation into the respiratory tract by a leech may become lethal because of hypoxia and death secondary to airway obstruction. A high index of suspicion of leech infestation sould be considered in patients presenting with hemoptysis, hoarseness and respiratory distress and a history of recent conta

Unusual cause of unilateral epistaxis: nasal leech infestation.

Common causes for unilateral nasal bleeding in adults are benign or malignant tumors and deviated nasal septum. Here we present two cases of recurrent unilateral nasal bleeding due to leech infestation. Though it is not a common cause for nasal bleeding, a clinician should suspect leech infestation for a recurrent nasal bleeding specially in tropical countries like Nepal.
