Limbal epithelial stem cells (LESCs) are responsible for the maintenance and repair of the corneal surface. Injuries and diseases of the eye may result in a vision condition called limbal stem cell deficiency (LSCD). Without limbal stem cells, the cornea becomes opaque, vascularized, and inflamed. Cultured LESC therapy as a treatment method was first described in 1997, and LESCs cultured from either patients or donors have been used to treat LSCD successfully. However, the main source of cornea for LSCD treatment is from donors, which are too few to meet the demand (less than 1:70 of cases). T…

PURPOSE: Disease that develops on the ocular surface in animals and humans that have their limbal stem cells erased or their niche severely damaged by physical or chemical trauma, is difficult to visually evaluate in a progressive and long-term manner. When limbal stem cell deficiency (LSCD) develops under such circumstances, intravital clinical microscopy or other techniques cannot be used to accurately detail what transpires on the cornea. Here, we investigated the evolution of LSCD and characterize cellular transformations that arise when the limbal niche is lost.
METHODS: We employed the …

The mammalian cornea is decorated with stem cells bestowed with the life-long task of renewing the epithelium, provided they remain healthy, functional, and in sufficient numbers. If not, a debilitating disease known as limbal stem cell deficiency (LSCD) can develop causing blindness. Decades after the first stem cell (SC) therapy is devised to treat this condition, patients continue to suffer unacceptable failures. During this time, improvements to therapeutics have included identifying better markers to isolate robust SC populations and nurturing them on crudely modified biological or biomat…

This approach aimed to assess the therapeutic significance of BM-MSCs in alleviating LSCD elicited in rats via corneal alkali burns and to explore the associated repairing mechanisms. Rats were assigned to (1) control group, (2) alkali burn group, and (3) stem cells group. Corneal surface investigations were performed using a slit lamp and fluorescence tests. The therapeutic impact of BM-MSCs was appraised by estimating the protein levels of TNF-α, SOD, VEGF, MMP-2, and IL-8 using ELISA. Moreover, mRNA transcription levels of corneal TNF-α, NF-kB, COX-2, and caspase-3 were quantified. Histolog

Mesenchymal stem cells (MSCs) have unique and beneficial properties and are currently used to treat a broad variety of diseases. These properties include the potential for differentiation into other cell types, secretion of different trophic factors that promote a regenerative microenvironment, anti-inflammatory actions, selective migration to damaged tissues, and non-immunogenicity. MSCs are effective for the treatment of ocular surface diseases such as dry eye, corneal burns, and limbal stem cell deficiency (LSCD), both in experimental models and in humans. LSCD is a pathological condition i…

Limbal Stem Cell Deficiency (LSCD) is a corneal disorder characterized by conjunctivalization of the cornea. Central corneal epithelial thickness, measured using Anterior Segment Optical Coherence Tomography (AS-OCT), is a biomarker for LSCD. Manual measurement of epithelial thickness remains the gold standard for diagnosis because existing automated commercial software is unreliable in severe cases. Standard machine learning approaches have limited applicability for automating this measurement due to the difficulty of distinguishing corneal epithelium from scar tissue, and imprecise clinical …

Ocular surface inflammation is a major disruptor of corneal epithelial homeostasis and a key driver of limbal stem cell deficiency (LSCD). Limbal stem cells (LSCs), residing within the specialized limbal niche, maintain corneal transparency through continuous epithelial renewal and by preventing conjunctival encroachment onto the corneal surface. Chronic or severe inflammatory insults-stemming from systemic autoimmune disorders, ocular surface diseases, infections, trauma, or environmental stressors-can damage both LSCs and their microenvironment, ultimately leading to limbal insufficiency. Th…

Limbal stem cell deficiency (LSCD) is a sight-threatening condition caused by the loss and/or dysfunction of limbal stem cells (LSCs), which are essential for corneal epithelial regeneration and homeostasis and are critical for maintaining corneal transparency. We have previously shown that specific inactivation of the endothelial mineralocorticoid receptor (MR) inhibits corneal neovascularization (CN) and that MR antagonists (MRA) improve corneal epithelial wound healing. This study investigated the therapeutic potential of MRA in LSCD and their mechanisms of action. Using a rat model of LSCD…
