In previous studies we have reported that patients with mild atopic eczema have enhanced lymphocyte mitogenesis while those with severe disease have markedly suppressed responses. A macrophage activating factor (MAF) has been previously described in mixed lymphocyte cultures (MLC) between the donor and the recipient of a mouse skin allograft. This factor can render macrophages cytotoxic against mouse mastocytoma targert cells. One of the major side effects induced by the in vivo administration of the murine monoclonal antibody OKT3 is a spontaneously reversible clinical syndrome associating in variable proportions depending on the patient: fever, chills, headaches, diarrhea and seldomly meningismus. Sera from 3 renal allograft recipients treated with OKT3 were studied and showed that a massive although transient release of some cytokines namely, Tumor Necrosis Factor alpha, Interleukin 2 and Interferon gamma is observed following the first OKT3 injection.

We studied polyclonal B-cell activation in twenty-six patients with discoid lupus erythematosus (DLE). Spontaneous plaque-forming cells of the IgA class (IgA-SPFC) as determined by a reverse haemolytic plaque assay were significantly more common in patients with DLE than in fifty control subjects. The patients showed a positive correlation between IgA-SPFC and OKT4/8 ratios and also had a significantly higher mean OKT4/8 ratio. Peripheral blood lymphocytes from some patients with rheumatoid vasculitis and giant cell arteritis were cytotoxic in vitro towards endothelial cells, but not fibroblasts. Use of cell surface markers and cell density showed that cytotoxicity correlated with numbers of HLA-DR bearing cells, but not with cells bearing CD3, the transferrin receptor, or less dense lymphocytes. We concluded that activated, cytotoxic lymphocytes were present in the peripheral blood of a sub-set of patients with vasculitis.

UNLABELLED: The activity of ectonucleoside triphosphate diphosphohydrolase (E-NTPDase; EC 3.6.1.5) was characterized in hepatic lymphocytes (HL) of rats. For this purpose, a specific method for the isolation of lymphocytes from hepatic tissue was developed. CGRP is a 37-amino acid neuropeptide found in the central and peripheral nervous systems as well as the nerve endings in lymphoid organs. Specific CGRP receptors are present on both T and B lymphocytes. There is increasing evidence that CGRP plays a role in regulation of the immune response. However, few investigations have examined the effects of CGRP on lymphocyte effector functions. In this report, CGRP (0.1 nM-1 microM) was shown to cause concentration-dependent inhibition of IL-2-activated lymphocyte growth inhibition of the fungus Candida albicans and cytotoxic activity for tumor cells.

OBJECTIVES: To investigate the pattern of glutamate dehydrogenase (GLDH) activity, GLUD1 and GLUD2 expressions in peripheral blood mononuclear cells (PBMC) of untreated B-chronic lymphocytic leukemia (B-CLL) in healthy individuals (HI) and patients with infectious mononucleosis (IM).
DESIGN AND METHODS: GLDH activity was determined in a supernatant obtained from pelleted PBMC. GLUD1 and GLUD2 mRNA expression was determined using a quantitative real-time polymerase chain reaction. CD19(+) B cells from PBMC were purified by using positive selection.

Treatment with anti-theta serum and the Wigzell column technique for cell separation was employed to study the separate functions of the B and T lymphocytes in the late states of immunity to the LCM virus in mice.
