Memory consolidation after learning involves spontaneous, brain-wide network reorganization during rest and sleep, but how this is achieved is still poorly understood. Current theory suggests that the hippocampus is pivotal for this reshaping of connectivity. Using fMRI in male mice, we identify that a different set of spontaneous networks and their hubs are instrumental in consolidating memory during post-learning rest. We found that two types of spatial memory training invoke distinct functional connections, but that a network of the sensory cortex and subcortical areas is common for both ta

OBJECTIVE: JM-20, a novel hybrid synthetic molecule, has been reported to have antioxidant, mitoprotective, anti-excitotoxic, anti-apoptotic and anti-inflammatory properties. However, the neuroprotective effect of JM-20 against memory impairment in preclinical AD-like models has not been analyzed. The aim of this study was to evaluate the potential neuroprotection of JM-20 that preserves essential memory process from cholinergic dysfunction and other molecular damages.
METHODS: The effects of JM-20 on scopolamine (1 mg/kg)-induced cognitive disorders were studied. Male Wistar rats (220-230 g)

Stressful events can generate emotional memories linked to the traumatic incident, but they also can impair the formation of nonemotional memories. Although the impact of stress on emotional memories is well studied, much less is known about the influence of the emotional state on the formation of nonemotional memories. We used the novel object-recognition task as a model of nonemotional memory in mice to investigate the underlying mechanism of the deleterious effect of stress on memory consolidation. BACKGROUND: Attention-deficit/hyperactivity disorder (ADHD) is a neurodevelopmental disorder commonly linked to deficits in executive function. However, some theoretical frameworks propose that ADHD may also involve disruptions in cognitive mechanisms central to statistical learning, such as predictive processing and implicit learning. Moreover, abnormal sleep electroencephalography in ADHD raises the possibility of altered sleep-dependent memory consolidation of statistical learning.
AIM: Here we examined how individuals with ADHD acquire and consolidate statistical regularities in auditory

Episodic memory is a critical cognitive function that enables the encoding, storage, and retrieval of new information. Memory consolidation, a key stage of episodic memory, stabilizes this newly encoded information into long-lasting brain "storage." Studies using fMRI to investigate post-encoding awake rest holds promise to shed light on early, immediate consolidation mechanisms. Here, we review fMRI studies during episodic memory to document common methods to investigate post-encoding consolidation, such as multivoxel pattern analysis (MVPA) and functional connectivity, and the current state

Memory consolidation is the process by which newly acquired experiences are stabilized into long-term memory, involving coordinated cellular and network-level activity across brain regions such as the hippocampus and prefrontal cortex. Dysregulation of this process has been implicated in psychiatric disorders including post-traumatic stress disorder (PTSD), which is characterized by the over-consolidation of traumatic memories. LIM kinase 1 (LIMK1), a key regulator of synaptic plasticity, is believed to play an important role in memory consolidation across hippocampal-cortical circuits. In thi

Two important themes in Ivan Izquierdo's research each offered both answers and questions about the topic of memory formation and maintenance. The first theme provided evidence supporting the view that short- and long-term memory were distinct processes and could be selectively modulated by several treatments, with some affecting only short-term, others only affecting long-term memory, and still others affecting both. Over many years, Izquierdo's laboratory documented molecular responses across time after training obtaining results that showed differences as well as similarities in the biochem

Sleep spindles benefit declarative memory consolidation and are considered to be a biological marker for general cognitive abilities. However, the impact of sexual hormones and hormonal oral contraceptives (OCs) on these relationships are less clear. Thus, we here investigated the influence of endogenous progesterone levels of naturally cycling women and women using OCs on nocturnal sleep and overnight memory consolidation. Nineteen healthy women using OCs (MAge = 21.4, SD = 2.1 years) were compared to 43 healthy women with a natural menstrual cycle (follicular phase: n = 16, MAge = 21.4, SD

Ethanol is one of the most consumed substance worldwide that impairs learning and memory processes, resulting in amnesia or blackout. Due to the genetic conservation, rich behavioral repertoire, and high pharmacological tractability, the zebrafish (Danio rerio) has emerged as a powerful model organism for assessing preventive strategies against the noxious effects of ethanol in vertebrates. Here, we used an inhibitory avoidance apparatus to investigate the potential preventive effects of taurine in a novel ethanol-induced amnesia model in zebrafish. The experimental tank consisted of two compa

Networks of neuronal synapses are the fundamental basis for making and retaining memory. Reduced synapse number and quality correlates with loss of memory in dementia. Heat shock factor 1 (HSF1), the major transcription factor regulating expression of heat shock genes, plays a central role in proteostasis, in establishing and sustaining synaptic fidelity and function, and in memory consolidation. Support for this thesis is based on these observations: (1) heat shock induces improvements in synapse integrity and memory consolidation; (2) synaptic depolarization activates HSF1; (3) activation of

Some patients with temporal lobe epilepsy display an atypical memory disorder known as very long-term amnesia or accelerated forgetting. The characteristics of the study patients were as follows: First, they could retain information long after the test was adminstrated; however, several weeks later, they could not remember the information (long-term anterograde amnesia: LAA). Second, they showed dense retrograde amnesia for decades (long-term retrograde amnesia: LRA). A plausible explanation of the symptoms was a consolidation deficit resulting from epileptiform activity. Indeed, 1 study has r

Based on a synthesis of recent advances, this review elucidates the distinct yet synergistic roles of non-rapid eye movement (NREM) and rapid eye movement (REM) sleep in memory consolidation. During NREM sleep, cortical slow oscillations (SOs) establish a temporal framework by alternating between UP and DOWN states, facilitating cross-regional communication. SOs phase-couple with thalamic spindles to coordinate hippocampal sharp-wave ripples (SWRs), thereby driving the reactivation and transfer of labile hippocampal memories to neocortical networks for long-term storage. Concurrent synaptic ho

Post-traumatic stress disorder (PTSD) is a mental health disorder triggered by exposure to a traumatic event, which manifests with anguish, intrusive memories, and negative mood changes. So far, there is no efficient treatment for PTSD other than symptomatic palliative care. However, the cytoplasmic polyadenylation element binding protein-3 (CPEB3) has been recently associated with a potential risk gene for PTSD. Considering that the CPEB3 protein is a functional amyloid whose importance for long-term memory consolidation in mammals is well established, we propose the active (amyloid) state of

Sleep benefits memory consolidation through periodic sleep spindle activity and associated memory reactivations. The temporal organization of spindles in "trains" is considered a critical sleep mechanism for the timed and repeated reactivation of memories. Evidence suggests that a timely phase-locking between slow oscillations (SO) and spindles facilitates learning-related synaptic plasticity. Here, we investigated the contribution of spindles' clustering and coupling with SO in motor memory consolidation by promoting local synaptic depression in sensorimotor cortical regions through upper-lim

There is increasing evidence that brief periods of quiet wakeful rest immediately after learning facilitate memory consolidation. Similar to consolidation during sleep, this effect has been assumed to derive from neocortical-hippocampal interactions that are accompanied by slow-oscillating cortical activity. This assumption is supported by studies showing that the magnitude of frontal slow-oscillating activity during quiet wakefulness predicts memory performance and that slow-oscillating transcranial direct current stimulation (so-tDCS) during sleep can enhance memory retrieval. We tested whet

Object recognition memory (ORM) allows individuals to discriminate between familiar and novel stimuli, providing a fundamental model for studying non-emotional memory in rodents. ORM reactivation during retrieval in the presence of a new object renders this memory labile and susceptible to interference during reconsolidation, a process with potential translational impact, as it contributes to the maintenance and updating of stored memories. Despite this relevance, the mechanisms underlying ORM reconsolidation are not well understood. Phosphodiesterase-4 (PDE4) regulates cyclic adenosine monoph

Stochastic resonance sensory input modulates the central nervous system's excitability, thereby possibly influencing motor skill learning and retention. We investigated the effects of transcutaneous spinal random noise stimulation (tsRNS) at the cervical level on motor skill learning and corticospinal transmission in healthy humans. Participants performed a 20 min visuomotor tracking training task requiring rapid shifts in pinch force, with motor performance tests conducted before, immediately after, 1 day after and 7 days after the training to assess motor skill learning and retention. Sleep disordered breathing (SDB) has its peak incidence in early childhood, but research into the impact of disrupted sleep on learning and overnight memory consolidation has been largely neglected in this population. Hence, we aimed to determine if children with SDB show deficits in learning and overnight consolidation of declarative and procedural memories compared to healthy control (HC) children. Forty-nine children aged 7-16 years (24 SDB, 25 HC) completed a cognitive battery, including two declarative (one verbal, one visual) and one procedural memory task. We manipulated the level of le
