Mercury Uptake by Desulfovibrio desulfuricans ND132: Passive or Active?

Recent studies have identified HgcAB proteins as being responsible for mercury [Hg(II)] methylation by certain anaerobic microorganisms. However, it remains controversial whether microbes take up Hg(II) passively or actively. Here, we examine the dynamics of concurrent Hg(II) adsorption, uptake, and methylation by both viable and inactivated cells (heat-killed or starved) or spheroplasts of the sulfate-reducing bacterium Desulfovibrio desulfuricans ND132 in laboratory incubations. We show that, without addition of thiols, >60% of the added Hg(II) (25 nM) was taken up passively in 48 h by live

[Organic mercury tolerance, absorption and transformation in Spartina plants].

Critical concentration of methylmercuric chloride (MeHgCl) in the nutrient solution to which Spartina plants are tolerant is 15 micromol/L, three times higher than that of tobacco plants. After being treated with methylmercuric chloride, total quantity of organic mercury within plants increased and that of nutrient solution decreased greatly so that total quantity of inorganic mercury rose. It is inferred that Spartina plants absorb organic mercury and partially transform organic into inorganic mercury, and then more inorganic mercury is accumulated in underground parts of plants. In addition,

Indirect detection of mercury-199 nuclear magnetic resonance spectra of methylmercury complexes.

Measurement of 199Hg NMR spectra of methylmercury species by the heteronuclear multiple quantum coherence (HMQC) indirect proton detection method and its application to the study of the solution chemistry of CH3HgII-thiol ligand complexes are described. A sensitivity enhancement factor of 16 is obtained for measurement of the 199Hg NMR spectrum of a 4 mM solution of the CH3HgII-glutathione complex by the indirect detection method, as compared to a theoretical enhancement of 74. The less-than-theoretical enhancement is attributed to loss of signal by relaxation during the HMQC pulse sequence. L

Voltage-activated calcium channel currents of rat DRG neurons are reduced by mercuric chloride (HgCl2) and methylmercury (CH3HgCl).

The actions of bath applied mercuric chloride (HgCl2) and methylmercury (CH3HgCl) on voltage-activated calcium channel currents (VACCCs) were tested, using the whole cell patch clamp recording technique with cultured dorsal root ganglion (DRG) neurons from 2-4 day old rat pups. Both metal compounds reduced the current irreversibly in a concentration dependent fashion, reaching a new (lower) steady state within 3 to 5 min after application. Inorganic mercury was more effective in reducing the VACCCs with an IC50 of 1.3 microM, while the IC50 for methylmercury was 2.6 microM. But the threshold c

Mercury modeling to predict contamination and bioaccumulation in aquatic ecosystems.

In response to increasing scientific evidence of the environmental toxicity of mercury and its organic compounds, this study reviews the state of knowledge about the mercury cycle in aquatic systems. It describes the aquatic chemistry of mercury and discusses the importance of biological and physicochemical parameters such as pH, temperature, dissolved organic carbon, oxygen concentration, mercury and methylmercury concentration and availability, as well as sulfate, manganese, and iron concentration in surfaced waters. There is still a paucity of information on environmental dependency of fact

Validation of methylmercury determinations in aquatic systems by alkyl derivatization methods for GC analysis using ICP-IDMS.

Isotope dilution mass spectrometry (IDMS), using an inductively coupled plasma quadrupole mass spectrometer (ICPMS) and a species-specific methylmercury spike was applied to validate the commonly used GC method for methylmercury (MeHg+) determination, which is based on the formation of volatile methylethylmercury by derivatizationwith NaBEt4. The spike compound, Me201Hg+, was synthesized by reaction of 201Hg-enriched mercury chloride with methylcobalamin. By analyzing different environmental aquatic samples, it was found that in most cases, transformation of MeHg+ into elemental mercury (Hg0)

Uptake and efflux of mercury sulfide nanoparticles in Escherichia coli.

Mercury sulfide nanoparticles (HgS NPs) are ubiquitous in nature and are methylated to neurotoxic methylmercury (MeHg) by microorganisms. However, there is a paucity of knowledge regarding the biokinetics of HgS NPs in microorganisms. Here, the biokinetic uptake and efflux of HgS NPs in the model bacterium (Escherichia coli) were quantified and compared with those of Hg-dissolved organic matter complexes (Hg-DOM). The results demonstrate that the uptake of HgS NPs was time- and concentration-dependent. The uptake rate constant of HgS NPs was 0.031 ± 0.006 L g-1 h-1, which was 2.4-fold higher t

Degradation of methylmercury into Hg(0) by the oxidation of iron(II) minerals.

Mercury contamination is a global concern, and the degradation and detoxification of methylmercury have gained significant attention due to its neurotoxicity and biomagnification within the food chain. However, the currently known pathways of abiotic demethylation are limited to light-induced photodegradation process and little is known about light-independent abiotic demethylation of methylmercury. In this study, we reported a novel abiotic pathway for the degradation of methylmercury through the oxidation of both mineral structural iron(II) and surface-adsorbed iron(II) in the absence of lig

Toxicity of organic and inorganic mercury to Saccharomyces cerevisiae.

In this study the effect if six different forms of mercury on the growth of the yeast Saccharomyces cerevisiae is presented. Five kinds of strains of S. cerevisiae were used. They were a wild type, a mercury-resistant type, and three mutants: mutation repair-deficient mutant, excision repair-deficient mutant, and recombination repair-deficient mutant. In terms of EC50 toward the wild-type strain, the toxicity order for the inorganic forms was Hg(NO3)2>HgSO4>HgCl2. Monovalent nitrate mercury Hg(NO3)2 was more toxic than bivalent Hg(NO3)2. The toxicity of organic mercury CH3HgCl on cell growth w

Form of mercury in stream fish exposed to high concentrations of dissolved inorganic mercury.

The form of mercury predominating in mercury-contaminated fish from both pristine and industrialized waters in North America and Europe has almost universally been methylmercury. Sunfish (Lepomis auritus) living in a stream contaminated with 0.5-1 micrograms/L dissolved inorganic mercury accumulated greater concentrations of total mercury at headwater sites, where the dissolved mercury concentrations were greatest, than they did at downstream sites. However, despite evidence from laboratory studies that dissolved inorganic mercury is rapidly accumulated by fish without transformation to methyl

Mechanisms of mercury (Hg) and methylmercury (CH3Hg+) removal by pristine and thiol-modified açaí waste nanobiochar.

Mercury (Hg) contamination remains a global environmental challenge, driven by sources such as artisanal gold mining, industrial emissions, and coal combustion. Developing efficient and sustainable remediation strategies is therefore critical. Here we evaluate for the first time the mechanisms controlling Hg and methylmercury (CH3Hg+) removal using nanobiochar derived from açaí (Euterpe oleracea Mart) seed residues and its thiol-functionalized counterpart. Nanobiochar (< 50 nm) was produced via pyrolysis at 600 °C and chemically modified to enhance sorption performance. Thiol functionalization

Degradation mechanism of methyl mercury selenoamino acid complexes: a computational study.

Density functional theory (DFT) calculations have been carried out on the possible degradation/demethylation mechanism of methyl mercury (CH(3)Hg(+)) complexes with free cysteine and seleonocysteine. The binding of CH(3)Hg(+) ions with one (seleno)amino acid is thermodynamically favorable. However, the binding with another acid molecule is a highly unfavorable process. The CH(3)Hg-(seleno)cysteinate then degrades to bis(methylmercuric)sulphide (selenide for the Se-containing complex) which in turn forms dimethyl mercury and HgS/HgSe, the latter being precipitated out as nanoparticles. The dime

[Bibliographical study of Minamata disease].

In 1958, Minamata Disease was suggested to be organic mercury compounds intoxication. This suggestion was based on Hunter and Russel's report on occupational exposure to methylmercury. This report is known to have established the typical symptoms of methyl mercury intoxication. However, it has been widely believed since the official recognition of Minamata Disease (1956) that, at the moment of outbreak, no reports were available on organic mercury formation from inorganic mercury in acetaldehyde production from acetylene, or on organic mercury intoxication among workers in acetaldehyde product

Water Activation on Sulfur Vacancies Enables Pyrite-Mediated Methylmercury Demethylation and Immobilization.

The accumulation of neurotoxic methylmercury (MeHg) in rice paddies presents a major global food safety challenge, yet effective in situ remediation strategies for flooded, anoxic soils are lacking. Here, we report that sulfur vacancy (SV)-rich pyrite efficiently degrades MeHg under dark, anoxic conditions via an innovative pathway involving vacancy-activated water (H2O). Experimental and theoretical evidence reveals that SVs serve a dual function: they enhance MeHg adsorption, weakening the carbon-mercury bond, while simultaneously activating ambient H2O to generate surface-bound hydroxyl rad

Predictive modeling of methylmercury in rice (Oryza sativa L.) and species-sensitivity-distribution-based derivation of the threshold of soil mercury in karst mountain areas.

The bioavailable mercury (Hg) in the soil is highly active and can affect the formulation of methyl-Hg (MeHg) in soil and its accumulation in rice. Herein, we predicted the concentration of MeHg in rice using bioavailable Hg extracted from soils; additionally, we determined the threshold value of soil Hg in karst mountain areas based on species sensitivity distribution. The bioavailable Hg was extracted using calcium chloride, hydrochloric acid (HCl), diethylenetriaminepentaacetic acid mixture, ammonium acetate, and thioglycolic acid. Results showed that HCl is the best extractant, and the pre
